    Toll-like receptor-4 (TLR4) is a primary receptor of the innate immune reaction and compelling evidence demonstrates its involvement in the pathoGenesis of atherosclerosis and stroke. TLR4 is constitutively expressed on monocytes and endothelial cells; it is highly expressed in atherosclerotic plaques and in peripheral blood of patients after ischemic stroke. Polymorphisms in the promoter region that alter the transcriptional regulation of this Gene may represent Genetic risk factors involved in the predisposition to atherosclerotic disease. In this study we investigated the effect on TLR4 Gene Expression of three polymorphisms in the upstream regulatory region at positions −1607T>C/rs10759932, −2026A>G/rs1927914 and −2604G>A/rs10759931 in peripheral blood of atherosclerotic patients. RNA from individuals homozygous for the −2604A allele showed a lower Expression of the Gene when compared to patients carrying the counterparts GG+GA. Electrophoretic mobility shift assays showed differences in the electrophoretic mobility of the DNA–nuclear protein complexes formed by the G>A variants, suggesting that the two alleles differ in their binding affinity to transcriptional factors.

    Abstract:

    Aims The aim of this study was to investigate the effect of ethanol exposure on the Expression Level of geranylgeranyl pyrophosphate synthase Gene involved in the metabolism of Oenococcus oeni to probe the mechanisms of ethanol tolerance correlated with adaptive changes. Methods and Results The evaluation of ten potential internal control Genes and the comparative study of their stability were performed to select the most stable internal controls for the normalization of Expression data. The Expression Level analysis by qPCR and changes after exposure to ethanol stresses highlighted a significant increase in the presence of higher ethanol concentrations. Conclusions The analysis of results suggest that O. oeni adjusts the Expression of Genes to adapt to stress conditions and the high Expression Level of ggpps would allow a flow of isoprenoid precursors towards the carotenoids and related pathways to stabilize bacterial cell membranes, improving the cell membrane disturbances and preventing cell death induced by ethanol. Significance and Impact of the Study The involvement of ggpps Gene in physiological changes of bacterial behaviour confirmed the exposure to stress requires the activation of defence mechanism to be more tolerant to adverse conditions. Improving the knowledge of stress tolerance and adaptation mechanisms of O. oeni is essential to enhance the efficiency of the malolactic starter in wine and to obtain the development of starters able to survive to direct inoculation with a large benefit for wine technology.

  • Predicting Gene Expression Level from Codon Usage Bias

    Abstract:

    The "Expression measure" of a Gene, E(g), is a statistic devised to predict the Level of Gene Expression from codon usage bias. E(g) has been used extensively to analyze prokaryotic genome sequences. We discuss 2 problems with this approach. First, the formulation of E(g) is such that Genes with the strongest selected codon usage bias are not likely to have the highest predicted Expression Levels; indeed the correlation between E(g) and Expression Level is weak among moderate to highly expressed Genes. Second, in some species, highly expressed Genes do not have unusual codon usage, and so codon usage cannot be used to predict Expression Levels. We outline a simple approach, first to check whether a genome shows evidence of selected codon usage bias and then to assess the strength of bias in Genes as a guide to their likely Expression Level; we illustrate this with an analysis of Shewanella oneidensis.

    Abstract:

    Background Recent studies have shown that the phosphatidylinositol 3-kinase (PI3K) signaling pathway is important regulator of many cellular events, including apoptosis, proliferation and motility. PI3K pathway alterations ( PIK3CA Gene mutations and/or amplification) have been observed in various human tumours. In the majority of diagnosed cases, mutations are localized in one of the three "hot spots" in the Gene, responsible for coding catalytic subunit α of class I PI3K ( PIK3CA ). Mutations and amplification of PIK3CA Gene are characteristic for thyroid cancer, as well. Methods The aim of our study was to examine a Gene Expression Level of PIK3CA in fine-needle aspiration biopsy (FNAB) thyroid specimens in two types of thyroid lesions, papillary thyroid carcinoma (PTC) and non-toxic goitre (NTG). Following conventional cytological examination, 42 thyroid FNAB specimens, received from patients with PTC (n = 20) and NTG (n = 22), were quantitatively evaluated regarding PIK3CA Expression Level by real-time PCR in the ABI PRISM^® 7500 Sequence Detection System. Results Significantly higher Expression Level (RQ) of PIK3CA in PTC group has been noted in comparison with NTG group (p < 0.05). Conclusion These observations may suggest role of PIK3CA alterations in PTC carcinoGenesis.
