Electroacupuncture ameliorates cognitive impairment through inhibition of nf κb mediated neuronal cell apoptosis in cerebral ischemia reperfusion injured rats

Nuclear factor‑κB (NF-κB)-mediated neuronal cell apoptosis is involved in the development of post-stroke cognitive impairment; therefore, it has become a promising target for the treatment of impaired cognition. Acupuncture at the Baihui (DU20) and Shenting (DU24) acupoints is commonly used in China to clinically treat post‑stroke cognitive impairment; however, the precise mechanism of its action is largely unknown. In the present study, we evaluated the therapeutic efficacy of Electroacupuncture against post-stroke cognitive impairment and investigated the underlying molecular mechanisms using a rat model of focal cerebral ischemia-reperfusion (I/R) injury. Electroacupuncture at Baihui and Shenting was identified to significantly ameliorate neurological deficits and reduce cerebral infarct volume. Additionally, Electroacupuncture improved learning and memory ability in cerebral I/R injured rats, demonstrating its therapeutic efficacy against post-stroke cognitive impairment. Furthermore, Electroacupuncture significantly suppressed the I/R-induced activation of NF-κB signaling in ischemic cerebral tissues. The inhibitory effect of Electroacupuncture on NF-κB activation led to the inhibition of cerebral cell apoptosis. Finally, Electroacupuncture markedly downregulated the expression of pro-apoptotic Bax and Fas, two critical downstream target genes of the NF-κB pathway. Collectively, our findings suggest that inhibition of NF-κB‑mediated neuronal cell apoptosis may be one mechanism via which Electroacupuncture at Baihui and Shenting exerts a therapeutic effect on post-stroke cognitive impairment.

Electroacupuncture at the quchi and zusanli acupoints exerts neuroprotective role in cerebral ischemia reperfusion injured rats via activation of the pi3k akt pathway

Acupuncture has long been used in China to clinically treat stroke. However, the precise mechanism of its neuroprotective activities remains largely unknown. Using a focal cerebral I/R injured rat model, in the present study we evaluated the in vivo therapeutic efficacy of Electroacupuncture and investigated the underlying molecular mechanisms. We found that Electroacupuncture at Quchi (LI11) and Zusanli (ST36) acupoints on the contralateral paralyzed limb significantly improved neurological deficits and cerebral infarction. In addition, Electroacupuncture profoundly activated PI3K/Akt signaling in ischemic cerebral tissues. Consequently, the upregulatory effect of Electroacupuncture on PI3K/Akt activation resulted in the inhibition of cerebral cell apoptosis. Moreover, Electroacupuncture increased the serum secretion levels of the PI3K activators BDNF and GDNF, as well as upregulated the anti-apoptotic Bcl-2/Bax ratio in ischemic cerebrum. Our data suggest that Electroacupuncture at Quchi and Zusanli acupoints exerts neuroprotective function in ischemic stroke via activation of the PI3K/Akt pathway.

Electroacupuncture at The Zusanli point could enhance microcirculation and blood perfusion in random skin flaps. OBJECTIVE: To determine whether Electroacupuncture at The Zusanli point can improve the survival of random skin flaps in a rat model. MATERIALS AND METHODS: Thirty-six male Sprague Dawley rats were randomly divided into 3 groups: control group (no Electroacupuncture), Group A (Electroacupuncture at a nonacupoint near The Zusanli point), and Group B (Electroacupuncture at The Zusanli point). McFarlane flaps were established. On postoperative Day 2, malondialdehyde (MDA) and superoxide dismutase were detected. The flap survival rate was evaluated, inflammation was examined in hematoxylin and eosin-stained slices, and the expression of vascular endothelial growth factor (VEGF) was measured immunohistochemically on Day 7. RESULTS: The mean survival area of the flaps in Group B was significantly larger than that in the control group and Group A. Superoxide dismutase activity and VEGF expression level were significantly higher in Group B than those in the control group and Group A, whereas MDA and inflammation levels in Group B were significantly lower than those in the other 2 groups. CONCLUSION: Electroacupuncture at The Zusanli point can effectively improve the random flap survival.

