    Abstract:

    We report the synthesis of three bioactive pyrene-bAsed fluorescent analogues of Ohmline which is the most efficient and selective inhibitor of SK3 ion channel. The interaction of these Ohmline-pyrene (OP1-3) with liposomes of different composition reveals that only OP2 and OP3 are readily integrated into liposomes. Fluorescence meAsurements indicate that, depending on their concentration, OP2 and OP3 exist either As monomer or As a mixture of monomer and excimers within the liposome bilayer. Among the three Ohmline Pyrene compounds (OP1-3) only OP2 is able to reduce SK3 currents and is the first efficient fluorescent modulator of SK3 channel As revealed by patch clamp meAsurements (- 71.3 ± 13.3% at 10 mM) and by its inhibition of SK3-dependent cancer cell migration at (32.5% ± 4.8% at 1 mM). We also report the first fluorescence study on living breAst cancer cells (MDA-MB-231) showing that OP2 is rapidly integrated in bio-membranes followed by cell internalization.

The originality of the new SK3 inhibitors comes from their amphiphilic nature and contrAst form others inhibitors that are peptides or heterocyclic compounds. We found that ether Lipids and ether phospho-glyco-Lipids are indeed efficient to modulate SK3 ion channel that consequently reduced cancer cell migration (in vitro Assays) and abolished the formation of bone metAstAses in a murine model of breAst cancer.

    Abstract The thermophilic cyanobacterium Thermosynechococcus elongatus wAs cultivated under controlled growth conditions using a new type of photobioreactor, allowing us to optimise growth conditions and the biomAss yield. A fAst large-scale purification method for monomeric and dimeric photosystem II (PSII) solubilized from thylakoid membranes of this cyanobacterium wAs developed using fAst protein liquid chromatography (FPLC). The obtained PSII core complexes (PSIIcc) were analysed for their pigment stoichiometry, photochemical and oxygen evolution activities, As well As Lipid and detergent composition. Thirty-six chlorophyll a (Chla), 2 pheophytin a (Pheoa), 9± 1 β-carotene (Car), 2.9±0.8 plAstoquinone 9 (PQ9) and 3.8±0.5 Mn were found per active centre. For the monomeric and dimeric PSIIcc, 18 and 20 Lipid As well As 145 and 220 detergent molecules were found in the detergent shell, respectively. The monomeric and dimeric complexes showed high oxygen evolution activity with 1/4 O2 releAsed per 37–38 Chla and flAsh in the best cAses. Crystals were obtained from dimeric PSIIcc by a micro-batch method. They diffract synchrotron X-rays to a maximum resolution of 2.9-A, resulting in complete data sets of 3.2 A resolution.

    The overall secretion of hepatic Lipid As VLDL may be regulated by (i) changing the number of particles secreted As a result of altering apolipoprotein B output and (ii) changing the degree of Lipidation of the particles with triacylglycerol so that their diameters vary over a threefold range. Substantial progress hAs been made in understanding the pathway of triacylglycerol incorporation into VLDL. Less is understood about the processes that commit apolipoprotein B either to secretion or to presecretory degradation, although both regulated translocation and an early Lipidation step of the nAscent particle with cholesterol or cholesterol ester have been implicated.
