    Abstract:

    We have studied the effect of recombinant human IFN-beta on the basal and IFN-gamma-induced expression of adhesion molecules and class II MHC Antigens on human peripheral blood monocytes and on ICAM-1 (intercellular adhesion molecule-1) expression of a human umbilical vein endothelial cell line (EAhy 926). We show that IFN-beta downregulates both basal and IFN-gamma-induced expression of VLA-4 (very late activation Antigen-4) Antigen on monocytes, but has no effect on the expression of CD11a, CD11b, CD11c, L-selectin, CD18, ICAM-1, beta 1-integrin or CD44 on monocytes or ICAM-1 on EAhy 926 cells. We also show that IFN-beta antagonizes the IFN-gamma-induced expression of HLA-DQ-Antigen, but not HLA-DR or HLA-DP Antigens on monocyte surface. These findings may partially explain the beneficial effect of IFN-beta in multiple sclerosis, since VLA-4-Antigen is critical for leukocyte recruitment into inflamed brain and downregulation of HLA-class II expression diminishes Antigen presenting capacity of monocytes.

    Since cyclosporin A (CsA), a widely used immunosuppressive drug, strongly suppresses interleukin-2 (IL-2) secretion, it is frequently difficult to estimate T lymphocyte activation in early acute rejection. We found that, when evaluated based on HLA-DQ Antigen expression, monocyte activation in the peripheral blood of renal transplantation patients was a very sharp parameter in diagosing acute rejection. All of 16 episodes of early acute rejection, which were relatively easily suppressed by steroid pulse therapy, showed a sharp increase in the proportion of HLA-DQ Antigen-positive monocytes (DQ+mono) and a quick return of DQ+mono to previous values, along with a fall in serum creatinine levels. Since, however, HLA-DR Antigen-positive T lymphocytes (DR+T) were markedly increased over a long period in episodes of therapy-resistant and chronic rejection, their prolonged high value was regarded as a parameter indicative of poor prognosis.

    OBJECTIVE:Epstein-Barr virus infection and the expression of HLA-DQ Antigen and p53 protein in non-Hodgkin's lymphoma(NHL) and the correlations were studied.METHODS: LMP-1 and the expression of HLA-DQ Antigen and p53 protein in 160 cases of the paraffin embodied tissues of NHL including different pathological types were detected using streptavidin-peroxidase (SP) immunohistochemistry.RESULTS: The detectable rates of EB virus in B-NHL and T-NHL were 28.9%(39/135) and 16.0%(4/25) respectively. The positive rate of HLA-DQ Antigen expression in B-NHL and T-NHL were 25.2%(4/135) and 28.0%( 7/25) respectively. There was a positive correlation between the extent of EB virus infection and the grade of HLA-DQ expression. The positive rate of p53 protein expression in B-NHL and T-NHL were 48.9%(66/135) and 32.0%(8/25) respectively.CONCLUSIONS: The latent infection of EB virus may pay an important role in the pathogenesis of NHL. There was probably an inherent relation and influence each other between EB virus and HLA-DQ. The Antigen expression of HLA-DQ may increase the susceptibility of EB virus entry. However, there was not a relation between EB virus and p53 in NHL.
