FURTHER EVIDENCE OF AN EXTENSIVE CROSS‐REACTIVITY BETWEEN THE THREE MAIN PARTS OF THE BOVINE Encephalitogenic Protein IN THE LYMPHOID CELL TRANSFORMATION TEST AND THE MACROPHAGE MIGRATION INHIBITION TEST
Acta Pathologica Microbiologica Scandinavica Section C Immunology, 2009Co-Authors: Håkan Bergstrand, Ben KallenAbstract:The results presented indicate that the three main peptide parts of bovine Encephalitogenic Protein (BEP) cross-react when examined by the macrophage migration inhibition test in guinea pigs and the lymph node cell transformation test in rabbits: 1. Guinea pigs were immunized with peptide 43—88 of BEP and the capillary migration of peritoneal cells from these animals was examined in the presence of the intact Protein or various peptide fragments derived from the latter. The complete Protein and peptides 1–42 and 92–169 were all capable of inhibiting cell migration, but none of these antigens were as efficient as the immunizing agent (peptide 43–88). 2. Lymph node cells from rabbits sensitized with peptide 1–42, 43–88, or 92—169 were examined using the lymphoid cell transformation test. Cells from animals immunized with any one region always showed a substantial degree of transformation also when stimulated by peptides covering the other two regions of the Protein, but the immunizing peptide consistently showed a transforming effect better than that of the other two peptides. 3. Control experiments showed that contamination of the immunizing peptide with other parts of the Protein could not explain the observed cross-reactivity between the three main regions of BEP.

Effect of basic Encephalitogenic Protein and some peptides derived from it on the migration in agarose gel of leukocytes from patients with multiple sclerosis, other neurological diseases, or carcinoma.
Acta neurologica Scandinavica, 2009Co-Authors: Håkan Bergstrand, Bengt Källián, Olle NilssonAbstract:Peripheral blood leukocytes from patients with multiple sclerosis (MS) or other diseases were studied using Clausen's direct agarose gel migration method, with bovine Encephalitogenic Protein (BEP) and four peptides derived from it as antigens. Cells from healthy individuals show a slight and dose-dependent inhibition with BEP. Of 35 cell samples from 24 MS patients, 16 showed a significantly stronger reaction, manifested either as inhibition or stimulation of migration, than that seen with cells from healthy individuals. Of 29 samples from 29 non-MS patients, 6 showed such a reaction. In most instances, the reactivity to BEP could be localized to one or more of the peptides studied. There was no correlation established between peptide activity in the test and disease. A reactivity to the region containing the single tryptophan residue of the molecule (residue 116) could be demonstrated with cells from four patients: three had MS, the fourth had amyotrophic lateral sclerosis. One patient with active MS was followed for approx. two months, and definite changes in reactivity to BEP and the peptides were seen.

THE RECONSTRUCTION OF A FRAGMENT OF PRIMORDIAL GENETIC INFORMATION FROM MODERN ProteinS: PGI‐F1 (AHAP)
International journal of peptide and protein research, 2009Co-Authors: Klausdieter BauerAbstract:In a continuation of earlier studies the connection between a reconstructed ancestral histone IV peptide and various sequences from non-histone Proteins was investigated. This peptide, AHAP, was found to be related to partial peptides from the human Encephalitogenic Protein, immunoglobulin L-chains, fibrinopeptide A, glyceraldehyde-3-phosphate-dehydrogenase, TMV coat Protein and several other Proteins and Protein families; for comparison, nodal sequences were employed wherever possible. The widespread occurrence of genetic information expressed in AHAP caused us to rename the peptide PGI-FI (primordial genetic information-fragment 1) since this peptide is apparently part of a very primitive and ancient genetic information, and may be called "protogene".
