Capillary electrophoresis of isoquinoline alkaloids from Chelidonium majus L.

TomèAbstract:The determination of six different isoquinoline alkaloids from Chelidonium majus (leaves and roots) using capillary electrophoresis is described. Analyses were performed on fused silica capillaries with 66 mM phosphate buffer at pH 2.4 containing 40% methanol. The analytes were monitored by on-line ultraviolet (UV) detection to identify sanguinarine, chelerythrine, berberine and chelidonine. Both coptisine and DL-stylopine were isolated by semi-preparative high performance liquid chromatography (HPLC), and their structure assessed by UV and mass spectrometry. Capillary electrophoresis is confirmed as a useful complementary technique to HPLC.

Sanguinarine, chelidonine, chelerythrine, berberine and coptisine were detected in all parts of the plant. While the berberine content was rather stable, the other alkaloids undergo remarkable daily variations probably due to the combined effect of light and temperature.

TomèAbstract:Tissue callus cultures originating from Chelidonium majus L. seeds were established on Gamborg B5, Murashige-Skoog (MS), and Schenk-Hildebrandt (SH) media. In all experimental conditions, calli accumulated a complex pattern of isoquinoline derivatives. The highest values, both in growth and in sanguinarine production, were obtained using SH medium.

A new method for alkaloid extraction from Chelidonium majus L.

TomèAbstract:The increasing interest in Chelidonium majus L. and its various metabolites (i.e., the isoquinoline alkaloids, sanguinarine, chelerythrine, chelidonine and berberine) has given rise to a need for a rapid and reproducible method for the standardization and evaluation of fresh C. majus extracts. Both quaternary and tertiary alkaloids can be isolated from vegetable material by means of ion pair extraction and the recovery of these compounds was found to be consistently greater than 90%.

One new alkaloid from Chelidonium majus L.

Their chemical structures were elucidated on the basis of physicochemical and spectroscopic data. Among them, 6-acetonyldihydrochelerythrine (4), 6-acetonyldihydrosanguinarine (5), 6-ketenesanguinarine (6), demethylchelerythrine (7) and demethylsanguinarine (11) were isolated for the first time from this plant. Compound 6 was identified as a new compound. These compounds were screened for cytotoxicity against human non-small lung carcinoma (H1299), breast cancer (MCF-7) and liver cancer (SMMC-7721). In a series of cytotoxic tests, compounds 9 and 10 displayed potent cytotoxic activity against H1299, MCF-7 and SMMC-7721, with the IC50 values of 8.16–35.25 μg/mL.
