    Abstract:

    OBJECTIVE To investigate potential reproductive effects of Pterocarpus Marsupium methanolic extract on testosterone propionate induced Polycystic Ovarian Syndrome (PCOS) in female albino rats. METHODOLOGY PCOS was induced in female albino rats by daily injecting testosterone propionate for 15 days intraperitoneally. Animals are divided into five groups with six rats per group. Group 1: Control group received olive oil, Group 2: Testosterone propionate+natural recovery, Group 3: Testosterone propionate + a dose of clomiphene citrate (standard), Group 4 and 5: Testosterone propionate + low dose (200mg/kg) and high dose (400mg/kg) b.w respectively for 15 days. Various biochemical and histopathological investigations were assessed. RESULTS Methanol extract of Pterocarpus Marsupium was able to exert its protective effect successfully by restoring all the parameters to normal and diminishing the cysts found in ovaries. CONCLUSION Pterocarpus Marsupium showed potential reproductive effects on testosterone propionate induced PCOS female albino rats and could be used as an alternative therapy in the treatment of PCOS.

    Abstract:

    Pterocarpus Marsupium is widely used as ‘Rasayana’ in ayurvedic system for curing several medical ailments. In view of this, petroleum ether and chloroform extracts of Pterocarpus Marsupium bark was scrutinized to unfold free radical scavenging by measuring their capability for scavenging DPPH radical, ABTS, phosphomolybdenum assay as well as reducing power capacity assessment and anti- inflammatory activity by inhibition of protein denaturation. Additionally, RBC’S were also used to explore the potentiality of Pterocarpus Marsupium different extracts to shield RBC in oxidative stress condition. Chloroform extract showed an effective pharmacological activity in all assays when compared with petroleum ether extract and their respective standards. This reveals the use of Pterocarpus Marsupium as a potent source of antioxidant, anti-inflammatory, anti-haemolytic agent and would provide an ample opportunity for further investigation. Isolation of active compounds from this plant responsible for producing such bioactivities is under process.

    Pterocarpus Marsupium is a well known plant in ayurvedic system of medicine. The plant has been found to possess diverse number of biological activities and is thus commercially exploited. In the present study, methanol and aqueous extracts of P.Marsupium bark were used to evaluate in vitro antioxidant, anti-inflammatory and anti-haemolytic activities. In vitro antioxidant activity was carried out by DPPH, ABTS, phosphomolybdenum, reducing power assays, anti- inflammatory activity by inhibition of protein denaturation and anti-haemolytic by hyposaline induced haemolysis. Total phenolic and flavonoid content was also estimated. Methanol extract showed an effective pharmacological activity in all assays when compared with their respective standards. The maximum phenolic and flavonoid contents were found to be (280 ±0.47mg/g) and (620±0.81mg/g) respectively in methanol extract of Pterocarpus Marsupium. This study indicates the presence of active constituents which can be exploited for the treatment of various diseases and also could be used in pharmaceutical industry.

    The aim of the present study was to assess the anti-diabetic activity of Pterocarpus Marsupium Roxb. heartwood in alloxan induced diabetic rats using extracts obtained by optimized conventional and non conventional extraction methods. Aqueous and ethanol extracts of Pterocarpus Marsupium heartwood were prepared by conventional methods (infusion, decoction, maceration and percolation) and non conventional methods, such as ultrasound-assisted extraction (UAE) and microwave-assisted extraction (MAE). The crude aqueous extracts were administered orally to both normal and alloxan induced male albino rats (Sprague-Dawley strain). The experimental set up consisted of 48 male albino rats divided into 6 groups: Normal control, diabetic control (sterile normal saline, 1 ml/100 g body weight), standard (gliclazide, 25 mg/1000g of body weight), groups 4-6 (crude aqueous percolation, optimized UAE and MAE extract, 250 mg/1000g of body weight).     Abstract:

    The Pterocarpus Marsupium belong to family Fabaceae and is widely distributed in central, western and southern regions of India. The role of Pterocarpus Marsupium as anti-diabetic has been very well established. Its extract has been prepared using many methods like infusion, maceration, decoction and percolation. Several chemical constituents like pterostilbene, (-)-epicatechin, pterosupin, marsupsin, etc., have been identified and isolated. Pterocarpus Marsupium extract also shows promising results in cataract and hypertriglyceridaemia. This plant also finds its use as cardiotonic and hepatoprotective agent. Studies have also been reported to demonstrate its ability as a specific COX- 2 inhibitor. The present review explores its description, traditional uses, extraction methods, chemical constituents, pharmacological activity and commercial importance so that its potential as a multipurpose medicinal agent can be understood and appreciated.

    An aqueous extract of Pterocarpus Marsupium wood was screened for hypoglycemic activity on alloxan-induced diabetic rats. During both acute and sub-acute tests, the water extract, at an oral dose of 250 mg/kg, showed statistically significant hypoglycemic activity.

