SAR and pharmacophore models for the rhodanine inhibitors of Plasmodium falciparum enoyl-acyl carrier protein reductase

Enoyl acyl carrier protein (ACP) reductase of Plasmodium falciparum (PfENR) is the rate determining enzyme of its elongation module. Hence, PfENR has been a target for the development of antimalarials as well as vaccines. Towards this endeavour, we had recently identified rhodanine class of compounds as inhibitors of PfENR. Here, we report a number of new inhibitors belonging to this class. These inhibitors have been divided into two broad subclasses: rhodanine-furans and rhodanine-phenyls. The inhibitory activity of all compounds was determined against purified PfENR. IC50 of these compounds were found to be in nanomolar to low-micromolar range. The structure–activity relationship of both the classes has been explored in detail for the first time. Separate 3D pharmacophore models for this enzyme have been generated for both rhodanine furans and phenyls. The pharmacophore model for rhodanine furan has a Hydrogen bond donor, two Hydrogen bond acceptors, two metal Ligators, three hydrophobic, and two aromatic ring features, whereas the pharmacophore model for the phenyl subclass has two hydrogen bond donors, two hydrogen bond acceptor, a metal Ligator, two hydrophobic, and two aromatic ring features. METHODS: Patients with rectal bleeding from grade II/III hemorrhoids had videoscopic anoscopy, which provided a magnified view, allowing accurate localization of the hemorrhoids and the dentate line before banding, and a photographic record, if required. Banding was performed using a suction Ligator that could be operated by one hand, allowing the other to control the anoscope. RESULTS: Of 39 patients with second- and third-degree hemorrhoids, 34 (87%) had no further bleeding after a single banding session and a further three had no recurrence after a second session. The only complications were pain (one patient) and infection (one patient). CONCLUSIONS: This method is convenient and effective, costing per procedure less than one-tenth of endoscope-mounted band Ligators. We recommend its use in preference if magnified views and a photographic record are required. However, its cost and complexity, compared with traditional hemorrhoid banding, may mean that the latter is preferred in the office setting.

Allowed and Forbidden d-d Transitions in Poly(3,5-dimethylpyrazolyl)methane Complexes of Nickel(II)†

A quantitative theoretical model involving coupled electronic states provides precise energies for the lowest-energy triplet and singlet excited states and allows compar-isons between complexes with a variable number of nitrogen and oxygen Ligator atoms. Singlet energies between 12 840 and 13 000 cm21 are determined for heteroleptic complexes. These energies are in an intermediate range between those for homoleptic complexes with either nitrogen or oxygen Ligator atoms with singlet states at approximately 12 000 and 14 000 cm21, respectively. The new theoretical approach is compared to the traditional ligand-field parameters obtained from the maxima of the broad, spin-allowed absorption bands
