Nifuroxazide, a potent STAT-3 inhibitor, exhibits diverse pharmacological properties. The present study aimed to elucidate a novel anti-colitis mechanism of nifuroxazide against the acetic acid-induced UC model. Methods Rats were grouped into control (received vehicle), UC (2 ml of 5% acetic acid by intrarectal infusion), UC plus sulfasalazine (100 mg/kg/day, P.O.), UC plus nifuroxazide (25 mg/kg/day, P.O.), and UC plus nifuroxazide (50 mg/kg/day, P.O.) and lasted for 6 days. Results The present study revealed that nifuroxazide significantly reduced UC measures, hematological changes, and histological alteration. In addition, treatment with nifuroxazide significantly down-regulated serum CRP as well as the Colonic expressions of MPO, IL-6, TNF-α, TLR-4, NF-κB-p65, JAK1, STAT-3, DKK1 in a dose-dependent manner. Besides, our results showed that the Colonic Wnt expression was up-regulated with nifuroxazide treatment. In a dose-dependent manner, nifuroxazide markedly alleviated acetic acid-induced cellular infiltration and improved Ulcer healing by increasing intestinal epithelial cell regeneration. Significance Our results collectively indicate that nifuroxazide is an effective anti-colitis agent through regulation of Colon inflammation and proliferation via modulation IL-6/STAT-3/Wnt signaling pathway.

Polski merkuriusz lekarski : organ Polskiego Towarzystwa Lekarskiego, 2008Co-Authors: Andrzej Jamry, Krzysztof Daniszewski, Marian Brocki, Szymon WcisloAbstract:UNLABELLED Nonspecific Colon Ulcer (NSCU) was described by Crulveilhier in 1832, but strict definition, occurrence rates and diagnostic and therapeutic guidelines have not been yet established. AIM to present a case of a rare disease of the Colon with discussion on diagnostic and therapeutic principles. CASE REPORT A 28-year-old female diagnosed for constipation and flatulence underwent endoscopy and a descending Colon lumen stenosis, measuring about 1.0 cm in length, with some mucosal erosions, was found. On the basis of the performed diagnostics, unspecific nature of the disease was determined and conservative treatment with endoscopic follow-up was initiated. In a follow-up examination, performed 2 months later, spontaneous Ulceration healing was revealed with a scar replacing the abovementioned lesion. DISCUSSION Clinical symptoms depend on disease dynamics and lesion location. Diagnosis is based on exclusion of known vascular, infective, unspecific inflammatory and traumatic causes. Therapy of uncomplicated causes is based on conservative treatment with endoscopic follow-up. CONCLUSIONS The disease does not cause any specific clinical symptoms and often mimics other abdominal diseases. Diagnosis is based on exclusion of known causes. Therapy of most uncomplicated cases comprises conservative therapy and endoscopic follow-up.

Free radicals increase the severity of the disease by decreasing the antioxidants. Silymarin is a flavonoid component that its extracted of Silybum marianum. Silymarin contains antioxidants and inhibits the inflammatory reactions. The aim of this research, is the investigation of antiinflammatory effect of silymarin in the treatment of Colon Ulcer induced acetic acid in mice Balb/C spicies.In this study, 32 mice are divided into 4 groups (n=8).These groups include:control group with no colitis report,the Sham group with colitis report that are not treated and the groups with colitis report that received 10 and 20 mg/kg B.W. of silymarin orally.The treatment groups received silymarin daily, for 1 week after having the disease. The drugs are fed to the mice per oral using gaster tubes. To induce colitis, 1 ml of acetic acid (%4) is injected into the rectum. In the one week after induction of colitis, the animals sacrificed and the damages of the Colon investigated in Morris and Murthy methods.The results of this study shown acetic acid causes severe inflammation and damages of the Colon. Also, silymarin is effective in improving of the inflammation and repair of the Colon Ulcer induced acetic acid. So, making use of silymarin can be considered as a suggestive way for the colitis treatment.
