It has been exploited for economical purposes, as cattle alternative in European and South American countries. In Brazil, the State of Rio Grande do Sul is outstanding, in rhea rearing and it is in the process of implantation in the Country Northeasty Region. This work aims to describe the morphology of the blood cells in rheas. In this work ten rheas were used, regardless age and sex. Two ml of peripheral blood were collected by puncture of the brachial vein with disposable syringe. The samples here partially used to make extensions with Leishman Stain. Seven types of nucleate cells have been observed through morphologic analysis on the light microscope. The erythrocyte revealed an elliptical form, with condensed nucleus of elliptical form; acidophilic cytoplasm. The thrombocyte revealed an elliptical form, with nucleus located in one of the polar regions; pale cytoplasm. As to the round-shaped leukocytes, within the granulocytes, the heterophils presented excentric, condensed, and lobulated nucleus; cytoplasm rich in fusiform salmon-colored granules. The eosinophils distinguish from the heterophils due to the round eosinophilic granules. The basophils stand out from the other granulocytes due to its large and central nucleus with round specific cytoplasmic and highly basophilic granules. Within the agranulocytes, the monocytes presented reniform nucleus, which is frequently central, with slack chromatin, with small areas of condensation; cytoplasm lightly basophilic and with vacuoles. The lymphocytes presented varies forms and sizes; large nucleus with slack chromatin with some nucleoli; scarce and basophilic cytoplasm. The cells of the peripheral blood of Rhea americana present on the light microscope morphology similar to the other birds which have already been studied.

No Brasil, destaca-se o Rio Grande do Sul e, em fase inicial, o Nordeste. O presente estudo objetivou descrever a morfologia das células sangüíneas em ema. Neste trabalho foram utilizados dez exemplares, desconsiderando-se idade e sexo. Foram colhidos 3mL de sangue periférico por punção da veia braquial, com seringa descartável. As amostras foram utilizadas, em parte, na confecção de extensões coradas com Leishman. Feita a análise morfológica ao Microscópio de Luz, foram observados sete tipos celulares nucleados. O eritrócito mostrou-se elíptico, com núcleo geralmente condensado, de forma elíptica; citoplasma acidófilo. O trombócito apresentou-se elíptico, com núcleo localizado em um dos pólos; citoplasma pálido. Quanto aos leucócitos, de forma arredondada, entre os granulócitos os heterófilos apresentaram-se com núcleo excêntrico, condensado, lobulado; citoplasma rico em grânulos fusiformes de coloração salmão. Os eosinófilos distinguem-se dos heterófilos pelos grânulos arredondados eosinofílicos. Os basófilos destacam-se dos outros granulócitos pelo núcleo grande e central, com grânulos citoplasmáticos específicos arredondados e fortemente basofílicos. Entre os agranulócitos, os monócitos mostraram núcleo rineforme, freqüentemente central, de cromatina frouxa, com pequenas áreas de condensação; citoplasma levemente basofílico e com vacúolos. Os linfócitos apresentaram-se variados em forma e tamanho; núcleo grande com cromatina frouxa, com alguns nucléolos; citoplasma escasso e basofílico. Although Giemsa Staining is most commonly used, the Leishman Staining method provides better visualization of the nuclear chromatin pattern of cells. It is less well known whether accuracy of parasitaemia assessment is equally accurate with the latter method. Peripheral blood thin and thick smears from consecutive febrile patients admitted to Ispat General hospital, Rourkela, Odhisa, India, were Stained with Giemsa and Leishman Stain. Methods were compared for species identification, parasite quantification, and ability for identification of alternative diagnoses. Blood films from 1,180 fever patients were compared according to Staining method, of which 111 were identified as parasitaemic using Giemsa and 110 with Leishman Staining. The Kappa value as a measure of agreement between methods was 0.995 (p < 0.001), and the log10parasitaemia between methods were strongly correlated (r2 = 0.9981). In parasite negative patients, thin smear assessment contributed to making a diagnosis in 276/1,180 (23%) of cases. These assessments were better made in Leishman-Stained preparations, especially for the assessment of morphological changes in red and white cells. Leishman’s Staining method for thin and thick smears is a good alternative to Giemsa’s Stain for identifying Plasmodium parasites. The Leishman method is superior for visualization of red and white blood cell morphology.

Blood was collected with heparinized syringes, and blood smears were prepared and Stained with Leishman Stain. Slides were examined and photographed using a Leica light microscope. Total white blood cell counts and differential counts of thrombocytes and leukocytes were obtained for statistical analysis. The group exposed to water samples from Lago Azul exhibited an increase in the number of leukocytes and in the total number of white blood cells, suggesting that the chemical contaminants in this environment were acting similarly to antigens in this fish species, causing the proliferation of defense cells. In the group exposed to detergent during 20 days, the number of thrombocytes decreased. These results suggest that the variations in the number of leukocytes were indicators of environmental pollution and that biodegradable detergents may, after a certain time of exposure, affect vital functions in fish, such as coagulation and prevention of infections, which directly involves thrombocytes. a comparative study between conventional and modified Leishman Stain
International Journal of Research, 2021Co-Authors: Rizwana Abdul Hye, Bindiya Gisuthan, Indira KariveettilAbstract:Introduction: Leishman Stain has been used as the Stain of choice for peripheral blood films since many decades .But it has a disadvantage of consuming 15 minutes for the procedure alone thereby increasing the turn around time of peripheral smear reporting. In this study modified Leishman Stain was made by adding phenol to conventional Leishman to reduce the Staining time to 3 minutes without interfering with the quality of Stain. Aim: To study the quality of modified Leishman Stain in comparison with conventional preparation on peripheral blood smears. Materials and Methods: The present cross-sectional study was carried out in Central Haematology laboratory of a tertiary health care centre in Southern India. A pilot study was done to determine the ideal ratio of phenol to Leishman powder in preparing modified Stain without compromising the Staining quality. After obtaining consent, the blood sample was collected in anticoagulated vials. Sample size was calculated to be 85.Two thin wedge smears were prepared from each sample and one was Stained with conventional Leishman Stain and other with modified Leishman Stain. The Staining characteristics were assessed and scored by an experienced pathologist in terms of 6 parameters- RBC pattern, nuclear pattern, neutrophil granules, eosinophil granules, platelets and background Staining. The grades were 1, 2, 3 & 4 (poor, satisfied, good and excellent). Smears each with grades 2, 3 and 4 were selected for study. The results were analysed by Statistical Package for the Social Sciences (SPSS) software version 16.0. Result: The mean value for the Staining quality using conventional method was 4.73 and for modified method was 5.39, and the total score possible is 6. Photomicrograph showed excellent results with modified Leishman Stain. Thus from these values we can interpret that modified method gave much more acceptable results than that of conventional method. Conclusion: Unlike the conventional method which requires a total of 15 minutes, to complete the Staining process, modified Leishman Staining techniques takes only 3 minutes. Blood films can be Stained within a short period of time thus aiding in rapid diagnosis and treatment of patients.
