STABLE OVEREXPRESSION OF TGF beta 1 IN A TRANSITIONAL Carcinoma Cell Line: IMPACT ON FIBRONECTIN PRODUCTION
The Journal of urology, 1998Co-Authors: Guangjian Zhang, David Ladehoff, William A. SeeAbstract:AbstractIntroduction: TGF beta 1 is a potent modulator of the biology of both benign and neoplastic Cells. Exocrine TGF beta 1 has been shown to alter fibronectin expression of bladder Carcinoma Cell Lines. The present study describes the development of a stable TGF beta 1 overexpressing transitional Carcinoma Cell Line, and evaluates the impact of autocrine TGF beta 1 production on fibronectin expression.Materials and Methods: The human transitional Carcinoma Cell Line 253J was transfected using the pcDNA3/hTGF beta 1 expression vector under control of the CMV promoter. TGF beta 1 mRNA expression was determined by Northern analysis. TGF beta 1 protein levels were analyzed by biological assay. Subsequently, the effect of TGF beta 1 autocrine production on fibronectin expression at both the mRNA and protein level was determined. Results were compared to Cells transfected with the pcDNA3/CAT and non-transfected 253J Cells. Results: Two 253J clones which uniformly expressed TGF beta 1 mRNA at 22 and four-fold...

Results were compared to Cells transfected with the pcDNA3/CAT and non-transfected 253J Cells. Two 253J clones which uniformly expressed TGFbeta1 mRNA at 22 and four-fold increases relative to controls were identified. mRNA overexpression correlated with marked increase in biologically active TGFbeta1 protein production. Autocrine production of Cellular TGFbeta1 showed a positive correlation with fibronectin expression at both the mRNA and protein levels. Autocrine expression of TGFbeta1 increases Cellular fibronectin production in a human transitional Carcinoma Cell Line. Therapeutic strategies altering urothelial production of TGFbeta1 and fibronectin may be a potential strategy to potentiate intravesical BCG activity.

Characteristics of a thyroid Carcinoma Cell Line derived from spinal metastasis.
Bioscience reports, 2016Co-Authors: Zhou Zhenhua, Xu Yan, Xudong Wang, Su Chen, Jianru XiaoAbstract:A thyroid Carcinoma Cell Line named THY28 was established through primary culture of the surgical specimens, which were derived from a Chinese patient with spinal metastasis. The Cell morphology, growth kinetics, Cell cycle, chromosome number, Cell capability of migration, tumorigenicity and cytogenetic features of the Cell Line were investigated. THY28 Cells were subcultured in vitro for more than 50 passages with a human karyotype. The modal number of its chromosomes was mainly from 67 to 85. The doubling time of THY28 Cells was 56 hours. The histopathological features of xenograft induced by THY28 Cells were consistent with the characteristics of thyroid cancer. The biological and molecular properties of THY28 Cells were not entirely consistent with those of other thyroid Carcinoma Cells such as SW579 and TT Cells, indicating biological differences between primary and metastatic thyroid Carcinoma Cell Lines. We have established a novel thyroid Carcinoma Cell Line derived from spinal metastasis, which will provide a useful model for biological or therapeutic studies of thyroid Carcinoma metastasis.
