Supercritical Carbon Dioxide Extraction of Seed Oil from Winter Melon (Benincasa hispida) and Its Antioxidant Activity and Fatty Acid Composition

Zaidul Islam, Taip, Farah Saleena, Rahman Russly, Ganjloo AliAbstract:In the present study, supercritical carbon dioxide (SC-CO2) extraction of seed oil from winter melon (Benincasa hispida) was investigated. The effects of process variables namely pressure (150–300 bar), temperature (40–50 °C) and dynamic extraction time (60–120 min) on crude extraction yield (CEY) were studied through response surface methodology (RSM). The SC-CO2 extraction process was modified using ethanol (99.9%)as co-solvent. Perturbation plot revealed the significant effect of all process variables on the CEY. A central composite design (CCD) was used to optimize the process conditions to achieve maximum CEY. The optimum conditions were 244 bar pressure, 46 °C temperature and 97 min dynamic extraction time. Under these optimal conditions, the CEY was predicted to be 176.30 mg-extract/g-dried sample. The validation experiment results agreed with the predicted value. The antioxidant activity and fatty acid composition of crude oil obtained under optimized conditions were determined and compared with published results using Soxhlet extraction (SE) and ultrasound assisted extraction (UAE). It was found that the antioxidant activity of the extract obtained by SC-CO2 extraction was strongly higher than those obtained by SE and UAE. Identification of fatty acid composition using gas chromatography (GC) showed that all the extracts were rich in unsaturated fatty acids with the most being linoleic acid. In contrast, the amount of saturated fatty acids extracted by SE was higher than that extracted under optimized SC-CO2 extraction conditions

ZaidulAbstract:In this study, the effect of different solvent including ethanol, n-hexane and ethyl acetate on antioxidant activity and total phenolic content (TPC) of winter melon (Benincasa hispida) seeds extract was investigated using conventional Soxhlet extraction (CSE). DPPH and ABTS scavenging activity and TPC results indicated that the seed extracts obtained using ethanol possessed the highest antioxidant activity and followed by ethyl acetate and n-hexane. By considering obtained results, it was clear that there was a high positive correlation between TPC and antioxidant activity. Linoleic acid forms a significant percentage of unsaturated fatty acids of the seed extract (60.6%). It is well known that essential fatty acids including linoleic acid and linolenic acid which are detected in extracts play important roles in preventing many disease and abnormal differentiation problems. B. hispida seeds are potential source of natural antioxidant compounds to replace synthetic antioxidants.

Comparison of different extraction methods for the extraction of total phenolic compounds from winter melon (Benincasa hispida) seeds

Effects of different UAE variables such as amplitude (25-75%), temperature (45-65 °C) and sonication time (20-60 min) on the TPC of extracts was determined using complete randomised design (CRD). The results showed that the TPC of extracts obtained UAE process significantly affected by independent variables. The maximum value of TPC (22.12 ± 1.23 mg GAE/g extract) was obtained at the combined treatment conditions of 75%, 55 °C and 40 min. The UAE results obtained were compared with those achieved by using conventional Soxhlet extraction (CSE) method. It was found UAE allowed extraction at lower temperature and the extracts obtained posses higher quality compare with CSE. UAE is a promising environment friendly technique for the extraction of bioactive compounds from winter melon (Benincasa hispida) seeds

