    Abstract:

    The blood glucose lowering effect of Urtica dioica (Stinging Nettle) as a medicinal plant has been noted in old writings such as those of Avicenna. Recently, there has also been other investigators that indicated the hypoglycemic effect of Urtica dioica. But so far, the mechanism of this effect has not been deduced. In this report, a perifusion system is arranged in which an exact number of Langerhans Islets were exposed to several fractions of extracts of Urtica dioica by TLC. The active ingredient fraction named F(1), caused a marked increase in Insulin secretion. A simultaneous assay of glucose showed that the increase in Insulin Level was associated with a decrease in glucose Level. Furthermore, the active component of Urtica dioica was found to increase the Insulin content of blood sera in normal and streptozotocin diabetic rats that were injected intraperitoneally (i.p.) with the active ingredient of the extract. The in vivo studies presented in this report show that not only an increase in Insulin Level of blood sera was observed in rats after 30 min from the initial point of injection but a simultaneous decrease of blood sugar was detected when similar sera was tested for glucose. The increase in Insulin Level was six times during the 120 min of our determination. The decrease in blood sugar was found to be similar both in the Level and time of initiation. On the basis of our findings, we assume that F(1) is the active ingredient of plant leaves extract. The results show that the blood lowering effect of the extract was due to the enhancement of Insulin secretion by Langerhance Isletes.

    Abstract Background Insulin resistance, often associated with obesity, is hypothesized to be involved in the pathogenesis of essential hypertension and may relate to increased left ventricular mass (LVM). Methods We examined correlations between echocardiographic LVM and fasting blood glucose and Insulin Levels in a cross-section of 216 black and white healthy children and young adults aged 13 to 27 years in Bogalusa, Louisiana. Anthropometric measurements and blood pressure readings were also obtained. Results Positive bivariate correlation was found between fasting blood glucose Level and LVM corrected for growth (LVMC) (LVMC = LVM/Height 2.7 ) with all race/sex groups combined ( r = 0.17, P ≤ .03). Multivariate analyses in a model including race, sex, age, and measures of body size showed no significant correlations between fasting blood glucose Level, Insulin Level, and LVMC. However, when patients were ranked in terciles by fasting Insulin Level and within each tercile by subscapular skinfold thickness or weight tercile, increasing LVMC with increasing Insulin Level was found in the highest subscapular skinfold thickness and weight terciles. The largest difference was between high and low Insulin groups ( P ≤ .03). When grouped by systolic blood pressure tercile, there was no difference in LVMC with increasing fasting Insulin tercile. We concluded that in heavier individuals, increased Insulin Levels may be a risk factor for the accumulation of increased LVMC independent of any relation among Insulin, obesity, and blood pressure.     Abstract:

    Objective: To assess whether circulating Insulin is a major contributor to adverse lipid profiles during the transition from adolescence to young adulthood. Methods: The association between fasting Insulin Levels and serum lipid and lipoprotein Levels was examined in a cross-sectional survey of 4136 young individuals aged 5 to 30 years from a biracial community. Results: Fasting Insulin Levels were strongly and positively correlated with serum triglyceride and very-low-density lipoprotein cholesterol Levels and negatively correlated with high-density lipoprotein cholesterol Levels in all age groups (5 to 11, 12 to 17, 19 to 24, and 25 to 30 years). An increasing impact of Insulin Level on low-density lipoprotein cholesterol Level was observed in young adults aged 25 to 30 years. In multivariate analysis, fasting Insulin Level was associated with very-low-density lipoprotein cholesterol Level for most of the age groups in both races independently of age, sex, glucose Levels, obesity, cigarette smoking, and alcohol intake. The independent relationship to low-density lipoprotein cholesterol Level persisted in young adults aged 25 to 30 years. The independent and negative association with high-density lipoprotein cholesterol Level remained in whites aged 5 to 24 years and blacks aged 19 to 24 years. When individuals were divided into tertiles according to Insulin concentration and subscapular skinfold thickness, the independent effect of Insulin Level and obesity on lipoprotein fractions was also noted. Furthermore, a stronger association of Insulin Level with lipoprotein fractions was observed in obese than in lean white males. Conclusions: These data indicate that an increasing association of Insulin Levels with adverse lipoprotein Levels in young adults, especially obese individuals, may have adverse consequence for adult cardiovascular diseases.     Abstract Objective: To determine whether body fatness, aerobic capacity, and fat distribution are associated with Levels of cardiovascular risk factors in children. Methods: Subjects (N = 57) were 7 to 11 years of age; their percentage of body fat ranged from 10% to 58%. The percentage of fat was measured with dual-energy x-ray absorptiometry, maximal aerobic capacity was measured on a tread-mill, fat distribution was expressed as the waist/hip circumference ratio, and blood pressures were measured with an automated monitor. Measurements of several lipoproteins and apoproteins were combined into one atherogenic index. Results: The percentage of fat was related to the atherogenic index (ρ = 0.38; p >0.01) and Insulin Level (ρ = 0.78; p >0.001). Aerobic capacity was inversely related to the atherogenic index (ρ = -0.27; p >0.05) and Insulin Level (ρ = -0.72; p >0.001). The waist/hip ratio was not related to the risk factors, and blood pressures were not related to fatness or aerobic capacity. In multiple regression analyses, only fatness explained significant independent proportions of the variance in the atherogenic index and Insulin Level. After control for fatness, the black children had higher Insulin Levels ( p >0.05). Conclusion: At early ages (7- to 11-year-old children), fatness was related to risk factors for cardiovascular disease and diabetes. (J PEDIATR 1994;125:847-52)
