    Abstract:

    To investigate the effects of active immunization of cyclic beef heifers with different doses of a human serum albumin-Cys-Gly-GnRH (HSA-GnRH) conjugate on Antibody titers, ovarian function, body growth, and carcass characteristics, 32 heifers (BW = 477 ± 7.1 kg ; mean ± SE) were assigned to one of four immunization treatments :.1 mg of HSA or.01,.1, or 1.0 mg of HSA-GnRH, respectively. All heifers received a primary (d 0) and booster (d 28) immunization using DEAE-dextran as adjuvant. The duration of the experiment was 158 d. Overall Antibody titers against GnRH were greater (P 21 d) were 1/8, 8/8, 7/8, and 8/8, respectively. The interval from primary immunization to anestrus (40.7 ± 6 d) and the duration of anestrus (78 ± 7 d) were not affected by dose of HSA-GnRH conjugate. The number of ovulations detected was reduced (P .10) of treatment on overall growth rate (ADG.54 ±.041 and.51 ±.054 kg for control and GnRH-immunized heifers, respectively) or carcass measurements. However, ADG was reduced for GnRH-immunized heifers between d 56 and 84. In , immunization of heifers with a HSA-GnRH conjugate invoked an immune response in all heifers and induced anestrus in 23/24 heifers for 82.8 ± 6.9 d ; dose of HSA-GnRH conjugate did not affect the incidence or duration of anestrus.

    Abstract:

    Abstract A novel electrochemical immunosensor based on carbon paste electrode (CPE) composed of ionic liquid (IL) and graphite was constructed. It demonstrated good efficiency for quick (each test in 30 s) determination of thyroid stimulating Hormone (TSH). Electrode surface was modified by gold nanoparticles in order to immobilize of the thyroid stimulating Hormone Antibody (anti-TSH) on the CPE. The immunoassay structure was established by sandwiching the antigen (TSH) between the thyroid stimulating Hormone Antibody on the CPE surface modified with gold nanoparticles and the secondary Antibody, polyclonal anti-human-TSH labeled with horseradish peroxidase (HRP-labeled anti-TSH). The signal of differential pulse voltammetry (DPV) was used as a basis for the determination of TSH concentration. This signal is generated by the reaction between O-aminophenol (OAP) and H2O2 catalyzed by HRP. The proposed immunosensor is able to measure the concentration of TSH in a linear range between 0.2 and 90.0 ng/mL with a detection limit 0.1 ± 0.02 ng/mL. In addition, high sensitivity and acceptable stability were achieved by this immunosensor which is promising in the clinical assay of TSH.

    Abstract Objective: To determine whether treatment with recombinant human growth Hormone (rhGH) enhances growth rate in growth-retarded children with chronic renal failure. Design: One hundred twenty-five prepubertal growth-retarded children with chronic renal failure were randomly assigned to receive either rhGH (n = 82) or placebo (n = 43) for 2 years. Setting: The study was undertaken at 17 pediatric nephrology centers in the United States. Measurements: Growth rate, standardized height, bone age, fasting and 2-hour postprandial glucose and insulin levels, biochemical values, and insulin-like growth factor I and anti-growth Hormone Antibody levels were evaluated serially during the 2-year study period. Results: Standardized height increased from −2.94 to −1.55 in the rhGH group after 24 months of treatment, and decreased from −2.82 to −2.91 in the placebo group ( p p p = 0.63). Mean fasting and postprandial insulin values were elevated at 12 months but not at 24 months in the rhGH-treated patients. Mean fasting and 2-hour postprandial glucose values at 24 months were not significantly elevated over baseline values in either group. Conclusions: Growth rate and standardized height were significantly increased in growth-retarded prepubertal children with chronic renal failure when rhGH was used. The acceleration in growth was not associated with undue advancement of bone age. No clinically significant side effects were associated with rhGH treatment. The use of rhGH in growth-retarded children with chronic renal failure may facilitate the achievement of the inherent growth potential of such children. (J P ediatr 1994;124:374-82)
