Susceptibility of Mycobacterium immunogenum and Pseudomonas fluorescens to Formaldehyde and Non-Formaldehyde Biocides in Semi-Synthetic Metalworking Fluids

This study investigated the biocidal efficacy of the frequently applied commercial formaldehyde-releasing (HCHO) biocides Grotan and Bioban CS 1135 and non-HCHO type biocides Kathon 886 MW (isothiazolone) and Preventol CMK 40 (phenolic) toward this emerging mycobacterial species (M. immunogenum) in HP-linked MWFs, alone and in presence of a representative of the Gram-negative bacterial contaminants, Pseudomonas fluorescens, using two semi-synthetic MWF matrices (designated Fluid A and Fluid B). Relative biocide susceptibility analysis indicated M immunogenum to be comparatively more resistant (2–1600 fold) than P. fluorescens to the tested biocides under the varied test conditions. In terms of minimum inhibitory concentration, Kathon was the most effective biocide against M. immunogenum. Fluid factors had a major effect on the biocide susceptibility. Fluid A formulation provided greater protective advantage to the test organisms than Fluid B. Fluid dialysis (Fluid A) led to an increased biocidal efficacy of Grotan, Kathon and Preventol against M. immunogenum further implying the role of native fluid components. Used fluid matrix, in general, increased the resistance of the two test organisms against the biocides, with certain exceptions. M. immunogenum resistance increased in presence of the co-contaminant P. fluorescens. Collectively, the results show a multifactorial nature of the biocide susceptibility of MWF-colonizing mycobacteria and highlight the importance of more rigorous efficacy testing and validation of biocides prior to and during their application in metalworking fluid operations.

Differential biocide susceptibility of the multiple genotypes of Mycobacterium immunogenum

Khan, Jagjit S YadavAbstract:The non-tuberculous mycobacterium Mycobacterium immunogenum colonizes industrial metalworking fluids (MWFs) presumably due to its relative resistance to the currently practiced biocides and has been implicated in occupational respiratory hazards, particularly hypersensitivity pneumonitis. With an aim to understand its inherent biocide susceptibility profile and survival potential in MWF, five different genotypes of this organism, including a reference genotype (700506) and four novel test genotypes (MJY-3, MJY-4, MJY-10 and MJY-12) isolated in our recent study from diverse MWF operations were evaluated. For this, two commercial biocide formulations, Grotan (Hexahydro-1,3,5-tris(2-hydroxyethyl)- s -triazine) and Kathon (5-chloro-2-methyl-4-isothiazolin-3-one) currently practiced for the control of microorganisms, including mycobacteria, in MWF operations were tested. Effect of the fluid matrix on the biocide susceptibility was investigated for the synthetic (S) and semi-synthetic (SS) MWF matrices. In general, the minimum inhibitory concentration values were higher for the HCHO-releasing biocide Grotan than the isothiazolone biocide Kathon. All genotypes (except the reference genotype) showed lower susceptibility in SS as compared to S fluid matrix for Grotan. However, in case of Kathon, a greater susceptibility was observed in SS fluid for majority of the test genotypes (MJY-3, 4 and 10). The test genotypes were more resistant than the reference genotype to either biocide in both fluid types. Furthermore, the individual genotypes showed differential biocidal susceptibility, with MJY-10 being the most resistant. These observations emphasize the importance of using the resistant genotypes of M. immunogenum as the test strains for formulation or development and evaluation of existing and novel biocides, for industrial applications.

The results indicated that M. immunogenum is more resistant than P. fluorescens to the tested formaldehyde-releasing biocides (Grotan and Bioban), isothiazolone (Kathon), and phenolic biocide (Preventol). Kathon was effective against mycobacteria at lower concentrations than the other three test biocides in MWF. In general, there was a marked increase in biocidal resistance of both the test organisms when present in MWF matrix compared to saline. Increased resistance of the two test organisms to biocides was observed when they were in a mixed suspension (1:1 ratio). The results indicate the protective effect of the MWF matrix against the action of commonly used biocides on the MWF-colonizing microbial species of occupational health significance, including mycobacteria.

toxicity evaluation of single and mixed antifouling biocides measured with acute toxicity bioassays
Analytica Chimica Acta, 2002Co-Authors: Amadeo R Fernandezalba, M D Hernando, Luis Piedra, Yusuf ChistiAbstract:Antifouling biocides used in boat paints were analyzed with a battery of toxicity bioassays to evaluate the toxic effects of these compounds on Vibrio fischeri, Daphnia magnaand Selenastrum capricornotum.The antifoulants tested were Irgarol 1051, Kathon 5287, chlorothalonil, diuron, dichlofluanid, 2-thiocyanomethylthiobenzothiazole (TCMTB) and tributyltin (TBT). In most cases, the sensitivity of the organisms towards the toxicants followed the order: S. capricornotum> D. magna> V. fischeri. Toxicity by concentration level had the following order: TBT = Kathon 5287 > chlorothalonil > Irgarol 1051 > diuron > dichlofluanid > TCMTB for S. capricornotum .F orD. magna(48 h test), the toxicity order of compounds was TBT > Kathon 5287 > chlorothalonil > TCMTB > dichlofluanid > Irgarol 1051 > diuron. For V. fischeri (30 min test), the compound toxicity had the following order: Kathon 5287 > TBT > TCMTB > dichlofluanid > Irgarol 1051 > chlorothalonil. Degradation products of Irgarol 1051 and diuron were also tested. Degradation product of Irgarol 1051 was found to be less toxic to the crustacean and the microalga but more toxic to the bacterium. Degradation products of diuron were less toxic to the microalga in comparison with the bacterium. For mixtures of compound, toxicities were additive in only 33% of the cases and 21% of mixtures were less toxic than expected based on the sum of concentrations of toxicants (antagonistic effect). Synergistic enhancements of toxicity were observed for a majority (46%) of the mixtures. The average reproducibility of the EC50 and LOEC measurements was 27, 24 and 28%, respectively, in the V. fischeri, S. capricornotum and D. magna bioassays. For single compound, the reproducibility of EC50 was better than ±20% for a vast majority of the measurements with the V. fischeri system, thus agreeing closely with the reported reproducibility values for this relatively well-known assay. © 2002 Elsevier Science B.V. Determination of selected antifouling booster biocides by high-performance liquid chromatography–atmospheric pressure chemical ionisation mass spectrometry

ThomasAbstract:A simple and rapid technique is described for the quantitative determination of four antifouling booster biocides [diuron, 4,5-dichloro-2-n-octyl-4-isothazolin-3-one (Kathon 5287), (2-thiocyanomethylthio)benzothiazole (TCMTB) and (2,3,5,6-tetrachloro-4-methylsulphonyl) (TCMS pyridine) in aqueous samples. The analytes were extracted with high recoveries (ca. 100±∼15%) from 2.7-l water samples, using C18 solid-phase extraction. Sample extracts were quantitatively analysed by reversed-phase HPLC and polarity switching atmospheric pressure chemical ionisation (APCI) MS using selective ion monitoring. Limits of detection for the four compounds were: diuron, 1 ng/l, Kathon 5287, 1 ng/l, TCMTB, 1 ng/l and TCMS pyridine, 5 ng/l. Analysis of samples collected from various UK marinas showed detectable concentrations of diuron to be present, however, concentrations of other three booster biocides were below their respective limit of detection.
