    Abstract:

    Abstract — We demonstrated the presence of anthocyanic vacuolar inclusion (AVI) in anthocyanin-producing cells of Camptotheca acuminata Decne cultured in vitro. During the four cell growth cycles, the number of cells with a col-ourless vacuole decreased. The percentage of cells with AVIs increased during the first and second cell growth cycles and then decreased. This decrease was accompanied by a considerable increase in the percentage of cells with no AVIs, with anthocyanins dissolved in the vacuolar solution. The cytochemical analysis applied to the transmission electron microscopy, revealed the presence of a protein component in the shell of the AVIs.     Abstract The metabolic profile of secondary products in calli and cell suspension cultures of Camptotheca acuminata Decaisne was investigated and compared to that of the leaves and roots taken from the plant. Neither in vitro system produced the anticancer quinoline alkaloid camptothecin (CPT); they accumulated discrete quantities of polyhydroxylated triterpenoids, different from those found in the plant organs, and ellagic acid derivatives. Nine ellagic acid derivatives ( 1a – 1d and 2a – 2e ) and eight triterpenoid acids ( 3a – 3e and 4a – 4c ) were isolated and characterised. All the identified triterpenes were related to ursane- or oleanane-type skeletons and their concentrations rose to 4.5% in suspended cells.

    Abstract:

    In this paper, a system of laticifers in Camptotheca acuminata Decne (Nyssaceae) is described. Laticifers were already present in the leaf primordia of the shoot apex. In the mature leaves, laticifers were found in the midrib and in the larger veins, both in the parenchymatic region delimited by vascular bundles and in the cortex just external to the phloem. In the stem, laticifers were present in both the primary and secondary body, running parallel to the longitudinal axis. They were located in the pith and in the cortex proximal to the phloem. No laticifers were found in the roots. The histochemical analyses indicated that the main compounds accumulated in laticifers were phenols. Neutral lipids and fatty acids were also present. Ultrastructural observations showed osmiophilic globules both in the vacuoles and in the peripheral regions of the cytoplasm of the laticifer cells. Plastids were present, although altered, with some parallel membranes and lacking starch grains. The discovery in C. acuminata of a laticifer system, which had never been described for the order Cornales, could be of taxonomic value, also considering that this order has traditionally represented one of the most problematic groups of flowering plants.

    An ionic liquid-based ultrasonic/microwave-assisted simultaneous distillation and extraction (IL-UMASDE) method for isolating camptothecin (CPT), 10-hydroxycamptothecin (HCPT), vincoside-lactam (VCS-LT), and essential oils (EOs) from Camptotheca acuminata Decne fruits was developed. The important parameters were optimized using single-factor and central composite design experiments. The optimum conditions were 0.75 M 1-octyl-3-methylimidazolium ([C8mim]Br) as the extraction solvent, a liquid–solid ratio of 13.7 mL/g, an extraction time of 33.2 min, a microwave power of 582 W, and a fixed ultrasonic power of 50 W. The yields of CPT, HCPT, and VCS-LT obtained under the optimum conditions were 2.463, 0.164, and 0.297 mg/g, respectively; these are 1.08-, 1.12-, and 1.04-fold higher, respectively, than those obtained by conventional 55% ethanol heat reflux extraction (HRE). The extraction time for the equilibrium yields of CPT, HCPT and VCS-LT using IL-UMASDE was 33.6%, 58.5%, 63.1%, and 66.8%, respectively, less than the corresponding times using IL-MASDE, IL-ultrasonic-assisted extraction (IL-UAE), 55% ethanol UAE and 55% ethanol HRE. The yield of EOs obtained using IL-UMASDE was 0.793 mg/g, i.e., 1.31-fold higher than that obtained by conventional hydrodistillation extraction (HDE). The components of the EOs obtained using IL-UMASDE and HDE were similar. The extraction time for the equilibrium yields of EOs using IL-UMASDE is 33.6%, 58.5%, 52.6%, and 72.3% lower than those for IL-MASDE, water-UMASDE, water-MASDE, and HDE, respectively. The ionic liquids with different cations and anions have been investigated, and 0.75 M 1-octyl-3-methylimidazolium bromide was selected as solvent. In addition, the ultrasonic parameters including ultrasonic power, time for ultrasonic treatment and solid–liquid ratio have been optimized by Response Surface Methodology. Compared with conventional solvent extraction, ILUAE exhibited higher efficiency (camptothecin increased by 13.96% and 10-hydroxycamptothecin increased by 24.04%, respectively) and shorter extraction time (decreased from 6 h to 1.75 h for extracting three times), which indicated that ILUAE was an efficient, rapid and simple sample preparation technique.

    A rapid and sensitive liquid chromatography-tandem mass spectrometry method with multiple reaction monitoring was developed for the simultaneous determination of camptothecin and 10-hydroxycamptothecin in Camptotheca acuminata. The separation of camptothecin and 10-hydroxycamptothecin was performed on an Agilent Eclipse XDB-C18 column with a mixture of methanol and water (1:1, v/v) containing 0.2% formic acid as a mobile phase. The limits of detection of camptothecin and 10-hydroxycamptothecin were 4.0 ng/mL and 7.0 ng/mL (S/N = 3), respectively. Analysis took 10 minutes, making the method suitable for rapid determination of camptothecin and 10-hydroxycamptothecin in C. acuminata.

    Abstract:

    Plantlet regeneration through shoot formation from young leaf explant-derived callus of Camptotheca acuminata is described. Calli were obtained by placing leaf explants on Woody plant medium (WPM) supplemented with various concentrations of 6-benzyladenine (BA) and naphthaleneacetic acid (NAA) or 2,4-dichlorophenoxyacetic acid (2,4-D). Callus induction was observed in all media evaluated. On the shoot induction medium, the callus induced on the WPM medium containing 19.8 μM BA and 5.8 μM NAA was the most effective, providing high shoot regeneration frequency (70.3 %) as well as the highest number of shoots (11.2 shoots explant−1). The good rooting percentage and root quality (98 %, 5.9 roots shoot−1) were achieved on WPM medium supplemented with 9.6 μM indole-3-butyric acid (IBA). 96 % of the in vitro rooted plantlets with well developed shoots and roots survived transfer to soil.

    Abstract:

    The Arcto-Tertiary relict genera, Camptotheca, Davidia and Nyssa represent deep lineages in the asterid order Cornales. Recent phylogenetic studies suggested that these genera should be placed in a newly circumscribed family, Nyssaceae. However, because these analyses were based upon a few genes, it is prudent and necessary to examine further evidence before adopting this taxonomic treatment. In this study, we determined the complete chloroplast (cp) genomes of Camptotheca acuminata, Davidia involucrata, and Nyssa sinensis. Their cp genomes ranged from 156672 to 158409 base pairs, which included 115 genes, and their genome features were highly similar to those of other species within the order Cornales. The phylogenetic relationships among the genera Camptotheca, Davidia, Nyssa, and 23 related taxa in the asterids were analyzed based on 73 protein-coding genes from the cp genomes. All of the previously recognized major clades (namely Cornales, Ericales, Campanulids, and Lamiids) in the asterids, as well as their relationships, were recovered with robust support. A clade including the genera Davidia, Nyssa, Camptotheca, and Diplopanax, was resolved as a well-supported monophyletic group, which was fully separated from the family Cornaceae by the family Hydrangeaceae. Our results provide novel evidence to support the acceptance of the family Nyssaceae outlined by the updated Angiosperm Phylogeny Group.
