    Pure autonomic failure (PAF) is an idiopathic sporadic Disorder characterized by orthostatic hypotension with evidence of more widespread autonomic failure. It has been suggested that PAF belongs to the group of synucleinopathies, as autopsy studies showed α-synuclein-positive Lewy bodies in sympathetic neurons of patients with PAF.1 REM sleep Behavioral Disorder (RBD) appears to be an almost universal feature of synucleinopathies. Whereas initial studies suggested that RBD specifically occurs in multiple system atrophy (MSA), subsequent studies showed that it is also a frequent feature of Parkinson disease (PD) and dementia with Lewy bodies (DLB).2 Furthermore, patients with isolated RBD often later develop PD.3 We here present a consecutive unselected series of three patients with PAF (M/F: 2/1, …

    Abstract:

    : The sapogenin isolated from seeds of Camellia oleifera shows central effects in our previous research. Aiming to improve its neuroprotective effects, we synthesize the zinc-sapogenin complex, which has been found with the nanoparticle characters and neuroprotective effects. Its structure is deduced as sapogenin:zinc:water (3:1:1) by ultraviolet spectra, infrared spectra, element analysis, and thermogravimetric analysis. The average size of zinc-sapogenin is 115 nm with plane circular shape by measurement of particle analyzer and scanning electron microscope. The nanoparticle improves DPPH radical scavenging activity due to the enhancement of electron transfer among atoms through the linkage of zinc with the sapogenin. Intraperitoneal injection of zinc-sapogenin reduced mice Behavioral Disorder and neuron damage induced by neurotoxicity of rotenone, increased antioxidative ability and promoted the neurotransmitters of dopamine and acetylcholine in mice brain. Zinc strengthens stability and effects of the sapogenin, and it is a superior candidate of drugs for neuroprotection without accumulation in brain.

    Abstract:

    Sasanqua saponin is a major active compound in the defatted seeds of Camellia oleifera but is always discarded without effective utilization. The sapogenin from hydrolysis of sasanqua saponin was purified, and its amination derivative was investigated on its neuroprotective effects, which were evaluated by animal models of Parkinson disease in mice induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP). The results showed that the sapogenin and its derivative increased dopamine content in striatum and tyrosine hydroxylase (TH) positive cells in substantia nigra and relieved inflammation and Behavioral Disorder, but the effect on movement was reversed by dopamine receptor antagonist haloperidol and was not intervened by adenosine receptor antagonist CGS 15943. Molecular simulation showed the interaction between dopamine receptor and the sapogenin or its derivative.
