    Ce travail s'inscrit dans le cadre d'un programme de recherche sur l'amélioration de la qualité de Plantes médicinales. Une démarche générale a été élaborée visant à augmenter la teneur et/ou le rendement en principes actifs et fait intervenir plusieurs voies de recherche : génétique, agronomique et technologique. Le mélilot (Melilotus officinalis L.) est une Légumineuse fourragère, utilisée comme matière première par l'industrie pharmaceutique. Ses extraits entrent dans la composition de médicaments à visée veinotonique. L'ensemble des études pharmacologiques publiées dans la littérature concerne la coumarine libre, qui possède des propriétés antiphlogistiques, antioedémateuses et qui est utilisée comme protecteur vasculaire. La coumarine libre n'existe pas dans le mélilot frais. Elle se trouve sous forme de glucosides d'acide ortho-hydroxy-cinnamique (GOHCA) trans et cis et localisés dans les vacuoles de la Plante, principalement au niveau des feuilles. Le GOHCA trans est isomérisé en GOHCA cis sous l'action des U.V., puis hydrolysé en coumarine libre par une ß-glucosidase inter-cellulaire spécifique, lors de la rupture des tissus de la Plante. Nous entendons par dérivés coumariniques la somme des deux glucosides précédemment cités exprimée en coumarine. L'objectif appliqué de ce travail est de produire du mélilot à forte teneur en dérivés coumariniques, et de transformer au mieux ces glucosides en coumarine libre au cours de la récolte et des étapes de transformation ultérieures afin de produire du matériel végétal à 0,7% de coumarine libre. En effet, la teneur moyenne en coumarine libre du mélilot utilisé par l'industrie pharmaceutique n'est que de 0,3%. L'objectif scientifique est de déterminer les facteurs susceptibles d'intervenir à la fois sur la synthèse des dérivés coumariniques du mélilot et sur leur transformation en coumarine libre. La première partie de ce travail a permis de comprendre et de maîtriser les mécanismes de formation de la coumarine libre durant les étapes de transformation postrécolte (séchage, extraction). Une technique d'extraction et une méthode de dosage en chromatographie en phase liquide ont été adaptées. Une étude de la variabilité génétique inter et intra origines concernant les caractères teneurs et rendements en dérivés coumariniques a permis la sélection de populations et de génotypes performants. Après avoir mis au point une technique de micropropagation, nous avons pu travailler avec des clones. La seconde partie de ce travail a permis d'avancer dans la compréhension des mécanismes de formation, de migration et de stockage des dérivés coumariniques dans la Plante. Nous avons pu montrer que les teneurs les plus fortes en dérivés coumariniques apparaissent dans les organes jeunes et physiologiquement actifs, comme les bougeons et les feuilles. Ceux-ci peuvent en contenir jusqu'à 3%. Le limbe, tissu possédant une forte activité photosynthétique, présente la plus importante teneur en dérivés coumariniques de la Plante à l'état végétatif. Nous avons également mis en évidence que : pour un génotype donné, la teneur en dérivés coumariniques est liée à l'âge de la Plante mais est indépendante de son poids ; les organes des niveaux inférieurs de la Plante possèdent toujours une teneur en dérivés coumariniques plus faible que ceux des niveaux supérieurs ; la teneur en dérivés coumariniques est indépendante de la teneur en azote ; la densité des Plantes en culture n'intervient pas sur la teneur en dérivés coumariniques alors qu'elle détermine de façon importante le rendement en matière sèche donc le rendement en dérivés coumariniques ; les dérivés coumariniques migrent des feuilles vers les racines en première année de culture ; la synthèse de dérivés coumariniques est dépendante à la fois de la quantité et de la qualité de la lumière. La troisième partie de ce travail concerne la mise au point d'un "itinéraire technique" visant à atteindre l'objectif de 0,7% de coumarine libre dans le matériel végétal sec. Aussi, nous sommes nous attachés à produire des organes jeunes en procédant à des récoltes précoces et multiples, effectuées sur une même parcelle. Du mélilot à forte teneur en dérivés coumariniques (plus de 1%) a été obtenu grâce à l'amélioration de la qualité du matériel végétal produit par le biais des techniques culturales. Le GOHCA cis a pu être totalement transformé en coumarine libre grâce à l'optimisation des techniques de récolte et de transformation du mélilot frais (conditions de séchage et d'extraction). La production de mélilot à plus de 0,7% de coumarine libre est maîtrisée. Les résultats obtenus, sur le mélilot, permettent ainsi de valider une démarche de recherche qui pourra être appliquée à l'amélioration des teneurs et rendements en principes actifs d'autres Plantes d'intérêt pharmaceutique.This work is included in a research programme concerning the quality improvement of medicinal Plants. Extraction and assay methods by liquid chromatography were performed. A technique of micropropagation was studied and applied to work with clones. The second part of this work consisted in the explanation of the mechanism involved in the synthesis and the storage of coumarin derivative in the Plant. In the literature only older, often in contradictions works are available. In our experiment, we showed that the higher coumarin derivative contents appeared in the young and physiologically active organs, such as buds and leaves. They may contain up to 3% of these molecules. The lamina with their high photosynthetic activity showed the highest level of coumarin derivative in the vegetative Plant. Experimentation during two years, on harvested clones, showed that: for a given genotype, the coumarin derivative level is determined by the age of the Plant and is independent