    Abstract Chemical investigations on secondary metabolites in natural products chemistry require efficient isolation techniques for characterization purpose as well as for the evaluation of their biological properties. In the case of phytochemical studies, the performance of the techniques is critical (resolution and yield) since the products generally present a narrow range of polarity and physicochemical properties. Several techniques are currently available, but HPLC (preparative and semipreparative) is the most widely used. To compare the performance of semipreparative HPLC and HPTLC for the isolation of secondary metabolites in different types of extracts, we have chosen carvone from spearmint essential oil (Mentha spicata L.), resveratrol from Fallopia multiflora (Thunb.) Haraldson, and rosmarinic acid from rosemary (Rosmarinus officinalis L.) extracts. The comparison was based on the chromatographic separation, the purity and quantity of isolated compounds, the solvent consumption, the duration and the cost of the isolation operations. The results showed that semipreparative HPTLC can in some case offer some advantages over conventional semipreparative HPLC.

    In April 2014, seedlings of Fallopia multiflora showing wilt symptom were first found at a greenhouse in Punggi-eup, Yeongju-si, Korea. A Fusarium-like fungus was isolated from the wilted plant and it was identified as Fusarium oxysporum based on morphological characteristics and nucleotide sequence data of translation elongation factor 1-a. The fungus isolated from the diseased plant was revealed to be pathogenic to the host plant through pathogenicity tests, and the reisolation of the pathogen confirmed Koch’s postulates. This is the first report of Fusarium wilt occurring on Fallopia multiflora in the world

    Abstract:

    A precise HPLC-DAD-based quantification together with the metabolomics statistical method was developed to distinguish and control the quality of Fallopia multiflora, a popular medicinal material in Vietnam. Multivariate statistical methods such as hierarchical clustering analysis and principal component analysis were utilized to compare and discriminate six natural and twelve commercial samples. 2,3,4′,5-Tetrahydroxystilbene 2-O-β-D-glucopyranoside (THSG) (1), emodin (4), and the new compound 6-hydroxymusizin 8-O-α-D-apiofuranosyl-(1⟶6)-β-D-glucopyranoside (5) could be considered as important markers for classification of F. multiflora. Furthermore, seven phenolics were quantified that the variation in the contents of selected metabolites revealed the differences in the quality of natural and commercial samples. Recovery of the compounds from the analytes was more than 98%, while the limits of detection (LOD) and the limits of quantitation (LOQ) ranged from 0.5 to 6.6 μg/ml and 1.5 to 19.8 μg/ml, respectively. The linearity, LOD, LOQ, precision, and accuracy satisfied the criteria FDA guidance on bioanalytical methods. Overall, this method is a promising tool for discrimination and quality assurance of F. multiflora products

    Four naphtolic glycosides (1–4), three anthraquinones (5–7), two stilbenes (8-9), one benzyl glycoside (10), and one flavonoid (11) were isolated from the roots of Fallopia multiflora. The new compounds were elucidated to be 6-hydroxymusizin 8-O-α-D-apiofuranosyl-(1→6)-β-D-glucopyranoside (1) and 6-methoxy-3-methyl-1,6,8-trihydroxy-2-naphthoic acid 8-O-β-D-glucopyranoside (3). Compound 3 could be an important marker for chemotaxonomy of F. multiflora. Benzyl gentiobioside (10) was isolated for the first time from the family Polygonaceae