    Rising popularity of phytomedicines in various diseased conditions have strengthened the significance of plant-research and evaluation of phytoextracts in clinical manifestations. Pterocarpus Marsupium Roxb., a medicinal plant, known for its anti-oxidant and anti-diabetic activity is a rich source of phytochemicals with antihyperglycemic and antihyperlipidemic activities. However, its possible role in diabetic complications is not evaluated yet. The present study explores the possible role of alcoholic extract of heartwood of P. Marsupium in the treatment of long-term diabetic complications. The alcoholic extract of P. Marsupium was evaluated for advanced glycation-end-products formation, erythrocyte sorbitol accumulation and rat kidney aldose reductase enzyme inhibition at the concentration of 25-400 μg/ml using in-vitro bioassays. Also the phytoextract at the concentration of 10-320 μg/ml was evaluated for its antioxidant potential by in-vitro antioxidant assays which includes, determination of total phenol content; reducing power assay; nitric oxide scavenging activity; superoxide radical scavenging activity; total antioxidant capacity; total flavonoid content; DPPH scavenging activity; and hydrogen peroxide scavenging activity. The alcoholic extract of P. Marsupium across varying concentrations showed inhibitory effect as evident by IC50 on advanced glycation-end-products formation (55.39 μg/ml), sorbitol accumulation (151.00 μg/ml) and rat kidney aldose reductase (195.88 μg/ml). The phytoextract also exhibited high phenolic and flavonoid contents with promising antioxidant potential against the antioxidant assays evaluated. The present investigation suggests that the phytoextract showed prominent antioxidant, antiglycation property and, inhibited accumulation of sorbitol and ALR enzyme, thus promising a beneficial role in reducing/delaying diabetic complications.

    Context: Heartwood of Pterocarpus Marsupium has been widely reported for its effect on diabetes clinical or preclinically. However, role in diabetic complications is yet to be revealed. Aims: To investigate the effect of alcoholic extract of the heartwood of Pterocarpus Marsupiumin experimentally induced diabetic nephropathy in rats. Methods: The streptozotocin (STZ) 55 mg/kg, i.p., once daily induced diabetes in Sprague-Dawley rats. These animals were treated orally with alcoholic extract of P. Marsupium (100, 200 and 400 mg/kg) or glimepiride (10 mg/kg) for 60 days. Body weight, blood glucose, glycosylated hemoglobin (HBA1c), biochemical markers of renal function were estimated on day 30 and at the end of study (day 60). Kidney weight measurement, oxidative stress markers such as lipid peroxidation (TBARS), catalase, superoxide dismutase, reduced glutathione were estimated in kidney tissues, and histopathological evaluation was carried out at the end of study period. Results: The administration of an alcoholic extract of P. Marsupium showed a decrease in blood glucose, HBA1c, kidney weight, serum creatinine, blood urea nitrogen, serum uric acid, urea, urine volume, urine albumin and the level of TBARS. While the increase in urine creatinine, the activity of superoxide dismutase and glutathione were observed when compared to the diabetic control group. This effect was observed significantly at the highest dose of the plant extract. The histopathological study also confirmed that alcoholic extract prevented structural kidney damage. Conclusions: These results suggest that the alcoholic extract of P. Marsupium has renoprotective effects against STZ induced diabetic nephropathy

    Abstract:

    Degenerative changes in the histoarchitecture of kidneys of diabetic rats were observed. Methanolic extracts of Pterocarpus Marsupium barks suppressed the serum glucose and blood urea levels and normalized the body-kidney weight ratio. Treatment with these extracts also improved the kidney antioxidant contents with a decrease in the thiobarbituric acid-reactive oxygen species and normalized the kidney histoarchitecture.

    Pterocarpus Marsupium Roxb. (Leguminosae) is widely used in Ayurvedic medicine as „Rasayana. for management of various metabolic disorders including hepatotoxicity. Treatment of diabetic rats with methanol extracts of P. Marsupium at 100 and 300 mg per kg-bwt per day for 21 days dose-dependently decreased serum glucose level. The higher dose exerted a protective effect on antagonized biochemical parameters such as reduced glutathione, superoxide dismutase and lipid peroxidation, and altered towards the normal levels hepatic mass, protein and glycogen content. In conclusion present results suggest antidiabetic, hepatoprotective as well as antioxidant effect of P. Marsupium.

  • Effect of Pterocarpus Marsupium in streptozotocin-induced hyperglycemic state in rats: comparison with glibenclamide.

    SUMMARY Pterocarpus Marsupium (Family: Leguminoceae) is widely used in ‘Ayurveda’ as ‘Rasayana’ for the management of various metabolic disorders including hyperglycemia. Treatment of diabetic rats with Pterocarpus Marsupium methanol extract (PMMtE, 300 mg/kg b.w./day) for 7 and 14 days showed normalization of streptozotocin-distressed serum glucose. At longer duration, PMMtE exerted a protective effect by correcting glycosylated hemoglobin (HbA 1c ), serum protein, insulin, alkaline and acid phosphatase (ALP and ACP), and albumin levels. Furthermore, tissue biochemical parameters viz. protein and glycogen altered towards normal in comparison to diabetic group. These results suggest oral administration of Pterocarpus Marsupium may have the ability to improve streptozotocin-induced chronic diabetic stress.
