NussdorferAbstract:: The mechanism of the trophic effect of chronic sodium restriction on the zona glomerulosa of the rat adrenals was investigated by combined morphometric and Biochemical Techniques. In the normal rats, prolonged sodium deprivation caused a significant hypertrophy of the zona glomerulosa and its parenchymal cells, coupled with a conspicuous rise in the plasma concentration of aldosterone. Analogous results were observed in animals simultaneously administered captopril and maintenance doses of angiotensin II. However, in this last case, the changes were significantly lower than those observed in the normal rats. These findings are compatible with the view that the activation of the renin-angiotensin system is not the sole mechanism involved in the stimulation of the growth and steroidogenic capacity of rat adrenal zona glomerulosa induced by chronic sodium restriction.

NussdorferAbstract:: The effects of chronic administration of metoclopramide or bromocriptine (two drugs which act as antagonist and agonist of dopamine receptors) on the zona glomerulosa of captopril-treated rats administered maintenance doses of angiotensin II, were investigated by combined morphometric and Biochemical Techniques. It was found that metoclopramide provoked a significant hypertrophy of the zona glomerulosa and its parenchymal cells, coupled with a persistent rise in the plasma concentration of aldosterone, only in rats fed a normal diet. Conversely, bromocriptine exerted an evident inhibitory effect on the zona glomerulosa growth and steroidogenic capacity only in chronically sodium-deprived animals. These findings strongly suggest that sodium balance modulates the activity of the dopaminergic system, whose chronic suppression may be involved in the mechanism underlying the extra-angiotensin adrenoglomerulotrophic effect of prolonged sodium restriction.

Isolation and Purification of the Schizont Stage of Theileria annulata from Host Leukocytes through Novel Biochemical Techniques

This uncontrolled replication happens in the macroschizont stage of the life cycle of the parasites. The development of a rapid and efficient technique is likely to necessitate for isolation of purified schizonts from host cells. This is necessary for the isolation of highly purified RNA, protein or glycoproteins of schizonts from host cells. This study aimed to isolate the purified schizont based on the aerolysin–nocodazole technique. Aerolysin which was purified from gram-negative pathogen aeromonas hydrophila has an ability to form discrete channels and unstable eukaryotic cell membranes in 0C• and low concentration. Nocodazole used for parasite separation from the microtubule network of infected lymphocytes and monocytes. In purified schizonts, no large nucleus of host cells visualized in giemsa and DAPI staining. The isolated schizonts were free and intact from host cells. Intact T.annulata schizonts obtained from this study are suitable for purification of RNAs, proteins, glycolipids, and glycoproteins of schizonts free of host cell debris.

Materials and Methods: This study approved based on the aerolysin - nocodazole. Aerolysin that purified from gram-negative pathogen aeromonas hydrophila has an ability to form discrete channels and unstable a eukaryotic cell membrane in 0°C and low concentration. Nocodazole used for parasite separation from Microtubule network of infected lymphocytes and monocytes. Results: In purified schizonts, no large nucleus of host cell visualize in Giemsa and 4’,6-diamidino-2-phenylindole staining. Schizonts are free and intact from host cells. Conclusions: intact T. annulata schizonts suitable for RNAs, proteins, glycolipids, and glycoproteins of schizonts free of host debris.

Identifying and characterizing the neuronal proteins interacting with AMPA-Rs have provided important information about the molecular mechanisms underlying synaptic transmission and plasticity. In this study, to identify more AMPA-R interactors in vivo, we performed proteomic analyses of AMPA-R complexes from the brain. AMPA-R complexes were isolated from the brain through various combinations of Biochemical Techniques for solubilization, enrichment, and immunoprecipitation. Mass spectrometry analyses of these isolated complexes identified several novel components of the AMPA-R complexes as well as some previously identified components. The identification of these novel components helps to further define the complex mechanisms involved in the regulation of AMPA receptor function and synaptic plasticity.
