    Abstract:

    Date palm fruit production has great economic significance for many countries. There is a fundamental necessity to conserve valuable date palm Germplasm, but there are various problems with in vivo and ex situ Conservation. In vitro storage has several advantages over conventional Germplasm Conservation methods. The in vitro technique offers a developed method of slow-growth storage, which is considered as an alternate solution for short- and medium-term storage of date palm Germplasm under controlled conditions. Minimal growth conditions for Germplasm Conservation are generally achieved by reducing growth rate through modification of environmental growing conditions and culture, by using low temperatures, and the addition of growth retardants and osmotic agents. This chapter describes a protocol for short-term in vitro Conservation of date palm shoot-tip and callus cultures under slow-growth storage conditions, using sucrose as an osmotic agent and abscisic acid (ABA) as a growth retardant at 15 °C for 12 months.

    Germplasm Conservation of the tropical forest trees,Cedrela odorata L.,Guazuma crinita Mart., andJacaranda mimosaefolia D. Don., at above-freezing temperatures following alginate-bead encapsulation was attempted. Shoot tips excised from in vitro plantlets were encapsulated in calcium-alginate beads and stored on different substrates at 12, 20, and 25 °C. Percent viability when encapsulated shoot tips were stored on substrate containing only water solidified with 1% (wt/vol) agar was 80% after 12 months at 12°C forC. odorata, 90% after 12 months at 25°C forG. crinita, and 70% after 6 months at 20°C forJ. mimosaefolia.

    Abstract:

    Cryopreservation approach for the Germplasm Conservation of three useful tropical forest tree species (Cedrela odorata L., Guazuma crinita Mart., and Jacaranda mimosaefolia D. Don.) were carried out using shoot-tip or roottip explants from in vitro-grown plantlets by four cryopreservation methods ((1) simple freezing, (2) rapid freezing, (3) slow pre-freezing, and (4) dehydration method). The effects of cold hardening treatments (5, 10, 15, and 20/10°C) and pre-culture (dehydration treatments) were also studied.The best results were achieved when the shoot-tips were cooled by slow pre-freezing before immersion in liquid nitrogen (LN). Survival and plant recovery rates of 50 and 20%; and, 50 and 15%; were obtained in cryopreserved shoot-tips of C. odorata and G. crinita, respectively. Although the effects of cold hardening and/or pre-culture treatments are not clear, apparently they were not effective in enhancing the survival or plant recovery rate after immersion in LN because these species may be intolerant of low temperature treatments of long duration and/or to drastic dehydration treatments.The cryopreservation of shoot-tips from tissue-cultured plants can be considered as a feasible alternative for the long-term storage of C. odorata, G. crinita, and J. mimosaefolia Germplasm.
