Many researches showed that it is used for treatment of variety of diseases, for example as remedy for asthma and diabetes, duo to its antibacterial effects. The present study aimed to access the effect of the sampling location on the antibacterial activity of the flavonoids extracted from Marrubium Vulgare L. leaves, against three pathogenic bacteria; Escherichia coli, Bacillus cereus, and Proteus mirabilis. The leaves were collected from three different sampling locations belonging northwest Algeria: Tessala Mountain, M’sila forest and Ain Skhouna. The flavonoid extraction was carried out using organic solvents with increasing polarity. The bacterial susceptibility testing by diffusion agar method showed that the diameters zones varied significantly (P < 0.001) according to the sampling location of the leaves, the flavonoid extract and its concentration. The inhibition diameters varied between 7.5–34.3 mm, which often exceed those induced by the standard antibacterial agents (chloramphenicol, gentamicin, aztreonam, nalidixic acid, ceftazidime, and imipenem). The agar dilution method showed that the minimum inhibitory concentrations (MICs) range between 25 and 100 μg/mL; revealing strong antibacterial inhibition. The phytochemical screening revealed the presence of flavans and flavanols, which may be responsible of this promising antimicrobial activity.

Les diametres d’inhibition enregistres oscillent entre 8,16 et 37,5 mm enregistrant meme une inhibition totale de la croissance fongique et depassent souvent ceux induits par les antifongiques commercialises (Amphotericine, Fluconazole, Terbinafine et le nitrate d’econazole). Les concentrations minimales inhibitrices (CMI) obtenues varient entre 6,25 et 100 μg/mL enregistrant une forte inhibition antifongique. Le screening phytochimique a revele l’existence de certaines classes flavonoiques telles que les flavanes et les flavanols qui pourraient etre responsables de ce remarquable pouvoir antifongique. Conclusion La localite d’echantillonnage des feuilles de Marrubium Vulgare influence nettement l’activite antifongique des flavonoides. Ces derniers se sont averes de tres bons fongistatiques et qui meritent d’etre valorises en pharmacologie.

optimization of extraction conditions of some phenolic compounds from white horehound Marrubium Vulgare l leaves

Distilled water and different organic solvents such as: methanol, ethanol and acetone, were used, with various concentrations (20% - 80%, v/v), temperatures (20°C - 60°C) and extraction times (30 - 450 min). Results showed that the maximum total phenolics amounts (293.34 ± 14.60 mg gallic acid equivalent/g dry weigh), were obtained with 60% aqueous methanol at 25°C for 180 min; total flavonoids (79.52 ± 0.55 mg catechin equivalent/g dry weigh) with 80% aqueous methanol at 20°C for 450 min, and condensed tannins (28.15 ± 0.80 mg catechin equivalent/g dry weigh) with 60% aqueous acetone at 50°C and for 180 min. ANOVA test showed the significant effect (***P < 0.001) of the extraction conditions tested on phenolic compounds. The Principal Component Analysis (PCA) exhibited the positive effect of low temperatures on total phenolics and flavonoids extraction, and the effect of high temperatures on the condensed tannins extraction. The Response Surface Methodology (RSM) provided predicted values of extraction conditions and maximum polyphenols amounts similar to those obtained experimentally.

Ils presentent un remarquable effet antibacterien sur les bacteries Gram+ et une activite antifongique moderee. Les concentrations minimales inhibitrices (CMI) varient entre 6,25 et 50 μg/ml en fonction de l’extrait flavonoique et de la souche microbienne. Le screening phytochimique et la chromatographie sur couche mince (CCM) ont revele l’existence de certaines classes flavonoiques qui pourraient etre responsables du pouvoir antimicrobien de Marrubium Vulgare L.

Materiels et methodes Les feuilles ont ete prelevees de trois differentes localites appartenant au nord-ouest Algerien : mont de Tessala, foret domaniale de M'sila et Ain Skhouna. L’extraction des flavonoides a ete realisee en utilisant des solvants organiques a polarite croissante. Un screening phytochimique a ete realise par des tests de coloration en tubes. Les diametres d’inhibition ont ete mesures par la methode de diffusion en milieu solide. Les concentrations minimales inhibitrices ont ete determinees par methode de dilution en milieu solide. Resultats L’activite antifongique varie significativement (p < 0,001) en fonction de la localite d’echantillonnage des feuilles, de l’extrait flavonoique teste et de sa concentration, et du type de la souche fongique. Les diametres d’inhibition enregistres oscillent entre 8,16 et 37,5 mm enregistrant meme une inhibition totale de la croissance fongique et depassent souvent ceux induits par les antifongiques commercialises (Amphotericine, Fluconazole, Terbinafine et le nitrate d’econazole). Les concentrations minimales inhibitrices (CMI) obtenues varient entre 6,25 et 100 μg/mL enregistrant une forte inhibition antifongique. Le screening phytochimique a revele l’existence de certaines classes flavonoiques telles que les flavanes et les flavanols qui pourraient etre responsables de ce remarquable pouvoir antifongique. Conclusion La localite d’echantillonnage des feuilles de Marrubium Vulgare influence nettement l’activite antifongique des flavonoides. Ces derniers se sont averes de tres bons fongistatiques et qui meritent d’etre valorises en pharmacologie.

optimization of extraction conditions of some phenolic compounds from white horehound Marrubium Vulgare l leaves

Distilled water and different organic solvents such as: methanol, ethanol and acetone, were used, with various concentrations (20% - 80%, v/v), temperatures (20°C - 60°C) and extraction times (30 - 450 min). Results showed that the maximum total phenolics amounts (293.34 ± 14.60 mg gallic acid equivalent/g dry weigh), were obtained with 60% aqueous methanol at 25°C for 180 min; total flavonoids (79.52 ± 0.55 mg catechin equivalent/g dry weigh) with 80% aqueous methanol at 20°C for 450 min, and condensed tannins (28.15 ± 0.80 mg catechin equivalent/g dry weigh) with 60% aqueous acetone at 50°C and for 180 min. ANOVA test showed the significant effect (***P < 0.001) of the extraction conditions tested on phenolic compounds. The Principal Component Analysis (PCA) exhibited the positive effect of low temperatures on total phenolics and flavonoids extraction, and the effect of high temperatures on the condensed tannins extraction. The Response Surface Methodology (RSM) provided predicted values of extraction conditions and maximum polyphenols amounts similar to those obtained experimentally.

The present study aimed to access the effect of the sampling location on the antibacterial activity of the flavonoids extracted from Marrubium Vulgare L. leaves, against three pathogenic bacteria; Escherichia coli, Bacillus cereus, and Proteus mirabilis. The leaves were collected from three different sampling locations belonging northwest Algeria: Tessala Mountain, M’sila forest and Ain Skhouna. The flavonoid extraction was carried out using organic solvents with increasing polarity. The bacterial susceptibility testing by diffusion agar method showed that the diameters zones varied significantly (P < 0.001) according to the sampling location of the leaves, the flavonoid extract and its concentration. The inhibition diameters varied between 7.5–34.3 mm, which often exceed those induced by the standard antibacterial agents (chloramphenicol, gentamicin, aztreonam, nalidixic acid, ceftazidime, and imipenem). The agar dilution method showed that the minimum inhibitory concentrations (MICs) range between 25 and 100 μg/mL; revealing strong antibacterial inhibition. The phytochemical screening revealed the presence of flavans and flavanols, which may be responsible of this promising antimicrobial activity.
