    Abstract:

    One of the efforts to increase the production of honey bee ( Apis cerana ) is by giving motivate to farmer and their family, because it can be done by increasing the income can be taken by honey bee cultivation (Apis cerana) in Buana Sakti v illage. This research was done from January until March 2013 to know at the level of farmers’ motivation and also the factors that has correlation and influence with the farmers’ motivation in honey bee ( Apis cerana ) cultivation . Based on the result of some model simulations in test the motivation of the farmers in honey bee ( Apis cerana ) cultivation in Buana Sakti village has been become high classification with result 18,78, and factors that has influence of motivation to farmers in honey bee Apis cerana cultivation is an easy in honey bee culti v ation 90,7%,  an easy of market result honey bee culti v ation 90%, frequency to follow socialitation 89,9%, farmer’s income 85,9%, and a long of honey bee culti v ation 78,8%.

  • Morphology and transcriptome differences between the haploid and diploid drones of Apis cerana

    Abstract:

    Abstract In general, drone honey bees are haploid and develop from unfertilized eggs. However, a diploid drone can arise in an inbred colony. In this study, the morphological characteristics and gene expression profile of the haploid and diploid drones of Apis cerana were analyzed to reveal the differences between them. The ploidy level of the drones was identified by Flow Cytometry (FCM). The characters of the fore wings, wet weight of reproductive organs and of newly emerged drones, were investigated. Then, a high throughput transcriptomic analysis was performed using RNA-seq in diploid and haploid drones. The results showed that the wet weight and reproductive organs of diploid drones were significantly lighter than those of haploid drones. About 201 million high-quality reads were generated from RNA-seq, and 75.99–78.12% of the data were mapped to Apis cerana genome. 360 genes were differentially expressed between diploid and haploid drone, with 152 up-regulated and 208 down-regulated in the diploid drones. Functional analysis identified that these genes were significantly enriched in 28 pathways. Comparative transcriptomic analysis detected several differentially expressed genes, which lay a foundation for future studies on molecular mechanisms underlying biology difference in drones in Apis cerana .

    Abstract:

    Apis mellifera and Apis cerana are two sibling species of Apidae. Apis cerana is adept at collecting sporadic nectar in mountain and forest region and exhibits stiffer hardiness and acarid resistance as a result of natural selection, whereas Apis mellifera has the advantage of producing royal jelly. To identify differentially expressed genes (DEGs) that affect the development of hypopharyngeal gland (HG) and/or the secretion of royal jelly between these two honeybee species, we performed a digital gene expression (DGE) analysis of the HGs of these two species at three developmental stages (newly emerged worker, nurse and forager). Twelve DGE-tag libraries were constructed and sequenced using the total RNA extracted from the HGs of newly emerged workers, nurses, and foragers of Apis mellifera and Apis cerana. Finally, a total of 1482 genes in Apis mellifera and 1313 in Apis cerana were found to exhibit an expression difference among the three developmental stages. A total of 1417 DEGs were identified between these two species. Of these, 623, 1072, and 462 genes showed an expression difference at the newly emerged worker, nurse, and forager stages, respectively. The nurse stage exhibited the highest number of DEGs between these two species and most of these were found to be up-regulated in Apis mellifera. These results suggest that the higher yield of royal jelly in Apis mellifera may be due to the higher expression level of these DEGs. In this study, we investigated the DEGs between the HGs of two sibling honeybee species (Apis mellifera and Apis cerana). Our results indicated that the gene expression difference was associated with the difference in the royal jelly yield between these two species. These results provide an important clue for clarifying the mechanisms underlying hypopharyngeal gland development and the production of royal jelly.

    Abstract:

    Abstract Transgenic Apis cerana cerana were produced by sperm-mediated gene transfer (SMGT). In the experiment, the foreign DNA was linearized and introduced with sperm during the instrumental insemination of virgin queen. The descendants of the experimental colonies were analyzed. In the green fluorescent-positive offspring, green fluorescence was observed for 1- to 2-day-old larvae, the predicted fragment was isolated by means of PCR amplification of genomic DNA and the expression of transferred genes was confirmed at transcriptional level by reverse transcription-polymerase chain reaction. These results showed that the exogenous gene could be integrated in a fraction of the germ line cells of the queen Apis cerana cerana and transmitted to offspring by SMGT.

    Abstract:

    In this study,the virgin queens of Apis cerana cerana were equipmently inseminated with a single male,2 males or mated naturally with more than 2 males.The unfertilized eggs laid by queens(QUGs) and workers(WUGs) were policed by the workers in each colony following standard methods.The results showed that: there was worker policing both in naturally mated colonies and in the equipmently inseminated coloyies of Apis cerana cerana.

    Abstract:

    In the present investigation,the DNA from fresh royal jelly of Apis cerana was extracted with modified phenol-chloroform method,and its content and purity were determined.The results showed that the mean content of DNA extracted from fresh royal jelly of Apis cerana was(82.95 ±10.39)μg/g,the mean purity(A_260/A_280)was 1.69±0.08.Taking the DNA as the template for RAPD-PCR,the obtained bands were clear,bright and complete.
