Characterization of the promoter region of the bovine SIX1 gene: Roles of MyoD, PAX7, CREB and MyoG

Previous studies have demonstrated that SIX1 is positively correlated with Body Measurement traits (BMTs). However, the transcriptional regulation of SIX1 remains unclear. In the present study, we determined that bovine SIX1 was highly expressed in the longissimus thoracis. To elucidate the molecular mechanisms involved in bovine SIX1 regulation, 2-kb of the 5′ regulatory region were obtained. Sequence analysis identified neither a consensus TATA box nor a CCAAT box in the 5′ flanking region of bovine SIX1. However, a CpG island was predicted in the region −235 to +658 relative to the transcriptional start site (TSS). An electrophoretic mobility shift assay (EMSA) and chromatin immunoprecipitation (ChIP) assay in combination with serial deletion constructs of the 5′ flanking region, site-directed mutation and siRNA interference demonstrated that MyoD, PAX7 and CREB binding occur in region −689/−40 and play important roles in bovine SIX1 transcription. In addition, MyoG drives SIX1 transcription indirectly via the MEF3 motif. Taken together these interactions suggest a key functional role for SIX1 in mediating skeletal muscle growth in cattle.

Novel polymorphisms of SIX4 gene and their association with Body Measurement traits in Qinchuan cattle.

SIX4 gene plays a crucial role in skeletal myogenesis, and its genetic variations or deficiency may cause hypopituitarism, suggesting that SIX4 gene is a potential candidate gene affecting Body Measurement traits (BMTs) in animals. Herein, the objectives of this study were to identify genetic polymorphisms of bovine SIX4 gene and to analyze potential association between single nucleotide polymorphisms (SNPs) and Body Measurement traits in Qinchuan cattle. In the present study, we investigated polymorphisms of SIX4 gene in 426 Qinchuan cattle using DNA sequencing and polymerase chain reaction–restriction fragment length polymorphisms. The polymorphisms of LYRM1 gene and their association with Body Measurement and ultrasound traits of Qinchuan cattle

Latest studies showed that LYR motif containing 1 (LYRM1) may be involved in influencing fatness deposition in animals. The objective of this study was to detect bovine LYRM1 gene polymorphism and analyze its association with Body Measurement and meat quality traits of cattle. Blood samples were taken from a total of 404 Qinchuan cattle aged from 18–24 months. Created restriction site-polymerase chain reaction-restriction fragment length polymorphism (CRS-PCR–RFLP) and DNA sequencing were used to find out LYRM1 single polymorphism nucleotide (SNPs). Sequence analysis of LYRM1 gene revealed two SNPs (g.165 C > A, g.193 A > G) in 3′ untranslated region (3′UTR) of exon 3. And g.165 C > A showed two genotypes namely AC and CC while g.193 A > G showed three genotypes: AA, AG and GG. Analysis results showed that there were significant associations between polymorphism of these two and Body Measurement and meat quality traits in Qinchuan cattle population. Based on the results obtained from this study, it is inferred that LYRM1 gene may have potential effects on Body Measurement and meat quality traits in Qinchuan cattle population and could be used for marker-assisted selection.

Abstract A113: Association between NSAIDs and PSA, prostate volume, and prostate cancer

SmithAbstract:Abstracts: Frontiers in Cancer Prevention Research 2008 A113 Purpose NSAIDs such as aspirin are commonly used to prevent cardiovascular disease, and several prior studies suggest NSAIDs may also reduce prostate inflammation and prostate cancer risk. We investigated the association between NSAID use, PSA, and prostate volume, hypothesizing lower PSA and volume levels with NSAID use. Methods The Nashville Men’s Health Study utilizes a multi-centered, rapid-recruitment protocol to collect clinical, biological, behavioral, and Body Measurement data from 1,277 men over age 40 years and scheduled for diagnostic prostate biopsy. Approximately 95% of eligible men approached for recruitment agree to participate. NSAID use was ascertained by survey and clinical interview, and medical charts were reviewed to ascertain current PSA levels, prostate volume, and clinical diagnoses following biopsy. The distributions of PSA level and prostate volume data were natural log transformed prior to analysis. In a linear model, mean PSA and volume scores across NSAID categories were adjusted for age; race (black/white); family history (yes/no); number of prior PSA tests (1,2, 3 or more); BMI; WHR; height; or treatment for BPH (steroid reductase inhibitors, other), diabetes (Yes/No), CVD (Yes/No), hyperlipidemia (Yes/No); and the diagnosis of PIN, atypical findings suspicious for cancer, low-grade cancer, or high-grade cancer following biopsy. PSA and prostate volume scores were back-transformed and geometric mean values are reported. Results Approximately 46% of subjects reported taking an NSAID, primarily aspirin (37% of subjects). After adjusting for age, race, family prostate cancer history, obesity, and treatment for BPH, CVD, hyperlipidemia, and diabetes; aspirin was significantly associated with lower PSA levels (7.3 vs. 8.0 ng/ml, p=0.01). Characterization of leptin receptor gene in Bubalus bubalis and association analysis with Body Measurement traits.

This property supports leptin and its receptor as candidate genes for evaluating genetic polymorphisms to associate with growth, milk yield and other economic traits. The aim of this study is to characterize the leptin receptor gene in Bubalus bubalis, to identify single-nucleotide polymorphism (SNP) sites in different coding and non-coding regions and to analyse potential associations between SNPs identified and the Body Measurements traits of growing buffalo heifers. A group of 64 animals were genotyped by direct sequencing and twenty-eight SNPs were detected. A sequence analysis revealed the presence of nine interesting SNPs in gene sequence. The association analysis of polymorphisms with the Body Measurements traits of growing buffalo heifers shows significant statistical effects on chest depth and sacrum height. Therefore according to the results obtained from this study, the leptin receptor gene appears to have potential effects on the Body Measurement traits of Bubalus bubalis.