Extraction of valuable compounds from winter melon (Benincasa hispida (thunb.) cogn.) seeds using ultrasonic-assisted solvent, supercritical carbon dioxide and pressure-swing methods
2012Co-Authors: B MandanaAbstract:Different extraction techniques have been used to isolate valuable compounds from different plants. Conventional Soxhlet extraction (CSE) is a standard technique which has been used for a long time. The main disadvantages of conventional techniques are long extraction time which is not always accepted by industries. This extraction method is also not suitable for the extraction of thermo-sensitive compounds due to the possibility of thermal decomposition of target compounds as extraction usually occurs at the boiling point of used solvent for a long time. The current study presented the effect of various extraction methods including conventional Soxhlet extraction (CSE), ultrasound-assisted extraction (UAE),supercritical carbon dioxide (CO2) extraction (SCE) and SCE with pressure swing technique (SCE-PST) for extraction of valuable compounds from winter melon (Benincasa hispida (Thunb.) Cogn.) seeds. Parameters examined were crude extraction yield (CEY), radical scavenging activity (RSA) in terms of 1, 1-diphenyl 2-picrylhydrazyl (DPPH˙) radicals scavenging percentage (%DPPHsc) and 2,2´-Azinobis (3-ethylbenzothiazoline-6-sulphonic acid) diammonium salt (ABTS+˙) radicals scavenging percentage (%ABTSsc) and total phenolic content (TPC) of B.hispida seeds. The overall optimum conditions for obtaining maximum dependent variables were predicted using response surface methodology (RSM). Under the optimised conditions, the fatty acid composition of extracts was identified and quantified using gas chromatography (GC). In CSE different solvents (n-hexane, ethanol and ethyl acetate) were used and the highest CEY (294.45 ± 1.22 mgg-1) obtained using ethyl acetate. Ethanol (EtOH) extracts showed the highest %DPPHsc and %ABTSsc (28.7 ± 0.7 and 27.0 ± 0.9, respectively) while n-hexane extracts possessed the lowest (13.1 ± 0.9 and 12.2 ± 0.6, respectively). TPC ranging from zero in n-hexane extract to 11.34 ± 1.3 mg GAE/g extracts obtained using EtOH which revealed the antioxidant potency of B. hispida seeds. Perubahan Kadar Glukosa Darah Kelinci Putih Jantan Akibat Air Perasan Buah Labu Bligo (Benincasa hispida Cogn.) sebagai Obat Alternatif Sakit Kencing Manis

Salah satu dari obat tradisional yang diduga dapat menurunkan kadar glukosa darah adalah labu bligo atau labu tangkua. Pada penelitian ini telah dilakukan uji efek anti diabetes dari air perasan buah labu bligo (Benincasa hispida cogn.) terhadap kelinci putih jantan diabetes karena pemberian streptozotocin. Dalam penelitian ini kelinci putih jantan dibagi menjadi 3 kelompok. Kelompok kontrol diberi air suling 8 ml/kg BB, kelompok pembanding diberi glucophage 500 mg/kg BB dengan kadar 50% dan kelompok uji diberi air perasan buah labu bligo 8 ml/kg BB. Berdasarkan penelitian yang telah dilakukan didapat hasil bahwa air perasan buah labu bligo dapat menurunkan kadar glukosa darah kelinci putih jantan sebesar 3,89% dibandingkan kelompok kontrol dan sebesar 5,84% dibandingkan kelompok pembanding. Jadi air perasan buah labu bligo efektif untuk menurunkan kadar glukosa darah pada kelinci putih jantan diabetes akibat streptozotocin. Pengobatan dari diabetes mellitus adalah dengan penggunaan obat antidiabetik oral dan insulin. Sementara itu penggunaan tanaman sebagai bahan obat tradisional memerlukan penelitian ilmiah untuk mengetahui kebenaran. Pada penelitian ini telah dilakukan uji efek antidiabetes air perasan buah labu bligo (Benincasa hispida Cogn.) terhadap kelinci putih jantan dengan metode toleransi glukosa. Dalam penelitian ini kelinci putih jantan dibagi menjadi 3 kelompok. Kelompok I diberi air suling 5 ml/kg BB, kelompok II diberi suspensi glibenklamid 5 mg/kg BB dan kelompok III diberi air perasan buah labu bligo (Benincasa hispida Cogn.) dosis 8 ml/kg BB. Dari hasil penelitian ini dapat ditarik kesimpulan bahwa air perasan buah labu bligo dapat menurunkan kadar glukosa darah kelinci putih jantan sebesar 97,92% dibanding kelompok kontrol (pemberian air suling) dan sebesar 70,03% dibanding kelompok pembanding (pemberian suspensi glibenklamid)

Inter-varietal variation in the composition of seeds and seed oils from winter mel on [Benincasa hispida (thunb.) cogn.] fruits.

