Germination and the role of extracellular matrix in adhesion of urediniosPores of Uromyces viciae-fabae to synthetic surfaces

Butt, Alan BeckettAbstract:UrediniosPores and sPorelings of Uromyces viciae-fabae on hydrophilic or hydrophobic synthetic substrata, produce extracellular matrix and adhere. Three phases of extracellular matrix release were observed with TEM & LTSEM: ( a ) from the sPore surface post-imbibition, ( b ) extrusion of digested polymers from the Germ Pore by growth of the incipient Germ-tube, and ( c ) progressive asymmetric distribution dorsiventrally along the extending Germ-tube. During the first 20 min post-hydration, fewer sPores adhered to cellulose acetate than to more hydrophobic substrata, indicating that hydrophobic interactions contribute to adhesion at this stage. On all surfaces, adhesion increased between 40–120 min due to release of lysed Pore material and extracellular matrix produced concurrent with Germ-tube growth. Inhibition of β-glucosidase with 50 m m-d -gluconic acid lactone, did not affect initial sPore adhesion and Germ Pore lysis but Germ-tube growth and the associated increase in adhesion were inhibited. It is proposed that the pre-invasion process of urediniosPores involves passive and active phases of non-specific adhesion.

High humidity or free water initiate Germination. SPores imbibe free water, swell and Germ Pores are lysed. Extracellular matrix is produced and possibly released through the Germ Pore. Emerging Germ tubes are enclosed in a sheath of matrix and adhere tightly to the substratum. This is a non- specific response. Several factors, either singly or in combination, may induce the formation of appressorium-like structures. These are discussed in relation to the availability of water and the topography of the substratum surface.

Another lineage of secotioid fungi is discovered: Psathyrella secotioides sp. nov. from Mexico

A phylogeny based on ITS, LSU, and ef -1α markers demonstrate that P. secotioides nests with strong support in the Psathyrellaceae, more precisely in the Psathyrella candolleana clade, representing the first secotioid form known in the Psathyrellaceae. Psathyrella secotioides is sister to a clade encompassing P. candolleana , P. badiophylla , P. leucotephra , and P. hymenocephala . Dark brown sPores without a Germ Pore and a hymeniform pileipellis structure are synapomorphic characters shared with the rest of the species of the P. candolleana clade ( Psathyrella sect. Spintrigerae). On the contrary, longer sPores with thicker walls, and the apparent loss of cheilocystidia are derived characters, probably linked with producing secotioid basidiomata and related to the loss of forcible sPore discharge. Estimation of divergence times using a molecular clock suggests that Psathyrella secotioides diverged from the core P. candolleana group around 3.6–5.1 Mya (1.6–9.5), approximately when the climate aridization that originated the Sonoran desert started. Psathyrella secotioides is described macro- and microscopically in detail, and microphotographs under optical microscope and SEM are provided.
