    Abstract:

    Pyrantel embonate and febantel are both constituents of Drontal Plus and Drontal Puppy broad spectrum anthelmintics for dogs. The effects of pyrantel and the febantel metabolite Fenbendazole were investigated against Toxocara canis in-vitro by studying changes in worm motility and tissue damage. Pyrantel and Fenbendazole were added to worms incubated in media for 8 h at the following concentrations: pyrantel: 12.2 microg, 25 microg, or 50 microg; Fenbendazole: 50 microg, 100 microg or 200 microg; mixture of pyrantel and Fenbendazole: 12.2 microg p + 50 microg f, 25 microg p + 100 microg f, 50 microg p + 200 microg f. Following this 8 h incubation period, one group of the worms was immediately fixed and studied by light- and electron microscopical examination. Other groups have been observed for further 8 h periods up to 56 hours and then studied in the same way.

    The effects of pyrantel and the febantel metabolite Fenbendazole - pyrantel and febantel are both constituents of Drontal® Plus and Welpan® - against Toxocara canisin-vitro were investigated by studying changes in worm motility and tissue damage. Pyrantel (P) (range : 12.2, 25 or 50 μg/ml) and Fenbendazole (F) (range : 50, 100 or 200 μg/ml) were added to the worms at different concentrations corresponding to plasma levels in different target animals obtained after oral dosing of the single drugs. Regarding motility changes there were no significant differences observable between pyrantel or Fenbendazole given as single drugs and the pyrantel/Fenbendazole combination. By contrast, when changes in tissue damages were studied there were significant differences detectable. Thus, hypodermis, muscles, gut, genital organs and nervous system got damaged severely at the concentrations of drug combinations P/F of 25/100 μg/ml. Similar effects could only be seen with the individual compounds at their highest level of concentration, e.g. 50 μg/ml of pyrantel or 200 μg/ml of Fenbendazole. In comparative analyses of irreversible tissue damages the synergistic effects observed were regarded as being so important that the conclusion must be drawn that these effects are also be present in-vivo in Drontal® Plus and Welpan® treated dogs resulting in a complete worm destruction.

    Abstract The aims of studies in 2002 and 2003 on three farms with 76 foals naturally infected with Parascaris equorum were to (i) identify if the nematode was resistant to ivermectin and moxidectin, and (ii) confirm the effectiveness of Fenbendazole and pyrantel pamoate for the parasite. Twelve clinical trials, each with a Fecal Egg Count Reduction Test, were conducted on two Thoroughbred and one Standardbred farms in southwestern Ontario, Canada. In each trial, Parascaris eggs/g feces were estimated for each foal pre- and post-treatment using the Cornell-Wisconsin double flotation and Cornell-McMaster dilution techniques. On each farm and for each trial, foals were randomized into treatment groups. Treatments were ivermectin, moxidectin, Fenbendazole, pyrantel pamoate administered at the manufacturers’ recommended dosages, and some foals were untreated. The overall efficacy for ivermectin was 33.5% (19 foals) and for moxidectin 47.2% (28 foals). Fenbendazole (16 foals) and pyrantel pamoate (21 foals) were highly effective for P. equorum each at 97.6%. For Fenbendazole, 15 foals had 100% and for pyrantel pamoate 17 foals had >97% with 14 at 100%.
