    Abstract A modified method of inter-retrotransposon amplified polymorphism (IRAP) with primers annealing to the conserved reverse transcriptase sites instead of the conserved long-terminal repeat (LTR) regions and retrotransposon–microsatellite amplified polymorphism (REMAP) markers were used to investigate Genetic Relationships of some Japanese persimmon ( Diospyros kaki Thunb.) genotypes from Japan and China and the related species. In total, 117 scorable amplification bands ranging from 250 to 2000 bp were generated by IRAP and REMAP analysis, among which 101 (86%) were polymorphic, with an average of six polymorphic bands per primer combination. The analysis of Genetic Relationships of 27 genotypes revealed that the Japanese, Chinese genotypes and related species were distinctly separated. The phyloGenetic trees constructed by unweighted pair-group method with arithmetic averages (UPGMA) and Wagner parsimony cluster methods agreed well with the geographic origin information. The Japanese persimmon genotypes from Japan and China may have different Genetic background, suggesting a long independent domestication, intercrossing and breeding history in both regions. ‘Baogaitianshi’ and ‘Xiaobaogaitianshi’ had a high similarity coefficient, implying that they possibly have the bud sport Relationships. The related species were clearly clustered in a separated group, suggesting that they are very closely related.

    Genetic Relationships among 104 accessions of Cucurbita pepo were assessed from polymorphisms in 134 SSR (microsatellite) and four SCAR loci, yielding a total of 418 alleles, distributed among all 20 linkage groups. Genetic distance values were calculated, a dendrogram constructed, and principal coordinate analyses conducted. The results showed 100 of the accessions as distributed among three clusters representing each of the recognized subspecies, pepo, texana, and fraterna. The remaining four accessions, all having very small, round, striped fruits, assumed central positions between the two cultivated subspecies, pepo and texana, suggesting that they are relicts of undescribed wild ancestors of the two domesticated subspecies. In both, subsp. texana and subsp. pepo, accessions belonging to the same cultivar-group (fruit shape) associated with one another. Within subsp. pepo, accessions grown for their seeds or that are generalists, used for both seed and fruit consumption, assumed central positions. Specialized accessions, grown exclusively for consumption of their young fruits, or their mature fruit flesh, or seed oil extraction, tended to assume outlying positions, and the different specializations radiated outward from the center in different directions. Accessions of the longest-fruited cultivar-group, Cocozelle, radiated bidirectionally, indicating independent selection events for long fruits in subsp. pepo probably driven by a common desire to consume the young fruits. Among the accessions tested, there was no evidence for crossing between subspecies after domestication.

    Cucurbita pepo (pumpkin, squash, gourd), an economically important species of the Cucurbitaceae, is extremely variable in fruit characteristics. The objective of the present study was to clarify Genetic Relationships across a broad spectrum of the C. pepo gene pool, with emphasis on domesticates, using AFLP, ISSR and SSR markers. Forty-five accessions were compared for presence or absence of 448 AFLP, 147 ISSR, and 20 SSR bands, their Genetic distances (GDs) were estimated and UPGMA cluster analysis was conducted. The results obtained from these three marker systems were highly correlated (P ≪ 0.001). Clustering was in accordance with the division of C. pepo into three subspecies, fraterna, texana and pepo, with the first two less distant to one another than to the last one. Within the clusters, sub-clustering occurred in accordance with fruit shape and size. The subsp. texana cluster consisted of six sub-clusters, one each for the representatives of its five cultivar-groups (Acorn, Crookneck, Scallop, Straightneck and Ovifera Gourd) and wild gourds. Within the subsp. pepo cluster, the representatives of two cultivar-groups (Zucchini and Orange Gourd) formed distinct sub-clusters and the representatives of two other groups (Cocozelle and Vegetable Marrow) tended to sub-cluster separately from one another but formed an assemblage with the representatives of the remaining group (Pumpkin). Within-group GDs were less than corresponding between-group GDs in nearly all comparisons. The smallest-fruited accession, 'Miniature Ball', appears to occupy a Genetically central position within C. pepo.
