  • Effects of arginine enriched TPN and standard TPN on the immune function of patients with Carcinoma

    Abstract:

    Objectives:To investigate the effects of arginine(Arg) enriched TPN and standard TPN on the funtion of immune system and intestional barrier of patients with Large Intestine Carcinoma. Methods: 20 cases of patients were divided into standard TPN group( n =10) and TPN+Arg group( n =10).They received standard TPN or TPN+Arg for 5 days respectively.The phenotype(CD3,CD4,CD8) of T lymphocyte in peripheral blood and proper lamina of small Intestine was assayed using SABC immunocyte chemistry method.The IgG,IgM,IgA in peripheral blood and the height and density of small Intestine villi were also determined. Results:In standard TPN group, as compared with the TPN+Arg groop,the CD4 positive cells were decreased,the CD8 positive cells were increased,the CD4/CD8 ratio was decreased,the production of IgG,IgM,IgA was low in peripherial blood,and the height and density of small Intestine villi were decreased in proper lamina .There were significantly different( P 0.05 or P 0.01) between the two groups. Conclusions:Standard TPN may weaken the bodys immune function and intestinal barrier.Arginine may protect the body against immune depression caused by TPN.

    Objective Construction of bacteriophage antibody library Large Intestine cancer,and combination of protomics technology to screen human of cancer.Methods Separates cancer cell's proteins with the 2D gel electrophoresis and transfers to PVDF membranes.2D gels were analysis by Melanie 11 2-DE software,the specific antigen were selected by bacteriophage antibody library throughout 3 rounds.Results ELISA test revealed that the 8 specific antigen were detected in tumor tissue and not in normal tissue.Conclusion Successfullyconstruct the human antibody library,and accord with the proteomics technology to screen and determine specific antigen.

Methods Prepare total RNA of patient of Large Intestine cancer serum, RT-PCR of weight chain Fd and light chain kcDNA, the PCR product were inserted into carrier pComb3, electransterred into E. coli XL1-Blu, added bacteriophage VCSM13. Separates cancer cell's proteins with the 2D gel electrophoresis and transfers to PVDF membranes. 2D gels were analysis by Melanie 11 2-DE software, the specific antigen were selected by bacteriophage antibody library throughout 3 rounds. Results Weight chain Fd and light chain mRNA were amplified, ELISA test revealed that the 8 specific antigen were detected in tumor tissue and not in normal tissue. Conclusion Successfully construct the human antibody library, and the Fab of lymph gland, and used the proteomics technology to screen and determine specific antigen.

    Abstract:

    Objective:To evaluate the effects of protein kinase C(PKC)on Large Intestine Carcinoma cells(HT-29)cytoskeleton by morphological study.Methods:Cytoskeleton components,such as microfilaments,tubulin,vinculin changes of Large Intestine Carcinoma HT-29 cells by using PKC activator PMA and inhibitor SP were displayed by coomassie staining and indirect immunofluorescence staining,in order to study the effects of PKC on HT-29 cells cytoskeleton.Results:Abundant cytoskeleton components of HT-29 cell,such as microfilaments,microtubules and the focal contact,were observed,but skeleton fibres were short,arrangement was in disorder and not polarized.PMA could induce the cytoskeleton components of HT-29 cell to be polarized and to be decreased,and the invasive structures of lamellae and lamellipodia were also observed.However,SP could inhibit the effects induced by PMA,and induce the cytoskeleton components of HT-29 cell to be increased markedly and to be arranged trimly,and to form the intact cytoskeleton framework.Conclusion:Activation of PKC by treatment with PMA causes depolymerization and reorganization of microfilaments,microtubules and focal adhesions.SP can inhibit the effects of PMA,and not further causes depolymerization and reorganization of microfilaments,microtubules and focal adhesions.We propose that HT-29 cells invasion and metastasis adjuested by PKC may involve the mechanism of effecting cytoskeleton with PKC.

    Abstract:

    Objective:To study the influence of arsenic trioxide (As_2O_3) injection combined with/or fiuorouracil (5-FU) on the growth and telomerase activity of human Large Intestine Carcinoma cell lines.Methods:The influence of growth on human Large Intestine Carcinoma cell lines LoVo and CoLo-320 induced by As_2O_3 injection combined with/or 5-FU was analysed by 3-(4,5-dimethytthiazole-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT) assay. Telomerase activity of the Carcinoma cells was measured by the method of Repeat Amplitcation Protocol (TRAP) combining with Polyacrylamide Gel Electrophosis (PAGE) Silver Staining method before and after treatment of As_2O_3 injection combined with/or 5-FU.Results:As_2O_3 injection combined with 5-FU may obviously improve the anti-tumor effeet on human Large Intestine Carcinoma, and may strengthen inhibiting the tolemerase activity comparing each individual drug.Conclusion:As_2O_3 injection combined with 5-FU may obviously improve the anti-tumor effect on human Large Intestine Carcinoma, and whose mechanism maybe related to its strengthen inhibiting the tolemerase activity.
