Method. We evaluated the preventive effect of ETFA by analysing the data on 821 cycles at high risk for SOHSS performed during the years 1987–99. In all cycles the high risk for SOHSS was defined by the estradiol values > 3000 pg/ml and more than 8 follicles > 14 mm plus intermediate and small follicles in each ovary on ultrasound. Results In the group of :729 female infertility cycles at high risk for SOHSS with preventive ETFA there were no cases of SOHSS. In the control group of 92 IVF/ET female infertility without ETFA there were 12 cases of SOHSS (P < 0.0001). No difference in live birth rate (17% vs. 16%) was noted. The results prove the preventive effect of ETFA. It is based on early timed withdrawal of paracine substances which are present in the follicular fluid. Conclusion The vaginal ultrasound guided ETFA is another successful option to decrease the incidence of severe OHSS in assisted reproduction.

Catabolite Repression of the Bacillus subtilis FadR Regulon, Which Is Involved in Fatty Acid Catabolism

Since the lcfA-fadRB-etfBA, lcfB, and fadNAE operons, whose gene products directly participate in the β-oxidation cycle, had been found to be probably catabolite repressed upon genome-wide transcript analysis, we performed Northern blotting, which indicated that they are clearly under CcpA-dependent catabolite repression. So, we searched for catabolite-responsive elements (cre's) to which the complex of CcpA and P-Ser-HPr binds to exert catabolite repression by means of a web-based cis-element search in the B. subtilis genome using known cre sequences, which revealed the respective candidate cre sequences in the lcfA, lcfB, and fadN genes. DNA footprinting indicated that the complex actually interacted with these cre's in vitro. Deletion analysis of each cre using the lacZ fusions with the respective promoter regions of the three operons with and without it, indicated that these cre's are involved in the CcpA-dependent catabolite repression of the operons in vivo.

Organization and function of the YsiA regulon of Bacillus subtilis involved in fatty acid degradation.

Northern and primer extension analyses indicated that this regulon comprises five operons, i.e. lcfA-ysiA-B-etfB-A, ykuF-G, yhfL, yusM-L-K-J, and ywjF-acdA-rpoE. YusJ and AcdA, YsiB and YusL, and YusK presumably encode acyl-CoA dehydrogenases, 3-hydroxyl-CoA dehydrogenase/enoyl-CoA hydratase complexes, and acetyl-CoA C-acyltransferase, respectively, which are directly involved in the fatty acid β-oxidation cycle. In addition, LcfA and YhfL are likely to encode long chain acyl-CoA ligases. On gel retardation and footprinting analyses involving the purified YsiA protein, we identified cis-sequences for YsiA binding (YsiA boxes) in the promoter regions upstream of ysiA, ykuF, yusL, yhfL, and ywjF, the equilibrium dissociation constants (Kd) for YsiA binding being 20, 21, 37, 43, and 65 nm, respectively. YsiA binding was specifically inhibited by long chain acyl-CoAs with 14–20 carbon atoms, acyl-CoAs with 18 carbon atoms being more effective; out of long chain acyl-CoAs tested, monounsaturated oleoyl-CoA, and branched chain 12-metyltetradecanoyl-CoA were most effective. These in vitro findings were supported by the in vivo observation that the knock-out of acyl-CoA dehydrogenation through yusJ, ETFA, or etfB disruption resulted in YsiA inactivation, probably because of the accumulation of long chain acyl-CoAs in the cells. Furthermore, the disruption of yusL, yusK, yusJ, ETFA, etfB, or ykuG affected the utilization of palmitic acid, a representative long chain fatty acid. Based on this work, ysiA, ysiB, ykuF, ykuG, yhfL, yusM, yusL, yusK, yusJ, and ywjF can be renamed fadR, fadB, fadH, fadG, lcfB, fadM, fadN, fadA, fadE, and fadF.

Early timed follicular aspiration prevents severe ovarian hyperstimulation syndrome
Journal of Assisted Reproduction and Genetics, 1996Co-Authors: T Tomazevic, Helena Meden-vrtovecAbstract:Purpose: Early timed follicular aspiration (ETFA) of one ovary 10–12 hr after administration of chorionic gonadotropin (hCG) is an attempt to prevent severe ovarian hyperstimulation syndrome (OHSS). After the introduction of early timed follicular aspiration ETFA of one ovary in IVF/ET cycles at high risk for severe OHSS, no cases of severe OHSS were registered in the Ljubljana IVF/ET program. In the period before preventive ETFA (1984–1992) there were 16 cases of severe OHSS of 4798 IVF/ET cycles followed by 577 clinical pregnancies. After the introduction of ETFA (1992–1993) there were no cases of severe OHSS of 2289 IVF/ET cycles followed by 364 clinical pregnancies. Methods: We attempted to evaluate the significance of this observation by comparing two groups of female infertility IVF/ET cycles at high risk for severe OHSS. The occurrence of severe OHSS and clinical parameters in the two groups of IVF/ET cycles at high risk for severe OHSS were compared. Results: In the group of 106 IVF/ET female infertility cycles at high risk of severe OHSS with preventive ETFA, there were no cases of severe OHSS. In the control group of 92 IVF/ET female infertility cycles at high risk for severe OHSS with normally timed follicular aspiration (NTFA) of both ovaries, severe OHSS occurred in 16 cases.
