    Abstract:

    The implementation Requirements are multiplex in Equipment Requirement demonstration, which leads to the flexible flows of Equipment Requirement demonstration. And the flexible flows have to achieve the corresponding contents. So the method designing the flexible flows of Equipment Requirement demonstration is proposed. Firstly, according to the characteristic of Equipment Requirement demonstration, the task breakdown structure and the activity meta-model of Equipment Requirement demonstration are designed. And the flow type and the method constructing the flows are proposed. And then the flows of Equipment Requirement demonstration can be built. Secondly, aiming at the design Requirement of the flexible flows, the flows of Equipment Requirement demonstration are differentiated into skeleton flow, operation flow and procedure flow. And the methods selecting and designing flexible activities are studied. The proposed methods in this paper are helpful to design flexible flows of Equipment Requirement demonstration.

    Aiming at the engineering-oriented application Requirements of the Equipment Requirement demonstration, the demonstration process driven by models is regarded as the essential goal. Firstly, the activities and the modeling Requirements of the Equipment Requirement demonstration are analyzed in detail. Thereafter, the model system of the Equipment Requirement demonstration is built. Focusing on the design of the model base, the model-driven model development process and the design pattern of the models based on MVC are put forward and discussed emphatically. The VV&A mechanism is designed to improve the quality of the models. According to the management Requirements of the model base, the model management technology based on ontology is put forward, and then the run mechanism of the model base is studied. Throughout all these designs, the models simulating the Equipment Requirement demonstration activities can be understood and reused better, and can satisfy the elementary Requirements building the toolkits based on these models. Simultaneity, the Equipment Requirement demonstration activities can be regulated and formalized with the toolkits.

    Firstly, in order to overcome the shortcoming of using only AD or TRIZ solely, and solve the problems currently existed in weapon Equipment Requirement demonstration, the paper construct the method system of weapon Equipment Requirement demonstration combining QFD, AD, TRIZ, FA. Then, we construct a CAI model frame of weapon Equipment Requirement demonstration, which include Requirement decomposed model, Requirement mapping model and Requirement plan optimization model. Finally, we construct the computer aided innovation model of weapon Equipment Requirement demonstration, and developed CAI software of Equipment Requirement demonstration.

  • Reconfigurable microfluidic dilution for high-throughput quantitative assays

    This paper reports a reconfigurable microfluidic dilution device for high-throughput quantitative assays, which can easily produce discrete logarithmic/binary concentration profiles ranging from 1 to 100-fold dilution in parallel from a fixed sample volume (e.g., 10 μL) without any assistance of continuous fluidic pump or robotic automation. The integrated dilution generation chip consists of switchable distribution and collection channels, metering reservoirs, reaction chambers, and pressure-activatable Laplace valves. Following the sequential loading of a sample, a diluent, and a detection reagent into their individual metering chambers, the top microfluidic layer can be reconfigured to collect the metered chemicals into the reaction chambers in parallel, where detection will be conducted. To facilitate mixing and reaction in the microchambers, two acoustic microstreaming actuation mechanisms have been investigated for easy integrability and accessibility. Furthermore, the microfluidic dilution generator has been characterized by both colorimetric and fluorescent means. A further demonstration of the generic usage of the quantitative dilution chip has utilized the commonly available bicinchoninic acid (BCA) assay to analyse the protein concentrations of human tissue extracts. In brief, the microfluidic dilution generator offers a high-throughput high-efficiency quantitative analytical alternative to conventional quantitative assay platforms, by simple manipulation of a minute amount of chemicals in a compact microfluidic device with minimal Equipment Requirement, which can serve as a facile tool for biochemical and biological analyses in regular laboratories, point-of-care settings and low-resource environments.

    There are two sections in support quantitative parameter determination for Equipment Requirement argumentation, which contains index selection and determination method. The paper analyzes the selections of quantitative parameter firstly, and then makes sure the needs and common process. For the characteristics of representative parameters, the paper builds up a support object model and system model for the research of support quantitative parameters used Object Oriented method. And last, the article gives an example of armored Equipment, and combines the theory of Discrete-Event simulation. The simulation experimentation gives a result of support parameters according to initialization design index, which will provide a good suggestion for the design of weapon Equipments.

    Loop-mediated isothermal amplification (LAMP) is a diagnostic method for amplification of DNA with rapid and minimal Equipment Requirement. In the present study, we applied the LAMP assay for rapid detection of cow components adulteration in buffalo milk/meat samples. The test can be completed within around 1 h 40 min starting from DNA extraction and can be performed in water bath without Requirement of thermocycler. The cow DNA in buffalo samples were identified in the developed LAMP assay by either visualizing with SYBR Green I/HNB dyes or observing the typical ladder pattern on gel electrophoresis. The test can detect up to 5 % level of cow milk/meat mixed in buffalo counterparts. Due to the simplicity and specificity, the developed LAMP test can be easily adapted in any laboratory for rapid detection of cow species identification in livestock by products.

    Fast and economical means of assaying SNP’s are important in diagnostic assays, especially when a large number of animals have to be screened for a genetic disease. This study was aimed at the development of a fast and economical screening assay for bovine leukocyte adhesion deficiency (BLAD) which is an important genetic disease of cattle industry. Four primers were designed where the outer primers amplify a 354 bp amplicon of CD18 gene carrying the polymorphism responsible for BLAD. The specifically designed inner primers in conjunction with the modified reaction mixture and cyclic conditions ensured amplification of either of wild or mutated alleles. Together with outer primers, the inner primers generated typical banding pattern in agarose gel which discriminated the normal animal against the carrier. We successfully used this protocol in 200 bulls for genotyping the BLAD allele which confirmed by sequencing, showing a cent percentage concordance. With the developed assay the need for restriction digestion or use of costly Equipment viz. real time PCR was eliminated. This genotyping assay ensured fast and economical genotyping and could be adopted in every laboratory with a minimum Equipment Requirement of thermocycler and gel documentation system.
