    ABSTRACTThe hagfishes of the genus Eptatretus (Myxinidae) from southern Africa are known from three poorly studied species: Eptatretus hexatrema, a common species from Namibia and South Africa; Eptatretus profundus, known only from the holotype collected off Cape Point (South Africa); and Eptatretus octatrema, known from two syntypes from the Agulhas Bank (South Africa). Taxonomic, morphological and distributional information about these three species are reviewed and updated based on the examination of additional specimens collected in South African waters. Eptatretus hexatrema differs from all congeners by having six pairs (rarely seven) of gill apertures arranged in a straight line, 3/2 multicusp pattern of teeth, total cusps 44–49, trunk pores 53–60, total pores 93–107, preventral length 45.1–57.4% TL, tail length 11.6–14.3% TL, tail depth 5.7–8.1% TL, and two bilaterally symmetrical nasal-sinus papillae.     A new species of seven-gilled hagfish Eptatretus astrolabium (Myxinidae) is described from a 400 mm total length female trapped 1 km east from Planet Rock, Astrolabe Bay, Papua New Guinea, at c. 500 m depth. This is the first hagfish species reported from the waters around New Guinea. It can be distinguished from other hagfishes by a combination of characters including seven pairs of gill apertures, three-cusp multicusps on the anterior and posterior rows of cusps, 10 posterior unicusps, 52 total cusps, 18-19 prebranchial pores, five branchial pores, 48-49 trunk pores, 83-84 total pores and no nasal-sinus papillae.

    Abstract:

    This paper revises and updates taxonomic and distributional information about hagfishes (Myxinidae) from Australia. It covers five species of the genus Eptatretus: Eptatretus cirrhatus known from eastern Australia and also distributed around New Zealand, Eptatretus longipinnis endemic to South Australia, Eptatretus strahani originally described from the Philippines and reported here as a new record from Western Australia and two new species described herein as Eptatretus alastairi and Eptatretus gomoni, both from Western Australia. Eptatretus alastairi is distinguished from all congeners by the unique combination of the following characters: six pairs of gill pouches; three-cusp multicusps on the anterior and posterior rows of cusps; anterior unicusps 9-12; posterior unicusps 8-11; total cusps 48-56; prebranchial pores 13-16; branchial pores 5-6; trunk pores 50-55; tail pores 11-13; total pores 83-88; two bilaterally symmetrical nasal-sinus papillae in the dorsal surface of the nasal sinus. Eptatretus gomoni is distinguished from all congeners by the unique combination of the following characters: eight pairs of gill pouches; three-cusp multicusps on the anterior and two-cusp multicusps on the posterior row of cusps; anterior unicusps 10-11; posterior unicusps 9-10; total cusps 50; prebranchial pores 12-13; branchial pores 7-8; trunk pores 57-58; tail pores 14-15; total pores 91-93; no nasal-sinus papillae. An identification key for the Australian species of Eptatretus is also provided.

    The fine structure of spermatid differentiation in a primitive vertebrate, the hagfish, whose spermatozoa bear acrosomes, was investigated. In early round spermatids, the acrosomal vesicles were spherical and located in a shallow nuclear indentation, flanked by the plasma and the nuclear membranes. The vesicle underwent a transition through lens-shaped and cap-shaped stages until it attained the shape of a bell in mature spermatozoa. Electron-dense acrosomal material that appeared as deposits in three portions of the vesicle finally joined in the center region at a late stage. Condensation of chromatin occurred in the anterior region of the nucleus. During transformation of the spermatids, many regularly spaced microtubules appeared beneath the plasma membrane except in the anteriormost region of the cell.     Spermatozoa of the hagfishes Eptatretus burgeri and Eptatretus stouti, caught in the sea near Japan and North America, respectively, were found to undergo the acrosome reaction, which resulted in the formation of an acrosomal process with a filamentous core. The acrosomal region of spermatozoa of E. stouti exhibited immunofluorescent labeling using an actin antibody. The midpiece also labeled with the antibody. The acrosomal region showed a similar labeling pattern when sperm were probed with tetramethylrhodamine isothyocyanate (TRITC)-phalloidin; the midpiece did not label. Following induction of the acrosome reaction with the calcium (Ca2+) ionophore ionomycin, TRITC-phalloidin labeling was more intense in the acrosomal region, suggesting that the polymerization of actin occurs during formation of the acrosomal process, as seen in many invertebrates. The potential for sperm to undergo acrosomal exocytosis was already acquired by late spermatids. During acrosomal exocytosis, the outer acrosomal membrane and the overlying plasma membrane disappeared and were replaced by an array of vesicles; these resembled an early stage of the acrosome reaction in spermatozoa of higher vertebrates in which no formation of an acrosomal process occurs. It is phylogenetically interesting that such phenomena occur in spermatozoa of hagfish, a primitive vertebrate positioning between invertebrates and high vertebrates.

