    We demonstrate a conduit for the delivery of a step change in the DNA analysis process: A fully integrated instrument for the analysis of multiplex short tandem repeat DNA profiles from reference buccal samples is described and is suitable for the processing of such samples within a Forensic Environment such as a police custody suite or booking office. The instrument is loaded with a DNA processing cartridge which incorporates on-board pumps and valves which direct the delivery of sample and reagents to the various reaction chambers to allow DNA purification, amplification of the DNA by PCR, and collection of the amplified product for delivery to an integral CE chip. The fluorescently labeled product is separated using micro capillary electrophoresis with a resolution of 1.2 base pairs (bp) allowing laser induced fluorescence-based detection of the amplified short tandem repeat fragments and subsequent analysis of data to produce a DNA profile which is compatible with the data format of the UK DNA databas...

    Abstract:

    : PCR-based DNA typing of biological evidence is now widely used in Forensic analyses due to the obvious advantages of enhanced sensitivity, the ability to distinguish discrete alleles and efficacy with degraded samples. A multiplex short tandem repeat (STR) system has been previously developed which successfully co-amplifies six STR loci HUMTH01, D21S11, D18S51, D8S1179, HUMVWF31/A and HUMFIBRA (FGA) in conjunction with the X-Y homologous gene Amelogenin. This is known as the second generation multiplex system (SGM). Detection of the PCR products is undertaken on ABD 373A or 377 automated sequencers using denaturing polyacrylamide gels coupled with fluorescent-based technology. We have evaluated this system for routine Forensic use and demonstrated that the technique is robust and reproducible under conditions consistent with those encountered in a Forensic Environment. A total of 132 stains from simulated and actual casework were analysed, together with relevant control areas and reference samples. The success rate was high with 76% of stains giving full profiles; we were also able to successfully detect and interpret mixtures. No mistyping was observed. A detailed examination of each of these profiles has assisted in the development of guidelines for casework interpretation. Although artefacts, stutter peaks and undenatured DNA were occasionally observed, these did not interfere with the accuracy of interpretation. In addition 38 samples, previously examined using the quadruplex system, were analysed with the SGM to enable a direct comparison to be made between the systems. The performance of the system with poor quality samples demonstrated its use as a rapid and powerful technique for individual identification.

    South Sumatra is one of the largest petroleum producing provinces in Indonesia, especially in the region of Musi Banyuasin Petroleum resources other than legally cultivated by Pertamina as government representatives, but on the other hand the community also participate through Illegal Drilling activities. This study aims to determine the hydrocarbon content and characterization of petroleum produced illegally by communities in the Sangadesa, Babattoman and Keluang districts through the biomarker analysis of the distribution of n-Alkane C10-C34 (m/z: 57), pristane, phytane, sterane C27-C29 (m/z: 217,218,259) and specific biomarker using Gas Chromatography Mass Spectroscopy agilent GCMSD 6890/5973i with data analysis using MSD Chemstation F.01.01.2317 and Library Database NIST14. Petroleum samples taken from 10 illegal wells with a depth range of 80-250 meters and production period of 3 months until 3 years. Oil is produced through The illegal drilling is not the main oil source rock but the result of migration. Hasil penelitian meninjukan karakteristik distribusi biomarker hidrokarbon dalam sampel  sampel minyak dari lokasi Babattoman dan Keluang  biomarker n-Alkana C12 sampai C30 tidak ditemukan untuk menunjukkan bahwa minyak bumi memiliki biodegradasi moderat, sampel minyak dari Keban sedikit terdegradasi. Rasio Pr / Ph, Pr / nC17, dan Ph / nC18 menunjukkan sumber bahan organik sumber laut yang tersimpan dalam kondisi lingkungan anoksik pengendapan.     The demand for high-throughput DNA profiling has increased with the introduction of national DNA databases and has led to the development of automated methods of short tandem repeat (STR) profile production; however, a potential bottleneck still exists at the gel electrophoresis stage. Capillary electrophoresis sequencers capable of processing 96 samples with considerably reduced manual intervention are now available. In this paper, we compare the ABI Prism 377 slab-gel sequencer currently used by the Forensic Science Service with three leading capillary array electrophoresis instruments: the ABI Prism 3700, the Amersham MegaBACE 1000 and the 16-capillary ABI Prism 3100. We describe the experiments used to evaluate and validate these platforms for Forensic use with the STR multiplex Ampf/STR SGMplus [1, 2], along with comparative data from the ABI Prism 377 sequencer.
