The objective of this study was to investigate the hypolipidemic properties of chrysin on Triton WR-1339-induced hyperlipidemia in female C57BL/6 mice. Triton WR-1339 was administered intraperitoneally (400 mg/kg) to overnight-fasted mice to develop acute hyperlipidemia. Chrysin was administered orally (10 mg/kg) 30 min before Triton WR-1339. At 24 h after Triton WR-1339 injection, blood samples were collected to measure plasma lipid levels. The hepatic thiobarbituric acid reactive substances (TBARS), carbonyl content, non-protein sulfhydryl (NPSH) and ascorbic acid (AA) levels, as well as catalase (CAT) and superoxide dismutase (SOD) activity were recorded. Chrysin administration significantly decreased total cholesterol levels. In addition, it partially decreased non-high density lipoprotein-cholesterol and triglycerides levels in plasma of hyperlipidaemic mice. In addition chrysin administration prevented the increase on TBARS levels and prevented the decrease in SOD activity induced by Triton WR-1339. These findings indicated that chrysin was able to decrease plasma lipids concentration and that its antioxidant properties was, at least in part, involved in the hypolipidaemic action of chrysin.

Food Chemistry, 2008Co-Authors: Hicham Harnafi, Hana Serghini Caid, Nour El Houda Bouanani, Mohammed Aziz, Souliman AmraniAbstract:Abstract The hypocholesterolemic and hypotriglyceridemic activities of the aqueous and organic extracts of Ocimum basilicum were studied using Triton WR-1339-induced hyperlipemic mice as an experimental model. Hyperlipidemia was developed by intraperitoneal injection of Triton (200 mg/kg body weight “BW”). The animals were divided into eight groups of eight mice each: normolipidemic control group (NCG), Hyperlipidemic control group (HCG), Hyperlipidemic plus DMSO control (HDCG), crude aqueous basil extract-treated group (CETG), dichloromethane extract-treated group (DETG), ethyl acetate extract-treated group (EETG), methanol extract-treated group (METG), and aqueous fraction-treated group (AFTG). After 7 h and 24 h of treatment, the intragastric administration of all extracts caused a significant decrease of plasma total cholesterol. Triglyceride levels were also significantly lowered but not in DETG. Similar results were observed for LDL-cholesterol concentrations. Although no significant change of HDL-cholesterol was noticed after 7 h of treatments, a significant increase of this cholesterol fraction was observed in EETG and AFTG after 24 h. Furthermore, crude aqueous basil extract and all polar solvent (methanol, ethyl acetate, water)-soluble fractions showed a significant ameliorative action on elevated atherogenic index (AI) and LDL/HDL-C ratios, while these atherogenic markers were not statistically suppressed by the dichloromethane-soluble extract. This finding indicates that O. basilicum may contain polar products able to lower plasma lipid concentrations and might be beneficial in treatment of hyperlipidemia and atherosclerosis.

TWEAK binds and activates the Fn14 receptor, and may regulate apoptosis, inflammation, and angiogenesis, in different pathological conditions. We have evaluated the effect of exogenous TWEAK administration as well as the role of endogenous TWEAK on proinflammatory cytokine expression and vascular and renal injury severity in Hyperlipidemic ApoE-knockout mice. Methods and Results— ApoE −/− mice were fed with Hyperlipidemic diet for 4 to 10 weeks, then randomized and treated with saline (controls), TWEAK (10 μg/kg/d), anti-TWEAK neutralizing mAb (1000 μg/kg/d), TWEAK plus anti-TWEAK antibody (10 μg TWEAK +1000 μg anti-TWEAK/kg/d), or nonspecific IgG (1000 μg/kg/d) daily for 9 days. In ApoE −/− mice, exogenous TWEAK administration in ApoE −/− mice induced activation of NF-κB, a key transcription factor implicated in the regulation of the inflammatory response, in vascular and renal lesions. Furthermore, TWEAK treatment increased chemokine expression (RANTES and MCP-1), as well as macrophage infiltration in atherosclerotic plaques and renal lesions. These effects were associated with exacerbation of vascular and renal damage. Conversely, treatment of ApoE −/− mice with an anti-TWEAK blocking mAb decreased NF-κB activation, proinflammatory cytokine expression, macrophage infiltration, and vascular and renal injury severity, indicating a pathological role for endogenous TWEAK. Finally, in murine vascular smooth muscle cells or tubular cells, either ox-LDL or TWEAK treatment increased expression and secretion of both RANTES and MCP-1. Furthermore, ox-LDL and TWEAK synergized for induction of MCP-1 and RANTES expression and secretion. Conclusion— Our results suggest that TWEAK exacerbates the inflammatory response associated with a high lipid–rich diet. TWEAK may be a novel therapeutic target to prevent vascular and renal damage associated with hyperlipidemia.