    Abstract:

    Fertilization of the hagfish or myxiniformes, a member of the most primitive vertebrate group and an animal of phylogenic interest, is unknown. Here, induction of an acrosome reaction for spermatozoa in the hagfish, Eptatretus burgeri, was successfully achieved by treatment of mature spermatozoa with ionomycin and excess Ca2+. The spermatozoon produced an acrosomal process that elongated from the apex of the long sperm head. The reaction bears resemblance to that of invertebrate spermatozoa rather than that of vertebrate spermatozoa. The result provides insights into the phylogenetical changes that have occurred in this sperm reaction.

    The whitish hagfish, Eptatretus albiderma sp. nov., was collected from the northeast Sea of Vietnam (Da Nang). This new species possesses six pairs of gill apertures, three multicusps, 18–21 prebranchial slime pores, absent or one branchial slime pore, and whitish to pinkish body color. Eptatretus albiderma sp. nov. can be readily distinguished from Eptatretus atami (Dean, 1904) and Eptatretus sheni (Kuo, Huang & Mok, 1994) by the numbers of total cusps (41–43 vs. 47–52 in E. atami vs. 48–53 in E. sheni), body color (whitish vs. darkish in E. atami vs. dark brown dorsally, lighter ventrally in E. sheni), eyespots (absent or faint vs. prominent in E. sheni), and total slime pores (74–78 vs. 71–78 in E. atami vs. 62–74 in E. sheni). The new species is clearly distinguished from E. atami by 0.026 pairwise genetic distance (d) of phylogeny based on mtDNA COI. We herein offer the key to 17 species of the genus Eptatretus Cloquet, 1819 in East Asia.

    Four specimens of the five-gilled white mid-dorsal line hagfish, Eptatretus wandoensissp. nov. were recently collected from the southwestern Sea of Korea (Wando). This new species has five pairs of gill apertures, 14-18 prebranchial slime pores, 4 branchial slime pores, a dark brown back with a white mid-dorsal line and a white belly. These hagfish are similar to Eptatretus burgeri and Eptatretus minor in having a white mid-dorsal line, but can be readily distinguished by the numbers of gill apertures (5 vs. 6-7), gill pouches (5 vs. 6), and prebranchial slime pores (14-18 vs. > 18), as well as the body color (dark brown back vs. gray or brown pale). In terms of genetic differences, Eptatretus wandoensis could be clearly distinguished from E. burgeri (0.9% in 16S rRNA and 8.5% in cytochrome c oxidase subunit I sequences) and E. minor (4.5% and 13.9%).

    Abstract Eptatretus walkeri was originally described based on 25 specimens from several coasts of Japan by McMillan and Wisner in 2004. We report the first collection of this species since its first description, based on 80 specimens caught at several localities along the southern Korean coast. Eptatretus walkeri is distinguished from its congeners by six gill pouches and gill apertures arranged in a closely spaced, linear pattern or a slightly crowded, irregular pattern; absence of branchial slime pores; 3/2 fused cusps; 36–44 total cusps; 68–79 total slime pores; faint or absent eyespots; poorly developed ventral fin-fold; and gill aperture surrounded by a pale margin. Based on molecular data (mtDNA), Eptatretus walkeri differs from other congeners by 0.9–4.5% in 16S ribosomal RNA gene, and 5.2–12.6% in cytochrome c oxidase subunit I. Because Eptatretus walkeri is poorly known, we redescribe the species based on additional voucher specimens, report its range expansion, and discuss its past taxonomic confusion with E. atami (Dean 1904)

