The difference in the secondary metabolisms of closely related plant species and their hybrids has rarely been addressed. In this study, we conducted a holistic secondary metabolomics analysis of three medicinal Glycyrrhiza species (G. uralensis, G. glabra, and G. inflata), which are used as the popular herbal medicine licorice. The Glycyrrhiza species (genotype) for 95 batches of samples were identified by DNA barcodes of the internal transcribed spacer and trnV-ndhC regions, and the chemotypes were revealed by LC/UV- or LC/MS/MS-based quantitative analysis of 151 bioactive secondary metabolites, including 17 flavonoid glycosides, 24 saponins, and 110 free phenolic compounds. These compounds represented key products in the biosynthetic pathways of licorice. The structures of 1–13 were identified on the basis of extensive NMR and MS data analyses. The sugar residues were identified by gas chromatography and ion chromatography coupled with pulsed amperometric detection after hydrolysis. Saponins containing a galacturonic acid (1–3) or xylose (5) residue are reported from Glycyrrhiza species for the first time. Compounds 1, 7, 8, and 24 exhibited good inhibitory activities against the influenza virus A/WSN/33 (H1N1) in MDCK cells with IC50 values of 48.0, 42.7, 39.6, and 49.1 μM, respectively, versus 45.6 μM of the positive control oseltamivir phosphate. In addition, compounds 24 and 28 showed anti-HIV activities with IC50 values of 29.5 and 41.7 μM, respectively.

Liquorice root is a significant medicinal herb due to its large spectrum of pharmacological effects and the numerous traditional indications in Asian and European cultures, e.g. bronchial catarrh, gastric and duodenal ulcers. Recent scientific knowledge about the liquorice root gives hope that a new important chapter in the fascinating history of this phenomenal medicinal herb may be written because of its antiviral effects.

They all mention its use for symptoms of viral respiratory tract infections and hepatitis. Randomized controlled trials confirmed that the Glycyrrhiza glabra derived compound glycyrrhizin and its derivatives reduced hepatocellular damage in chronic hepatitis B and C. In hepatitis C virus-induced cirrhosis the risk of hepatocellular carcinoma was reduced. Animal studies demonstrated a reduction of mortality and viral activity in herpes simplex virus encephalitis and influenza A virus pneumonia. In vitro studies revealed antiviral activity against HIV-1, SARS related coronavirus, respiratory syncytial virus, arboviruses, vaccinia virus and vesicular stomatitis virus. Mechanisms for antiviral activity of Glycyrrhiza spp. include reduced transport to the membrane and sialylation of hepatitis B virus surface antigen, reduction of membrane fluidity leading to inhibition of fusion of the viral membrane of HIV-1 with the cell, induction of interferon gamma in T-cells, inhibition of phosphorylating enzymes in vesicular stomatitis virus infection and reduction of viral latency. Future research needs to explore the potency of compounds derived from licorice in prevention and treatment of influenza A virus pneumonia and as an adjuvant treatment in patients infected with HIV resistant to antiretroviral drugs. optimization of extraction process of Glycyrrhiza glabra polysaccharides by response surface methodology
Carbohydrate Polymers, 2008Co-Authors: Liang RenjieAbstract:Abstract Experimental design was used to investigate the effect of three parameters (extraction time, extraction number and ratio of water to raw material) on polysaccharides yields. The ranges of the factors investigated were 3.5–4.5 h for extraction time ( X 1 ), 4–6 for extraction number ( X 2 ), and 25–35 for ratio of water to raw material ( X 3 ). The statistical analysis of the experiment indicated that extraction time and ratio of water to raw material had significant effect on Glycyrrhiza glabra polysaccharides yields. The central composite design showed that polynomial regression models were in good agreement with the experimental results with the coefficients of determination of 0.924 for Glycyrrhiza glabra polysaccharides yield. The optimal condition for Glycyrrhiza glabra polysaccharides yield within the experimental range of the variables studied was at 4.3 h, 6, and 35. At this condition, the predicted yield of polysaccharides extracted was 3.6%.

Antimycobacterial activity of Glycyrrhiza glabra was found at 500 μg/mL concentration. Bioactivity guided phytochemical analysis identified glabridin as potentially active against both Mycobacterium tuberculosis H37Ra and H37Rv strains at 29.16 μg/mL concentration. It exhibited antimicrobial activity against both Gram-positive and Gram-negative bacteria. Our results indicate potential use of licorice as antitubercular agent through systemic experiments and sophisticated anti-TB assay.
