    An uncultured bacterium associated with the ectomycorrhizal fungus Tuber borchii Vittad. was identified as a novel member of the Cytophaga-Flexibacter-Bacteroides group. Utilizing a quantitative PCR targeting the 16S rRNA gene, we relatively quantified this bacterium in the host. The estimated number of bacteria was found to be approximately 106 cells per 30-day-old T. borchii mycelium culture. This represents the first molecular attempt to enumerate an uncultured bacterium associated with a mycorrhizal fungus.

    Mycorrhizal ascomycetous fungi are obligate ectosymbionts that colonize the roots of gymnosperms and angiosperms. In this paper we describe a straightforward approach in which a combination of morphological and molecular methods was used to survey the presence of potentially endo- and epiphytic bacteria associated with the ascomycetous ectomycorrhizal fungus Tuber borchii Vittad. Universal eubacterial primers specific for the 5′ and 3′ ends of the 16S rRNA gene (16S rDNA) were used for PCR amplification, direct sequencing, and phylogenetic analyses. The 16S rDNA was amplified directly from four pure cultures of T. borchii Vittad. mycelium. A nearly full-length sequence of the gene coding for the prokaryotic small-subunit rRNA was obtained from each T. borchii mycelium studied. The 16S rDNA sequences were almost identical (98 to 99% similarity), and phylogenetic analysis placed them in a single unique rRNA branch belonging to the Cytophaga-Flexibacter-Bacteroides (CFB) phylogroup which had not been described previously. In situ detection of the CFB bacterium in the hyphal tissue of the fungus T. borchii was carried out by using 16S rRNA-targeted oligonucleotide probes for the eubacterial domain and the Cytophaga-Flexibacter phylum, as well as a probe specifically designed for the detection of this mycelium-associated bacterium. Fluorescent in situ hybridization showed that all three of the probes used bound to the mycelium tissue. This study provides the first direct visual evidence of a not-yet-cultured CFB bacterium associated with a mycorrhizal fungus of the genus Tuber.

    Abstract:

    Macroscopic and microscopic features of natural and experimental Flexibacter maritimus infection, and epidemiological aspects of the disease, have been reported in a number of species of fish in Tasmanian aquaculture including Atlantic salmon, Salmo salar L., rainbow trout, Oncorhynchus mykiss (Walbaum), greenback flounder, Rhombosolea tapirina Gunther, and striped trumpeter, Latris lineata (Bloch & Schneider). There is a great deal of consistency in the pathology in salmonids and non-salmonid species, with erosive lesions of external surfaces being the most prominent clinical sign. Experimentally induced disease of salmonids and flounder is similar to natural infection. Mature lesions show dermal and gill erosion, with dermal bacterial invasion into the dense connective tissue and occasionally underlying musculature, but a remarkable lack of inflammatory response. The earliest lesions show consistent fragmentation and degeneration of the epithelium, with infiltration of amorphous protein-like materials and occasionally intra-epithelial cellular inflammatory cells, plus congestion and haemorrhage of the superficial dermis, but without visible bacteria in standard sections. Variable scale loss, oedema and a low level of inflammation in scale pockets, plus variable small adherent bacterial mats, are evident before full epithelial erosion.

    Abstract The susceptibility of freshwater and marine fish to infection by Flexibacter columnaris and Flexibacter maritimus was assessed in laboratory based experiments. Flexibactor columnaris produced a more severe disease in barramundi compared to goldfish. Both species suffered greater mortality at 25 °C than at 20 °C. Atlantic salmon ( Salmo salar ) and rainbow trout ( Oncorhynchus mykiss ) had similar levels of susceptibility to F. maritimus infection in seawater with approximate LC 50 values of 2.3 × 10 5 and 1.6 × 10 6 CFU mL −1 respectively. However, at salinity of 15 %. greenback flounder ( Rhombosolea tapirina ) were more resistant to F. maritimus than Atlantic salmon.

    . In vitro and in vivo efficacies of commonly used chemotherapeutants were determined for Cytophaga johnsonae, Cytophaga psychrophila, Flexibacter colutnnaris and Flexibacter maritimus. Treatmeat of barramundi, Lutes calcarifer (Bloch), with oxolinic acid (OA) as a bath (50 ppm) or by mouth (10 mg kg−1 body weight) resulted in serum levels above the minimum inhibitory concentration (MIC) for F. columnaris and produced significant clinical efficacy (P < 0.05). Amoxycillin (AM) was found to produce adequate serum levels against F. maritimus when used as a bath (200 ppm) or given orally (80 mg kg−1 body weight) to Atlantic salmon, Salmo salar L. and rainbow trout, Oncorhynchus mykiss (Walbaum), respectively. At these dose rates, this antibiotic was also clinically efficacious against F. maritimus (P <0.05). Trimethoprim produced more than adequate serum levels for the control of F. maritimus when given as a bath (50 ppm) or orally (10 mg kg−1 body weight). Trimethoprim was significantly more protective than AM when tested in vivo (P < 0.05). For C. johnsonae and C. psychrophila, the MIC values for OA and oxytetracycline were low, whereas that for TMP was high. Also, based on MIC values, C. psychrophila strains were more sensitive to AM and norfloxacin than C. johnsonae.
