  • Effect of "Xinshuai Granule" on Echocardiography and Myocardial Fibrosis in Patients with Chronic Heart Failure

    Abstract:

    Objective: To investigate the effect of "Xinshuai Granule" on cardiac function and myocardial Fibrosis in patients with chronic Heart failure(CHF) of qi-deficiency,blood-stasis and water-retention syndrome.Methods: 60 patients were randomly assigned to treatment group receiving routine therapy combined with "Xinshuai Granule" and control group receiving routine therapy only for six weeks.Before and after the treatment,precollagen Ⅲ peptide(PⅢP),laminin(LN) and echocardiography were examined.Results: Compared with control group,treatment group has lower left ventricular mass index(P 0.05),higher levels of EF and VE/VA,and lower levels of PⅢP and LN(P 0.05).Conclusion: "Xinshuai Granule" can improve Heart function and suppress Heart Muscle Fibrosis of patients with CHF.

  • Experimental Study of Formula of Jiang-ya Qing-xin on Myocardial Fibrosis and Neuroendocrine in Spontaneous Hypertensive Rat

    Abstract:

    Objective: To observe the curative effect of Formula of Jiang-ya Qing-xin on Heart Muscle Fibrosis,AngⅡ and Ⅲ pro-collagen amino peptide(PⅢNP) in spontaneous hypertensive rat.Methods: 24 male rats of eight to ten weeks' age with spontaneous hypertension were divided into 4 groups randomly,which were model control group,TCM group(group of Formula of Jiang-ya Qing-xin),western medical group(group of ramipril),and integrated group(group of Formula of Jiang-ya Qing-xin and ramipril).There were 6 rats in each group.In addition,normal control group included 8 SD rats of eight weeks'age.After four weeks' administration,AngⅡ,PⅢNP and pathology were detected.Results: In model control group AngⅡ and PⅢNP were higher than those in normal control group(P0.01),but those in TCM group,western medical group and integrated group were lower than in model control group,and significant difference showed between western medical group and integrated group and model control group(P0.05).In normal control group cardiac Muscle fibers lined up in order,and their size and shape were normal.In model control group cadiocytes were in overgrowth,cardiac Muscle interstitial collagen fibers obviously increased,and the collagen fibers around cardiac Muscle cells arranged in disorder.In TCM group myocardial interstitial was in mild hyperplasia,and cardiac Muscle cells were fat lightly and moderately.In western medical group cardiac Muscle fibers lined up in order,but cellular diameter transverse was swelling lightly.In integrated group cardiac interstitial collagen fibers grew downwards markedly,but intercellular collagen fibers lined up in order.Conclusion: Formula of Jiang-ya Qing-xin can inhibit Heart Muscle Fibrosis of spontaneous hypertensive rat,and the mechanism is to inhibit AngⅡ and PⅢNP to bring into full play.

  • The Construction and Pharmacodynamic Action of a Novel Anti-renal Hypertension DNA Vaccine Targeted AngiotnensinII

    Abstract:

    In this article,we designed a DNA vaccine targeted angiotensin II to investigate its pharmacodynamic action and safety and tried to explain the mechanism in model rats.Methods:This DNA vaccine has six tandem repeats of angiotensin II as the immunological epitope and hepatitis B virus core antigen as a vaccine carrier.As angiontensin II is endogenous,we used the plasmid of Interleukin Ⅳ as the adjuvant to avoid the immune tolerance.We injected this vaccine into model rats of renal hypertension to investigate its immunogenicity and pharmacodynamic action.Results:This DNA vaccine can induce high titer specific antibody against angiotensin II for almost eight weeks accompanied with the degradation of serum angiotensin II level.When the antibody titer reaches the peak,the systolic blood pressure(SBP) degraded 20mmHg and the diastolic blood pressure(DBP) degraded 9mmHg.Besides,the histopathologic results indicated that this DNA vaccine could also inhibit the progress of Heart Muscle Fibrosis and effectively retroconverse the myocardial hypertrophy of model rats.Conclusion: The angiotensin II as a potencial target of hypertension can be inhibited by DNA vaccine in immunological pathway.This article shows a novel design and provides a method for antihypertension vaccine research.
