    Abstract:

    O estudo de novos fármacos e a otimização dos processos de biodistibuição, tem sido alvo de diversos estudos em nanobiotecnologia, com essa abordagem o presente trabalho tem como principal objetivo interligar o sistema microscópio, advindo de polímeros naturais e de berberina, que formaram microesferas, com nanopartículas de ouro obtidas a partir da metodologia modificada de Turchevich. No sistema de nanopartículas de ouro foi associado antibiótico citostático doxorrubicina, sendo esta associação realizada por duas metodologias: associação eletrostática junto as cargas de íons citrato na superfície da partícula de ouro; ou por acoplamento EDC/NHS do fármaco a superfície modificada das nanopartículas de ouro pelo THPAL. Os sistemas micro e nanoestruturados foram avaliados frente a características físico-químicos tais como: distribuição de tamanho, potencial zeta, analises térmicas, análises espectroscópicas de absorção na região Uv-vis, infravermelho com transformada de Fourie e análises de estrutura morfológica utilizando microscopia eletrônica de transmissão e varredura. Os resultados obtidos com ambos os sistemas micro e nanoestruturado isoladamente e associados mostraram que os sistemas mistos obtidos são promissores para o carreamento de fármacos. As microesferas apresentaram diâmetro médio de 0,65 mm quando secas e entumescidas de 0,85 mm, com parâmetros físico-químicos medidos (0,25 ± 0,05) fração de água retida, (30,15 ±10,6%) percentual de hidratação, (24,55±5,3%) inchaço e 1,25 ± 0,05 difusão. As análises térmicas dos sistemas mistos contendo ou não AuNPs mostraram ter transição vítrea no geral até a temperatura de 80ºC e que há uma diferença entre os sistemas que contem AuNP quanto a perda de massa (TGA). ______________________________________________________________________________________________ ABSTRACTThe study of new drugs and the optimization of biodistribution processes, has been the subject of several studies in nanobiotechnology, with this approach the present work aims to link the microscope system, arising from natural polymers and berberine, which formed microspheres with gold nanoparticles obtained from Tuchevich methodology. In the gold nanoparticles system has been associated Antineoplastic Antibiotic, doxorubicin citrate particles and particles coupled to THPAL and covalently linked to doxorubicin. Were preparation of the systems evaluated the physicochemical and morphological parameters, such as, size, zeta potential, simultaneous differential thermal and thermogravimetric, electronic absorption spectra in the UV-vis region, FTIR transform infrared Fourier electronic microscopy scanning and transmission. The results obtained with both micro- and nanostructured systems and associated alone showed that the obtained mixed systems are promising for the carrying of drugs. The microspheres had an average size of 0.65 mm and 0.85mm, with measured physical and chemical parameters (0.25 ± 0.05) retained water fraction (30.15 ± 10.6%) hydration percentage (24.55 ± 5.3%) swelling and 1.25 ± 0.05 diffusion. The thermal analysis of mixed systems with or without AuNPs shown to have glass transition in general until the temperature of 80ºC and that there is a difference between the systems containing AuNP as the weight loss (TGA)

The cells, the extracellular matrix (ECM) proteins, and the growth factors regulating the synthesis, degradation, and deposition of the ECM proteins will be discussed. Therapeutics with particular emphasis given to gene therapies and their effects on specific signaling pathways are described. Bleomycin (BM), a potent Antineoplastic Antibiotic increases TGF-beta1 transcription, TGF-beta1 gene expression, and TGF-beta protein. Like TGF-beta1, BM acts through the same distal promoter cis-element of the COL1A1 gene causing increased COL1 synthesis and lung fibrosis. Lung fibroblasts exist as subpopulations with one subset predominantly responding to fibrogenic stimuli which could be a specific cell therapeutic target for the onset and development of pulmonary fibrosis.
