Odontologia.Os compostos de silício orgânico (SO), sulfato de glucosamina (SG) e o Metilsulfonilmetano (MSM) têm sido utilizados rotineiramente, via oral, para tratamento de problemas articulares, tendo função analgésica, anti-inflamatória e efeitos sobre o metabolismo ósseo. Sendo assim, o objetivo deste estudo foi avaliar uma composição de silício orgânico (SO), sulfato de glucosamina (SG) e MSM em Teste de histocompatibilidade em dorso de rato, e posteriormente analisar o efeito de sua aplicação no processo de regeneração de defeitos ósseos mandibulares de ratos da raça Wistar. Para o Teste de histocompatibilidade foram utilizados 9 ratos que receberam no dorso o implante de 2 tubos de polietilieno, um contendo a composição de 70% SO + 15% SG + 15% MSM e o outro contendo guta percha (controle). As amostras foram coletadas após 7, 14 e 21 dias e processadas para inclusão em parafina e coloração em hematoxilina e eosina. Foi analisado a presença de tecido de granulação, neutrófilos, eosinófilos, linfócitos, macrófagos e abscesso. Em relação ao processo inflamatório este foi classificado como ausente, presente e infiltrado. No Teste de histocompatibilidade a composição obteve boa resposta tecidual, com as amostras apresentando inflamação moderada à intensa em 7 dias e leve em 14 e 21 dias, possibilitando então o encaminhamento da mesma para ser utilizada localmente em defeitos ósseos mandibulares de ratos. Para tanto, foram utilizados 9 ratos Wistar. Um defeito ósseo com altura e profundidade de 1mm e 3mm de comprimento, no lado direito e esquerdo da mandíbula, sob a coroa do primeiro molar inferior foi realizado. No lado direito foi aplicada a composição (grupo experimental); o lado esquerdo não recebeu tratamento (controle). Após 7, 14 e 28 dias de pós-operatório as amostras foram coletadas e processadas para a análise em Microscopia Eletrônica de Varredura (MEV). Nos defeitos de 7 dias as margens estavam regulares, sem presença significativa de osso novo em ambas as amostras. Aos 14 dias, no lado controle houve diminuição da profundidade do defeito; no lado tratado o defeito apresentava limites irregulares e ampla reabsorção nas margens, na cortical vestibular adjacente e na crista óssea alveolar. Aos 28 dias, no lado controle houve recobrimento do defeito por osso novo; no lado tratado houve reabsorção extensa da cortical vestibular e crista alveolar com perda dos limites do defeito. Apesar de a composição utilizada apresentar bom desempenho no Teste de histocompatibilidade, sua aplicação local em defeitos ósseos mandibulares ocasionou reabsorções intensas.Organic silicon (OS), glucosamine sulphate (GS), and methylsulfonylmethane (MSM) have been related to bone and connective tissue health and have been considered as basic therapy for osteoarthrosis disorders. The present study investigated Histocompatibility of these three components in association and its application in the regeneration in mandibular bone defects in rats. Nine rats were used for Histocompatibility Test. In each animal was implanted the composition (70% OS + 15% GS + 15% MSM) and gutta percha (control) under the dorsal subcutaneous tissue. The samples were collected in 7, 14 and 21 days post-surgery, processed for paraffin inclusion and staining in hematoxylin and eosin, and inflammatory events analyzed. The presence of granulation tissue, neutrophils, eosinophils, lymphocytes, macrophages and abscess was analyzed. For the Histocompatibility Test both groups had a moderate inflammatory process in seven days post-surgery and mild inflammatory process in 14 and 21 days, allowing its application in rats mandibular bone defects. Nine rats were used for local application Test. Were prepared a bone defect in the right and left mandible, under the crown of the lower first molar. The composition was engrafted in the right and bone defects without treatment (left) were the control group. The analyzes were performed after 7, 14 and 28 days post-surgery and samples were evaluated by scanning electron microscopy (SEM). In the 7 day there were no significant presence of new bone in both samples. At 14 days, on the control side there was a decrease in the depth of the defect; on the treated side the defect presented irregular borders and wide resorption in the margins and in the cortical and alveolar bone crest. At 28 days, on the control side there was an advanced bone regeneration with no more depth in the cavity with newly formed bone; on the treated side there was extensive reabsorption of the cortical vestibular and alveolar crest with loss of the defect limits. In conclusion, although the composition had positive result in the Histocompatibility Test, its direct application in mandibular bone defects caused intense resorption

You cannot accurately predict a positive or negative FCXM result-not even when the sera have donor-specific antigens-unless you actually perform a crossmatch. Using fluorescence intensity as a surrogate for antibody concentration does not correlate quantitatively with the occurrence of a positive or negative crossmatch. Therefore, it is imperative to give each recipient a chance at being offered a donor organ by performance of a real-time crossmatch and not rely on a virtual evaluation.
