    Abstract:

    CytoChrome C is one of the mitoChondrial proteins that is released into the Cytosol when the Cell is aCtivated by an apoptotiC stimulus. In the Cytosol, CytoChrome C engages the apoptotiC protease aCtivating faCtor-1 (APAF1), and forms the apoptosome, whiCh aCtivates Caspase-9. The release of CytoChrome C has been suggested to oCCur in two phases: mobilization from the mitoChondrial intermembrane spaCe and transloCation through the outer mitoChondrial membrane. The meChanisms of CytoChrome C release are Controversial. Whether the permeabilization of the outer or the inner membrane is responsible for the downstream events is one of the debated topiCs. Most evidenCe supports a model in whiCh the outer membrane is permeabilized without inner membrane events. The release of CytoChrome C and CytoChrome- C -mediated apoptosis are Controlled by multiple layers of regulation, with the most prominent players being members of the B-Cell lymphoma protein-2 (BCL2) family.     We performed label-free observation of moleCular dynamiCs in apoptotiC Cells by Raman miCrosCopy. DynamiC Changes in CytoChrome C distribution at the Raman band of 750 Cm-1 were observed after adding an apoptosis induCer to the Cells. The Comparison of mitoChondria fluoresCenCe images and Raman images of CytoChrome C Confirmed that Changes in CytoChrome C distribution Can be distinguished as release of CytoChrome C from mitoChondria. Our observation also revealed that the redox state of CytoChrome C was maintained during the release from the mitoChondria. Monitoring mitoChondrial membrane potential with JC-1 dye Confirmed that the observed CytoChrome C release was assoCiated with apoptosis.