PLOS ONE, 2014Co-Authors: Jia Shi, Lirong Gong, Shuan Dong, Yuan Zhang, Xinshun CaoAbstract:NF-E2 related factor 2 (Nrf2) is a major transcription factor and acts as a key regulator of antioxidant genes to exogenous stimulations. The aim of current study was to determine whether Nrf2/ARE pathway is involved in the protective effect of Electroacupuncture on the injured lung in a rabbit model of endotoxic shock. A dose of lipopolysaccharide (LPS) 5 mg/kg was administered intravenously to replicate the model of acute lung injury induced by endotoxic shock. Electroacupuncture pretreatment was handled bilaterally at Zusanli and Feishu acupoints for five consecutive days while sham Electroacupuncture punctured at non-acupoints. Fourty anesthetized New England male rabbits were randomized into normal control group (group C), LPS group (group L), Electroacupuncture + LPS group (group EL) and sham Electroacupuncture + LPS (group SEL). At 6 h after LPS administration, the animals were sacrificed and the blood samples were collected for biochemical measurements. The lungs were removed for calculation of wet-to-dry weight ratios (W/D), histopathologic examination, determination of heme oxygenase (HO)-1 protein and mRNA, Nrf2 total and nucleoprotein, as well as Nrf2 mRNA expression, and evaluation of the intracellular distribution of Nrf2 nucleoprotein. LPS caused extensive morphologic lung damage, which was lessened by Electroacupuncture treatment. Besides, lung W/D ratios were significantly decreased, the level of malondialdehyde was inhibited, plasma levels of TNF-α and interleukin-6 were decreased, while the activities of superoxide dismutase, glutathione peroxidase and catalase were enhanced in the electroacupucnture treated animals. In addition, Electroacupuncture stimulation distinctly increased the expressions of HO-1 and Nrf2 protein including Nrf2 total protein and nucleoprotein as well as mRNA in lung tissue, while these effects were blunted in the sham Electroacupuncture group. We concluded that Electroacupuncture treatment at ST36 and BL13 effectively attenuates lung injury in a rabbit model of endotoxic shock through activation of Nrf2/ARE pathway and following up-regulation of HO-1 expression.

It is proposed that compound Electroacupuncture waveform is an inevitable trend for Electroacupuncture technique development. Based on the clinic and experiment needs, three different waveforms are taken as the model of compound Electroacupuncture waveforms. Controlled by the microcomputer technique, the compound Electroacupuncture waveforms are more multiple and precise, which will expand the range of application of Electroacupuncture.

METHODS: Analyze and study on the relevant literature about the application of Electroacupuncture in the past 5 years and analyze the parameters of Electroacupuncture in the literature in detail. RESULTS: Most of literature paid greater attention to the selection of parameters of Electroacupuncture, but different Electroacupuncture apparatuses were used with different Electroacupuncture parameters. CONCLUSION: The selection of the Electroacupuncture apparatus and Electroacupuncture parameters does not have an unified standard, influencing comparability and reference of experimental and clinical results.

Analysis and Study about Stimulating Parameters of Electroacupuncture on the Clustering Technique

Method: On the base of the study on parameters quantification of Electroacupuncture and application of the Electroacupuncture on the clinical practice, propose the means which analysis and study the large numbers of parameters of Electroacupuncture by the clustering technique, and consult the specialists' explain about the result of clustering. Result: It is a new idea to use the clustering technique to analyze the parameter of Electroacupuncture, and it can remedy the deficiency of the current study on parameters of Electroacupuncture in a certain extent. Conclusion: The result of clustering can provide the theory foundation for the different curative effect in the kinds of parameters of Electroacupuncture and provide some reference for choosing the optimal parameters' combination in clinic work.

Analgesic effects of Electroacupuncture of different pulse waveforms in the rat of adjuvant arthritis

METHODS Forty Wistar rats were randomly divided into 5 groups. The Electroacupuncture groups of AA were treated with different pulse waveforms of Electroacupuncture, respectively. Pain threshold, swelling degrees of joint, and contents of 5-HT, beta-EP and LEK in local tissues of inflammation were observed. RESULTS After the treatment, pain thresholds of the Electroacupuncture groups with different pulse waveforms were higher than that of the model group (P < 0.05, P < 0.01); the local swelling degrees of inflammation in the Electroacupuncture group were higher than that in the normal group (P < 0. 01), but was lower than that in the model group (P < 0.05, P < 0. 01); the contents of 5-HT in local tissues of inflammation in the Electroacupuncture groups were lower than that in the model group (P < 0.05, P < 0.01); in comparison with the model group, the Electroacupuncture of sound-electric wave increased the contents of beta-EP and LEK (P < 0.01, P < 0.05) and the Electroacupuncture of intermittent wave increased the content of beta-EP in local tissues of inflammation (P < 0.05). CONCLUSION Sound-electric wave is the best pulse waveform of Electroacupuncture for treatment of adjuvant arthritis.
