    We have previously reported that ADP ribosylation factor like 2 (Arl2), a small GTPase, content influences microtubule dynamics and cell cycle distribution in breast tumor cells, as well as the degree and distribution of phosphorylated P53. Here we show, in two different human breast adenocarcinoma models, that Arl2 content has a major impact on breast tumor cell Aggressivity both in vitro and in vivo. Cells with reduced content of Arl2 displayed reduced contact inhibition, increased clonogenic or cluster formation as well as a proliferative advantage over control cells in an in vitro competition assay. These cells also caused larger tumors in SCID mice, a phenotype which was mimicked by the in vivo administration of siRNA directed against Arl2. Cells with increased Arl2 content displayed reduced Aggressivity, both in vitro and in vivo, with enhanced necrosis and were also found to contain increased PP2A phosphatase activity. A rt-PCR analysis of fresh human tumor breast samples suggested that low Arl2 expression was associated with larger tumor size and greater risk of lymph node involvement at diagnosis. These data underline the role of Arl2, a small GTPase, as an important regulator of breast tumor cell Aggressivity, both in vitro and in vivo.

    This study investigated the mediating effect of trait Aggressivity on the relation between agreeableness and alcohol-related aggression in a laboratory setting. Participants were 116 healthy male social drinkers between 21 and 30 years of age. Agreeableness and trait Aggressivity were measured using the Big Five Inventory and the Buss-Perry Aggression Questionnaire, respectively. Following the consumption of an alcohol or no-alcohol control beverage, participants completed a modified version of the Taylor Aggression Paradigm, in which electric shocks were received from and administered to a fictitious opponent during a competitive task. Aggression was operationalized as the proportion of the most extreme shocks delivered to the fictitious opponent under conditions of low and high provocation. Results indicated that lower levels of agreeableness were associated with higher levels of trait Aggressivity. In turn, higher levels of trait Aggressivity predicted extreme aggression in intoxicated, but not sober, participants under low, but not high, provocation. Findings highlight the importance of examining determinants of intoxicated aggression within a broader theoretical framework of personality.

    Abstract:

    The research literature on the relation between cognitive functioning and alcohol consumption is inconsistent and difficult to interpret. The purpose of this study was to test a causal model that might help reconcile some of these conflicts. The model specifies that Aggressivity is an important intermediary mechanism underlying the relation between executive functioning (EF) and alcohol consumption. Participants were 310 (152 men and 158 women) healthy social drinkers between 21 and 35 years of age tested in a laboratory setting. EF was measured with a battery of neuropsychological tests. Aggressivity and alcohol consumption (as well as cigarette and drug use frequency) were measured with self-report and interview formats. Aggressivity played a mechanistic role in the relation between EF and alcohol/drug use for men but not for women. Women evinced some unexpected positive relations between EF and alcohol use. This study serves as a first step in trying to reconcile previous inconsistent findings regarding the relation between cognitive functioning and alcohol use by demonstrating that a better understanding of this relation involves considering Aggressivity as an intermediary variable.

Method: Subjects were 204 healthy social drinkers (111 men) between 21 and 35 years of age. Dispositional Aggressivity was measured using the Buss-Perry Aggression Questionnaire. Following the consumption of either an alcohol or placebo beverage, subjects were tested on a modified version of the Taylor Aggression Paradigm, in which electric shocks are received from and administered to a fictitious opponent during a competitive task.     This report examines relative vehicle crashworthiness and Aggressivity of the Australian light passenger vehicle fleet in the four major crash types in which they are involved. These are single vehicle crashes, crashes with unprotected road users such as pedestrians and bicyclists, crashes with heavy vehicles and crashes with other light passenger vehicles. Analysis focuses on 1982-2000 model vehicles classified into 8 broad market groups. The ratings used were based on data on crashes in Victoria and New South Wales during 1987-2000 and in Queensland and Western Australia during 1991-2000. Results of the research have been able to identify the vehicle market groups that have relatively good and bad secondary safety performance in each crash type considered. Results have shown clear differential relative performance between market groups across the major crash types considered. They have also been able to quantify the relative risk of serious injury or death between crash types, further highlighting the high serious injury risk to unprotected road users in particular, as well as to light vehicle drivers in both single vehicle crashes and in crashes with articulated heavy vehicles.     Crashworthiness and Aggressivity ratings for 1982-2002 model vehicles were developed based on data on crashes in Victoria and New South Wales during 1987-2002 and in Queensland, Western Australia and New Zealand during 1991- 2002. Crashworthiness and Aggressivity were measured by a combination of injury severity (of injured drivers) and injury risk (of drivers involved in crashes). The crashworthiness estimates and their associated confidence limits were obtained for 255 vehicle models and were sufficiently sensitive that they were able to identify 128 models of passenger cars, four-wheel drive vehicles, passenger vans and light commercial vehicles that have superior or inferior crashworthiness characteristics compared with the average vehicle. Aggressivity rating estimates and their associated confidence limits were obtained for 202 vehicle models and were sufficiently sensitive that they were able to identify 80 models of passenger cars, four-wheel drive vehicles, passenger vans and light commercial vehicles that have superior or inferior Aggressivity characteristics compared with the average vehicle.     The relationship between vehicle Aggressivity and the year of manufacture of Australian passenger and light commercial vehicles manufactured from 1964 to 2000 was investigated. Trends were examined by year of manufacture both for the fleet as a whole and by market group for vehicles manufactured from 1982 to 2000. Aggressivity was measured by a combination of injury severity (of injured drivers) and injury risk (of drivers involved in crashes). The ratings were adjusted for the sex and age of the focus driver (that is, the driver of the vehicle impacted by the vehicle of the specific year of manufacture and/or market group), the speed limit at the crash location, the state in which the crash occurred and the year in which the crash occurred. These factors were strongly related to injury risk and/or severity for Aggressivity. The Aggressivity rating estimates the risk of the focus driver being killed or admitted to hospital when involved in a tow-away crash, to a degree of accuracy represented by the confidence limits of the rating in each case. The results of this report are based on a number of assumptions and warrant a number of qualifications that should be noted. (a)

    This paper describes an empirical comparison of seven vehicle Aggressivity rating methods in order to identify the most satisfactory vehicle Aggressivity 'rating' system. Vehicles were distinguished by make and model, and the Aggressivity of each model was estimated from data on real two-vehicle crashes. For comparison purposes, two common crash databases were used for estimation of ratings by each method: Police-reported tow-away crash data from three US states, and accident compensation claims from Finland. New methods of vehicle Aggressivity rating were also proposed and tested.
