To comprehend the kinetics of R,S-Goitrin and promote its curative application, a rapid and sensitive UHPLC-MS/MS method was developed. The selected reaction monitoring transitions were m/z 130.0 → 70.0 for R,S-Goitrin and m/z 181.1 → 124.0 for the internal standard in a positive-ion mode. The established UHPLC-MS/MS method achieved good linearity for R,S-Goitrin at 10-2000 ng/mL. The intra- and interday accuracy levels were within ±9.7%, whereas the intraday and interday precision levels were <11.3%. The extraction recovery, stability and matrix effect were within acceptable limits. The validated method was successfully applied for the pharmacokinetic analysis of R,S-Goitrin in rats after oral administration. Moreover, a total of six metabolites were structurally identified through UHPLC-Q/TOF-MS. The proposed metabolic pathways of R,S-Goitrin in rats involve demethylation, acetylation, glutathionylation and oxygenation.

A standard Goitrin–epiGoitrin racemic mixture was used for method development. A total of six chiral stationary phases (CSPs) were screened and the (S,S)-Whelk-O 1 (4.6 × 250 mm, 10 μm) column was chosen as it offers baseline resolution for the enantiomeric pair with separation accomplished in 6 minutes. A single quadrupole MS and a PDA detector were used in line for the detection. The validated method was applied successfully in the analysis of different samples. Results indicated that the developed assay was fast, sensitive and reproducible. SFC should be an integral part of the overall analytical platform for TCM and natural product research, especially in the area of chiral analysis.

Ninety-three batches of samples were investigated and the common fingerprinting profile of water-soluble extracts was established with professional analytical software recommended by the State Food and Drug Administration (SFDA). The reference chromatogram was also generated with 24 common peaks showing large peak areas and good separation from adjacent peaks, the total peak areas of 24 common peaks were more than 85% of the total peak areas. modulation of glutathione s transferase activity and isozyme pattern in liver and small intestine of rats fed Goitrin and t3 supplemented diets

BjeldanesAbstract:Goitrin is a potent goitrogen that has been shown to induce glutathione S-transferase (GST) activity and to increase aflatoxin detoxification. In the present study with rats, dietary Goitrin (200 mg/kg diet) produced a hypothyroid state and significantly increased levels of hepatic GSSG (1.4-fold), GST protein (1.4-fold) and GST activity against chlorodinitrobenzene (CDNB) (1.7-fold). Cotreatment with dietary triiodothyronine (T3) reversed these effects in a dose-related manner. Intestinal GST activities against CDNB and epoxynitrophenoxypropane did not change with Goitrin or T3 treatment. HPLC analysis showed that, in the liver, Goitrin treatment increased the levels of GST-1b and -7 by 3.5- and 5-fold, respectively, and decreased the level of GST-3 by 50%. Cotreatment with T3 returned levels of GST-7 and -3 to control levels but only partially reduced the level of GST-1b. In the small intestine, Goitrin increased the level of GST-1b by 28% and decreased the level of GST-7 by 34% compared with those of controls; thyroid hormone treatment produced no additional effect on GST in this organ. Selenium deficiency altered thyroid hormone status but significantly affected the level only of hepatic GST-3, which was reduced by 30% compared with that of controls. These results indicate that a modified thyroid hormonal status plays an important role in the GST-inducing effects of Goitrin. A possible mechanism of thyroid-dependent GST induction by Goitrin is discussed.

Some evidence suggests that it arises, in part, from variation in ability to perceive Goitrin (5-vinyloxazolidine-2-thione), a potent antithyroid compound found naturally in crucifers. Individuals vary in ability to perceive synthetic compounds similar to Goitrin, such as 6-propyl-2-thiouracil (PROP) and phenylthiocarbamide (PTC), as the result of mutations in the TAS2R38 gene, which encodes a bitter taste receptor. This suggests that taste responses to Goitrin itself may be mediated by TAS2R38. To test this hypothesis, we examined the relationships between genetic variation in TAS2R38, functional variation in the encoded receptor, and threshold taste responses to Goitrin, PROP, and PTC in 50 subjects. We found that threshold responses to Goitrin were associated with responses to both PROP ( P= 8.9 · 10 4 ; rs = 0.46) and PTC ( P= 7.5 · 10 4 ; rs = 0.46). However, functional assays revealed that Goitrin elicits a weaker response from the sensitive (PAV) allele of TAS2R38 (EC50 = 65.0 lM) than do either PROP (EC50 = 2.1 lM) or PTC (EC50 = 1.1 lM) and no response at all from the insensitive (AVI) allele. Furthermore, Goitrin responses were significantly associated with mutations in TAS2R38 ( P= 9.3 · 10 3 ), but the same mutations accounted for a smaller proportion of variance in Goitrin response (r 2 = 0.16) than for PROP (r 2 = 0.50) and PTC (r 2 = 0.57). These findings suggest that mutations in TAS2R38 play a role in shaping Goitrin perception, but the
