    Abstract:

    The study was conducted in the “Mare aux Hippopotames” Biosphere Reserve of Burkina Faso and aimed at assessing the number, age structures and seasonal mobility of Hippopotamus amphibius. Methodology used was based on three consecutive-year (2006-2008) monitoring and ground inventory inside and outside the reserve. The results of the inventories have led to the identification of 41 Hippopotamus in 2008 versus 35 in 2006 which are divided into 3 distinct herds. The structure in age group of that population was composed of 32 were adults, 5 sub-adults and 4 juveniles. Location of places where they take a rest varies with the level of the water in the pond. The inventory has identified eight sites on each side of the pond as used by hippos to harvest food. In addition, exploration of the reserve has identified four temporary ponds located near the water meadows used by hippos during the rainy season. These areas were surrounded by agricultural fields that were often raided by these mammals for food. It is urgent to establish agricultural zoning and to grant a special conservation status to the four additional ponds to minimize conflicts between humans and Hippopotamus. Finally, local participation in conservation activities is probably greatly responsible for an increase in the size of the hippo population.

Progesterone metabolites were extracted from the feces with 80% MeOH and quantified using a progestagen EIA. Fecal progestagen analysis indicated hippopotami had a cycle length of 35.3±1.3 days and ovulated throughout the year. The observed calving interval in females with lactational anovulation was 17 months. The majority of births (63.6%) occurring during the summer rainy season and the inconsistency of lactational anovulation suggested the possibility of environmental influences on reproduction.

    The population decline of the common Hippopotamus (Hippopotamus amphibius) has necessitated the preservation of their genetic resources for species conservation and research. Of all actions, cryopreservation of fibroblast cell cultures derived from animal biopsy is considered a simple but efficient means. Nevertheless, preserving viable cell cultures of the common Hippopotamus has not been achieved to our knowledge. To this end, we established and characterized fibroblast cell cultures from the skin sample of a newborn common Hippopotamus in this study. By combining the tissue explant direct culture and enzymatic digestion methods, we isolated a great number of cells with typical fibroblastic morphology and high viability. Neither bacteria/fungi nor mycoplasma was detectable in the cell cultures throughout the study. The population doubling time was 34 h according to the growth curve. Karyotyping based on Giemsa staining showed that the cultured cells were diploid with 36 chromosomes in all, one pair of which was sex chromosomes. Amplified mitochondrial cytochrome C oxidase subunit I gene sequence of the cultured cells was 99.26% identical with that of the registered Hippopotamus amphibius complete mitochondrial DNA, confirming the species of origin of the cells. Flow cytometry and immunofluorescence staining results revealed that the detected cells were positive for fibroblast markers, S100A4 and Vimentin. In conclusion, we generated the fibroblast cell cultures from a common Hippopotamus and identified their characteristics using multiple techniques.
