    : It is supposed that plant functional foods, rich in phytochemicals, may potentially have preventive effects in carcinogenesis. In this study, the anticancer effects of Cloves in the in vivo and in vitro mammary carcinoma model were assessed. Dried flower buds of Cloves (CLOs) were used at two concentrations of 0.1% and 1% through diet during 13 weeks after the application of chemocarcinogen. After autopsy, histopathological and immunohistochemical analyses of rat mammary carcinomas were performed. Moreover, in vitro evaluation using MCF-7 cells was carried out. Dietary administered CLO caused the dose-dependent decrease in tumour frequency by 47.5% and 58.5% when compared to control. Analysis of carcinoma cells in animals showed bcl-2, Ki67, VEGFA, CD24 and CD44 expression decrease and Bax, caspase-3 and ALDH1 expression increase after high-dose CLO administration. MDA levels were substantially decreased in rat carcinomas in both CLO groups. The evaluation of histone modifications revealed increase in lysine trimethylations and acetylations (H4K20me3, H4K16ac) in carcinomas after CLO administration. TIMP3 promoter methylation levels of CpG3, CpG4, CpG5 islands were altered in treated cancer cells. An increase in total RASSF1A promoter methylation (three CpG sites) in CLO 1 group was found. In vitro studies showed antiproliferative and pro-apoptotic effects of CLO extract in MCF-7 cells (analyses of cytotoxicity, Brdu, cell cycle, annexin V/PI, caspase-7, Bcl-2 and mitochondrial membrane potential). This study showed a significant anticancer effect of clove buds in the mammary carcinoma model in vivo and in vitro.

    Abstract The present study was designed to study the effect of storage duration on changes of antioxidant capacities and bioactive compounds in garlic clove by DPPH and FRAP assays and GC–MS analysis. Both DPPH and FRAP assays displayed that the antioxidant capacities in garlic clove extracts reached maximum values at 8 weeks. The levels of soluble sugar decreased during storage, and the contents of total polyphenols and organosulfur compounds reached maximum value at 6 and 8 weeks, respectively, and then decreased significantly. The GC–MS analysis results of ethyl acetate extract showed that among of the eighteen typical organosulfur compounds, ten compounds reached maximum levels at 8 weeks. The present study indicated that organosulfur compounds in garlic Cloves might play a key role in antioxidant capacities due to their positive relationship during storage. The results in this study would provide beneficial information for garlic processing industries and consumers to make full use of garlic during storage.

    : The cyanogenic polymorphism in Trifolium repens L. (white clover) has been used as the basis of a study of the genetic control of cyanogenesis. The Ac locus controls the presence of two cyanoglucosides in white clover. Biochemical characterization of cyanoglucoside biosynthesis in plants containing different Ac alleles has shown that this is a complex locus which affects more than one step in the pathway. A study of the in vivo synthesis and processing of the cyanogenic beta-glucosidase (linamarase) of white clover led to the isolation of cDNA clones for this enzyme. The cloning strategy and structure of the cDNA clones is described. Together with biochemical and genetic data, these clones have been used to characterize the Li locus which controls linamarase activity in white clover. It has been shown that 'null' alleles of the Li locus result in very reduced levels of transcription of homologous mRNA sequences. The use of these cDNA clones to investigate the genomic organization of cyanogenesis genes in both white clover and other cyanogenic species is described, and their use in structural analysis of the cyanogenic beta-glucosidase is discussed.

    The nucleotide sequence and derived amino acid sequence of two different β-glucosidase cDNA clones were determined. One clone (TRE104) was identified as the cyanogenic β-glucosidase by homology with the N-terminal and internal peptide amino acid sequence of the purified enzyme. The biological function of the other β-glycosidase (TRE361) is not known. Co-segregation of genomic restriction fragments uniquely identified by each cDNA clone shows that these two genes are linked in the white clover genome. Both TRE104 and TRE361 fragments co-segregate with cyanogenic β-glucosidase activity. Extensive homology was found between the white clover β-glucosidase sequences and a group of prokaryote and mammalian β-glycosidases. This group of sequences has no homology with a separate set of β-glucosidase genes isolated from fungi and the thermophilic bacterium Clostridium thermocellum .

    In attempting to increase the reliability of clover contribution in clove/ryegrass systems it is important to understand the roles of (1) specific traits of the clover genotype (2) climate and (3) their interactions in determining clover behaviour in swards. Overwintering and spring growth of white clover (cultivars AberHerald and Huia) grown in binary mixtures with perennial ryegrass were measured at 12 European sites ranging in latitude from Reykjavik, Iceland (64 degrees 30' N) to Pordenone, Italy (46 degrees 30' N). In the overwintering period, tiller density of the grass was assessed and detailed morphological and chemical measurements were made on the clover at each sampling time. During the growing season, the clover contribution to total available biomass was recorded, Detailed climatic data were available at all sites. The annual growth cycle of swards was divided into four functional periods (spring, summer, autumn and winter). Within each functional period community responses were modelled. The models incorporated independent biotic variables characterizing each community within each site at the start of the period and independent variables characterizing the climate at each site during the period. The models were linked dynamically by taking, as response variable(s) for a functional period. the independent biotic variable(s) of the succeeding period. In general, the modelling strategy was successful in producing a series of biologically meaningful linked models. Essential prerequisites for this were (a) the establishment of a well-devised common protocol prior to the experiment and (b) the extensive gradients of climatic and other variables obtained by using numerous sites, AberHerald generally performed as well as. or better than, Hula throughout the annual cycle across the range of climatic conditions encountered, and especially under low temperature conditions in winter and autumn. Clover leaf area index appeared to be a key variable in determining clover performance over winter and through the following growing season. Grass tiller density had a strong negative effect on clover content in spring but only at low temperatures, This emphasizes the importance of a high clover leaf area index in autumn as the main biotic factor related to spring clover content in milder conditions. The importance of climatic variables in the models is their use in explaining the reliability of the contribution of clover in clover,: ryegrass systems. Temperature was the primary climatic determinant of clover response in all periods, having a direct effect on clover content and leaf area index or mediating the effect of the associate species. Radiation strongly influenced Clover dynamics during winter and spring but not in the other periods, possibly because it was confounded with the effect of higher temperature. Precipitation was positively related to clover growth during spring and autumn and was related to tiller density in a complex manner during autumn and winter.
