V wurde durch das Acetat VI und das Formiat VII charakterisiert. VI und VII werden durch 1-proz. methanolische HCl bei 20° vollstandig hydrolisiert, wahrend Digitoxigenin-acetat (II) dabei unverandert bleibt. 3-Epi-Digitoxigenin (V) zeigte an der Katze keine digitalisartige Wirkung.

TammAbstract:Digitoxigenin (I) wurde auf mikrobiologischem Wege (mit Fusarium Zini (Bolley)) durch Einfuhrung einer Hydroxylgruppe in die 12β-Stellung in Digoxigenin (VII) ubergefuhrt. Der gleiche Mikroorganismus vermochte 3-De-hydro-Digitoxigenin (X) in 3-epi-Digitoxigenin (XII), 3-Dehydro-digoxigenin (XI) und 3-epi-Digoxigenin (XIV) umzuwandeln. XI und XIV sind neue Stoffe. Zum Konstitutionsbeweis wurde XI durch katalytische Oxydation mit O2, und Pt aus Digoxigenin (VII) hergestellt. NaBH4,-Reduktion von XI ergab 3-epi-Digoxigenin (XIV).

The α-l-rhamnose and α-l-amicetose digitoxin monosaccharide analogues displayed stronger apoptosis inducing activity and cytotoxicity against nonsmall cell human lung cancer cells (NCI-H460) than its d-diastereomeric isomers in a sugar-chain length dependent manner.

a stereoselective synthesis of digitoxin and digitoxigen mono and bisdigitoxoside from Digitoxigenin via a palladium catalyzed glycosylation
Organic Letters, 2006Co-Authors: Maoquan Zhou, George A OdohertyAbstract:A convergent and stereocontrolled route to trisaccharide natural product digitoxin has been developed. The route is amenable to the preparation of both the digitoxigen mono- and bisdigitoxoside. This route featured the iterative application of the palladium-catalyzed glycosylation reaction, reductive 1,3-transposition, diastereoselective dihydroxylation, and regioselective protection. The natural product digitoxin was fashioned in 15 steps starting from Digitoxigenin 2 and pyranone 8a or 18 steps from achiral acylfuran.