from its weight ; the organs of the lower parts of the Plant always contain less coumarin derivatives than the upper parts ; the coumarin derivative level is independent from nitrogen content ; the Plant density in the field has no effect on the coumarin derivative level but interferes with the dry matter yield and therefore on the coumarin derivative yield ; the translocation of the coumarin derivatives from the root to the leaves takes place at the end of the first year of cultivation ; the coumarin derivative synthesis is linked both to the light intensity and composition. The third part of this work concerns the determination of a crop management that would produce 0.7% free coumarin in the dry matter. So, we emphasized the production of young organs by having early and multiple cuts. Some sweetclover with high coumarin derivative content (above 1%) were obtained by the improvement of the quality of the Plant material through agricultural techniques. The GOHCA cis could be totally transformed into free coumarin by the optimization of harvesting techniques and the transformation of fresh material (drying and extraction conditions). It was possible to control the production of Melilot with more than 0.7% of free coumarin. The results obtained here validate the present research approach, which could be applied to the improvement of content, and yield of other Plants of pharmaceutical interest

    Nendrinis dryžutis (Phalaris arundinacea L.) yra plačiai paplitęs paupiuose. Dėl didelės kūno masės šis augalas pastaruoju metu tiriamas kaip galimas biokuro šaltinis, fitoremediacijai tinkamas organizmas. Didelio dėmesio susilaukė nendrinio dryžučio invazija Amerikos žemyne. Nendrinis dryžutis dėl plataus prisitaikymo ribų dirvožemio drėgniui yra vertingas pašarinis augalas kintančio klimato sąlygomis. Nuo senų laikų nendrinio dryžučio dekoratyvinės formos yra auginamos įvairiuose pasaulio kampeliuose. Dekoratyvinių šio augalų formų tyrimai nesusilaukė pakankamo dėmesio Baltijos šalyse. Ypač pasigendama duomenų apie nendrinio dryžučio dryžuotosios formos augalų genetinę įvairovę. Nendrinis dryžutis – dažnas Lietuvos sodybų puošmenos elementas. Genetinė dryžuotosios formos nendrinio dryžučio įvairovė visiškai netirta, nežinomi šio augalo sklaidos keliai. Darbo tikslu buvo nendrinio dryžučio dryžuotajai formai tirti tinkamų DNR žymenų paieška. Darbo metodu pasirinktas DNR paprastųjų kartotinių sekų (mikrosatelitų) metodas, kaip vienas iš pastaruoju metu labiausiai naudojamų genetinės įvairovės vertinimo būdų. Darbe aprašoma DNR žymenų pritaikymo tyrimai. Iš penkiolikos žinomų mikrosatelitų žymenų, keičiant polimerazės grandininės reakcijos ciklų skaičių ir temperatūras, pavyko pritaikyti keturiolika. Šie molekuliniai žymenys bus naudojami surinktų įvairiose Lietuvos miestuose bei gyvenvietėse dryžuotosios formos nendrinio dryžučio individų tyrimamsReed canary grass (Phalaris arundinacea L.) is a widespread riparian species. Due to massive body, this Plant has recently been studied as a potential source of biofuel, an organism suitable for phytoremediation. Much attention was given to the invasion of the reed invasions along the America continent. Due to wide scale adaptations for soil moisture, it is valuable Fodder Plant in changing climate conditions. Since ancient times, the ornamental varieties of striped reed canary grass have been grown in the gardens of various corners of the world. Despite very popular in the greenery of allotments of the Baltic States, ornamental forms of reed canary grass has not received enough attention among breeders. In particular, there is a lack of information on the genetic diversity of striped forms of reed canary grass spread in Lithuania. The genetic diversity of the striped reed streamer is completely unknown, and the pathways of introduction of ornamental forms of this Plant are unknown for the Baltic States. The purpose of the work was to find out DNA markers suitable for the investigation of genetic variety of reed canary grass. Our research aimed at adjustment of microsatellite (SSR) DNA markers for evaluation of genetic diversity of striped form of reed canary grass accessions collected in Lithuania village and urban settlements. The paper describes the search for the conditions of application of the DNA SSR method. Of the fifteen examined microsatellite markers, fourteen were adapted changing conditions of polymerase chain reactions: number of cycles and temperature. Darbo tikslu buvo nendrinio dryžučio dryžuotajai formai tirti tinkamų DNR žymenų paieška. Darbo metodu pasirinktas DNR paprastųjų kartotinių sekų (mikrosatelitų) metodas, kaip vienas iš pastaruoju metu labiausiai naudojamų genetinės įvairovės vertinimo būdų. Darbe aprašoma DNR žymenų pritaikymo tyrimai. Iš penkiolikos žinomų mikrosatelitų žymenų, keičiant polimerazės grandininės reakcijos ciklų skaičių ir temperatūras, pavyko pritaikyti keturiolika. In particular, there is a lack of information on the genetic diversity of striped forms of reed canary grass spread in Lithuania. The genetic diversity of the striped reed streamer is completely unknown, and the pathways of introduction of ornamental forms of this Plant are unknown for the Baltic States. The purpose of the work was to find out DNA markers suitable for the investigation of genetic variety of reed canary grass. Our research aimed at adjustment of microsatellite (SSR) DNA markers for evaluation of genetic diversity of striped form of reed canary grass accessions collected in Lithuania village and urban settlements. The paper describes the search for the conditions of application of the DNA SSR method. Of the fifteen examined microsatellite markers, fourteen were adapted changing conditions of polymerase chain reactions: number of cycles and temperature.
