    Cascara sagrada bark is extensively used in the treatment of occasional constipation. Despite its wide diffusion, Cascara sagrada dried extracts with good flow and compactability properties for tablet formulation by direct compression and simple methods for the determination of Cascara sagrada extract release from tablets are not available. To overcome this lack of information, this article is focused on the design of tablets based on a Cascara sagrada extract obtained by spray-drying and on the quantification of its active constituents using a novel and isocratic HPLC-UV technique, which was validated. Flow properties of the Cascara sagrada spraydried extract and of the physical mixtures of it with conventional excipients were evaluated. The different formulated powders were compacted and their properties (i.e., hardness, friability, disintegration and dissolution) were studied. The proposed tablet formulations, based on the Cascara sagrada spray-dried extract, reached the pharmacopoeial requirements for immediate release tablets. In particular, the formulas containing a superdisintegrant showed the best pharmaceutical performance. Nevertheless, the use of excipients to improve the powder flow and/or the tablets properties was not strictly necessary. Besides, the HPLC-UV method used to monitor the extract release from tablets was validated, proving to be linear within the concentration range adequate for therapeutic purposes and appropriate for routine analysis due to its simplicity, good sensitivity, accuracy and precision.

    Abstract The aim of this work was to study Rhamnus purshiana (Cascara sagrada) extract spray-drying in order to obtain powders with good rheological properties for direct compression (DC) and stability attributes. The experiments were carried out in a Buchi B-290 Mini Spray Dryer using colloidal silicon dioxide as carrier agent. A 2 5 − 1 fractional factorial statistical design was used to find adequate spray-drying operating conditions to produce Cascara sagrada powders with good flow, good compactability properties, high process yield, low moisture content, low hygroscopicity, and caking tendency. Morphology, size, structural and thermal properties of the particles were also evaluated. The operating variables studied were air inlet temperature, atomization air flow rate, pump feed rate, aspiration capacity, and feed solids concentration. The 2 5 − 1 factorial experimental design used was a good strategy to establish spray-drying operating conditions to produce Cascara sagrada extract powders with good properties for direct compression and high process yields. Colloidal silicon dioxide was an effective drying adjuvant. Its use in relatively high concentrations together with low atomization air flow rates improved powder flowability, increased the product's glass transition temperature, and decreased its moisture content and hygroscopicity.

    Among phytotherapic medicines, tablets of Cascara sagrada (Rhamnus purshiana) dried bark, usually used as laxative, are commercially widespread in our market. Taking into account natural origin and/or inappropriate procedures that may allow the occurrence of toxinogenic Aspergillus flavus group, a study on susceptibility to aflatoxin contamination and natural aflatoxin incidence was performed by TLC and HPLC methods. This survey allows one to conclude that bark of Cascara sagrada is a good substrate for the growth of A. parasiticus NRRL 2999 and for aflatoxins production. Natural anatoxins presence was detected on 2 from 9 raw material samples. One of them (irradiated sample) had only aflatoxin B1 (10 μg/kg) and the other (pasteurized) was positive for aflatoxin B1 (19 μg/kg); G1 (6 μg/kg) and B2 (1.46 μg/kg). Only one from 10 lots of tablets analyzed was positive for aflatoxin B1 (5.42 μg/kg) and B2 (0.32 μg/kg).Therefore, adequate quality control including an aflatoxins assay must be performed to guarantee the harmlessness of natural drugs.