    Este trabalho contem informações extensivamente revisadas sobre a taxonomia e distribuição dos peixes-bruxa. Ele inclui todas as espécies nominais da família Myxinidae, a qual compreende 2 subfamílias, 6 gêneros e 69 espécies. Informações sobre cada espécies incluem: nomes válidos e descrições originais; sinônimos; material examinado; características diagnósticas; tamanho máximo; distribuição e habitat; interesse pesqueiro; observações (quando necessário); e nomes vernaculares. Pela primeira vez são apresentadas chaves de identificação e mapas de distribuição para todos os gêneros e espécies, observando-se ampliações na distribuição de muitas delas. As subfamília Paramyxininae Berg, 1947 e Quadratinae Wisner, 1999 são consideradas sinônimos júnior de Eptatretinae Bonaparte, 1850. O gênero Quadratus Wisner, 1999 é considerado sinônimo júnior de Paramyxine Dean, 1904. Uma espécie de Eptatretus e duas de Myxine são sinonimizadas. Duas novas espécies são descritas: Eptatretus lakeside Mincarone & McCosker, 2004, do Arquipélago de Galápagos, e Eptatretus goliath Mincarone & Stewart, 2006, da Nova Zelândia. A inclinação da curva acumulativa de diversidade temporal indica que o número de espécies de Myxinidae ainda está longe de ser conhecido. Cruzeiros de pesquisa direcionados a regiões pouco exploradas e o uso de novas tecnologias de coleta poderão revelar espécies ainda não descritas

The phylogenetic analysis using the sequence of mitochondrial gene cytochrome oxidase C (CO1) suggested the Eptatretus sp. collected from India is distinct from all other species where sequence data is available and P ID (Liberal) for Eptatretus sp. collected from India is 0.96, indicating a high probability that an unknown member of the putative species would be correctly identified as a member once more specimens are collected

    A recent phylogenetic analysis of the Myxinidae based on the 16S rRNA gene resulted in synonymization of Paramyxine with Eptatretus. This created homonymy of Paramyxine fernholmi with Eptatretus fernholmi and Paramyxine wisneri with Eptatretus wisneri. In order to resolve this nomenclatural dilemma, we made a more extensive phylogenetic assessment of the Myxinidae and examined the nomenclature of the family. We used 75 sequences (37 of which new for this study) of a 561 bp fragment of the 16S rRNA gene, representing 33 species, and 72 sequences (37 of which new for this study) of a 687 bp fragment of the cytochrome c oxidase subunit I (COI) gene, representing 23 species, to reconstruct the phylogeny of Myxinidae. The monophyly of the subfamily Myxininae, traditionally characterized by having a single pair of external gill openings, was rejected (0.50 Bayesian posterior probability) by the 16S analysis, but supported by the COI and combined COI+16S analyses (0.99 and 0.81 Bpp, respectively). The monophyly of the subfamily Eptatretinae, characterized by having several pairs of external gill openings, was not supported by the 16S analysis and rejected by the COI and combined COI+16S analysis due to the placement of Eptatretus lopheliae as the earliest branch of Myxinidae (0.71 and 0.57 Bpp, respectively). Eptatretus lopheliae and Eptatretus rubicundus formed a monophyletic group and were allocated to a new genus, Rubicundus, characterized by the presence of an elongated tubular nostril and reddish coloration. A new monotypic subfamily, Rubicundinae, was proposed for Rubicundus. The synonymy of the genera Paramyxine and Quadratus with Eptatretus was confirmed. E. fernholmi is renamed Eptatretus luzonicus. Eptatretus wisneri was renamed Eptatretus bobwisneri. Petromyzon cirrhatus Forster, 1801, Homea banksii Fleming, 1822, and Bdellostoma forsteri M€ 1836 are synonyms, but no type specimens are known to exist. Petromyzon cirrhatus was designated as type species of Eptatretus, conserving present usage. Gastrobranchus dombeyi Shaw, 1804 has priority over other names for Chilean myxinids. Bdellostoma stoutii was designated as type species of Polistotrema Gill. The validity of the Western Atlantic Myxine limosa as distinct from the Eastern Atlantic Myxine glutinosa was confirmed.