In the present work, physicochemical properties of seeds and the extracted seed oils were examined and compared among three cultivars namely round, oval and hybrid of winter melon. The seeds from round, oval and hybrid fruits, exhibited protein, fiber and ash contents 28.18-42.03, 19.36-26.21 and 5.02- 11.81%, respectively. The oils were extracted based on Soxhlet method by petroleum ether, whilst yields ranged from 17.78- 32.53% (wt/wt). The extracted oils were analyzed for physicochemical parameters, and fatty acids, tocopherols and sterols profiles. The results for specific gravity (25°C), refractive index (nD 25°C), iodine value (IV), saponification value (SV), peroxide value (PV), and free fatty acid (% as oleic acid) were 0.89-0.91 g/mL, 1.4627-1.4646, 119.9-125.1 g I/100 g oil, 182.3-194.1 mg KOH/ g oil, 1.13-1.33 mequiv.O2 /kg, 1.57-2.10%, respectively. The oil color intensity in terms of yellow and red units was 6.9Y + 1.0 R to 8.9Y + 1.9R. The amounts of oil tocopherols as analyzed by HPLC varied widely among the cultivars tested showing α-tocopherol 31.1-207.6 mg/kg and δ-tocopherol 60.4-146.0 mg/kg. According to the GLC analysis linoleic acid (C18:2) was established to be the principal fatty acid (63.10-70.64%) followed by C16:0 (12.45-17.59), C18:1 (8.46-12.87%) and C18:0 (5.13-7.48%). Analysis of oil sterol fractions, using GC and GC-MS, revealed the presence of β-sitosterol (54.62–60.50%), campesterol (15.10–18.50%), stigmasterol (11.00–14.30% and Δ 5 -avenasterol (6.40–8.14 %) as the four main components. Most of the properties of the seed oils analyzed varied significantly among fruit cultivars tested. Overall, we concluded that the seeds, which are under-utilized and often discarded as an agrowate, from winter melon should be explored for extraction of high-linoleic oil with additional tocopherols and phytosterol benefits

Cogn.], a member of the family Cucurbitaceae, is one of the famous crops that are grown primarily for its fruits and usually recognized with its nutritional and medicinal properties especially in Asian countries. Kundur fruit has been valued as a nutritious vegetable as it provides a good source for natural sugars, amino acids, organic acids, mineral elements and vitamins. A number of medicinal properties such as anti-diarrheal, anti-obesity, anti-ulcer, and antioxidant and diuretic have been ascribed to this fruit of high economic value. As a rich source of functionally important bioactives and therapeutics such as triterpenes, phenolics, sterols, and glycosides, the fruit has been widely used for the treatment of epilepsy, ulcer, and other nervous disorders in the native medicine system of Asia. Nutritional composition and oil fatty acids of Kundur [Benincasa hispida (Thunb.) Cogn.] seed

Cogn.], a fruit vegetable plant with high functional properties (especially in medicinal treatment), were analysed for nutritive parameters (dietary fiber, crude protein, crude fat, crude fiber, ash, and energy) and oil fatty acids composition. Proximate analysis of the seeds revealed the total dietary fiber as the principal component, comprising 58.43% of the seed. The contents of crude fat and crude protein were found to be 20.70 and 11.63%, respectively. The extracted Kundur seed oil mainly consisted of linoleic acid (C18:2ω6), accounting for 67.37% of the total fatty acids. Other important fatty acid detected were palmitic (C16:0), oleic (C18:1 cis) and stearic (C18:0) acids with contribution of 17.11, 10.21 and 4.83%, respectively. The fatty acids profile of the tested Kundur seed oil was quite comparable to those of previously reported cucurbitaceae seed oils and some other wholesome oils, revealing that it can be used as a potential seed oil crop
