Diagnostico citologico de células redondas neoplasicas de piel y tejido subcutaneo en canis familiaris mediante punción y aspiración con aguja fina (paaf) y tincion wright
Universidad Privada Antenor Orrego - UPAO, 2017Co-Authors: Romero Rodriguez, Rodrigo EduardoAbstract:The present study was carried out in the research laboratory of Veterinary Medicine of the Private University Antenor Orrego, in order to obtain information about the round cell neoplasias in skin and subcutaneous tissues, more frequent in Canis familiaris by means of cytology using the puncture technique And aspiration with fine needle and wright staining, the study was performed in 58 dogs with cutaneous and subcutaneous neoplasms of different sex, age and body condition, of which 57% were male and 43% female; It was observed that the round cell neoplasms were the most numerous with 53.4% of the total samples obtained, with the Mastocytoma being the highest incidence within this group, with 41.9% followed by the transmissible venereal tumor with 32.3% of the Total of dogs, the mestizos were the ones that obtained the greater percentage of neoplasias with 48.4%, followed by the American pitbull with 12.9%. Concluyéndose que el diagnóstico citológico mediante PAAF y tinción wright es un método eficaz, rápido, poco invasivo y económico para el diagnóstico de nuestros pacientes

Experimental Hematology, 1999Co-Authors: Cheryl A London, Stephen J Galli, Toshifumi Yuuki, Stuart C Helfand, Edwin N GeisslerAbstract:Spontaneous mast cell tumors (MCT) are the most common malignant neoplasm in the dog, representing between 7% and 21% of all canine tumors, an incidence much higher than that found in humans. These tumors often behave in an aggressive manner, metastasizing to local lymph nodes, liver, spleen, and bone marrow. The proto-oncogene c-kit is known to play a critical role in the development and function of mast cells. Point mutations in the kinase domain of c-kit leading to tyrosine phosphorylation in the absence of ligand binding have been identified in three Mastocytoma lines, (P815, RBL, and HMC-1), and some human patients with various forms of mastocytosis. We now demonstrate that although c-kit derived from canine MCT did not contain the previously described activating point mutations, 5 of the 11 tumors analyzed possessed novel mutations consisting of tandem duplications involving exons 11 and 12. We also show that one such duplication, detected in a canine Mastocytoma cell line, was associated with constitutive phosphorylation of c-kit protein (KIT), suggesting that these mutations may contribute to the development or progression of canine MCT.

phenotypic characterization of skin lesions in urticaria pigmentosa and Mastocytomas
Archives of Dermatological Research, 1995Co-Authors: Norbe Haas, K Hama, Jurge Grabbe, Eate M CzarnetzkiAbstract:In order to identify possible cellular abnormalities in human mastocytosis, sections from 13 urticaria pigmentosa lesions and 5 Mastocytomas were compared with 5 normal skin specimens using histochemical, enzyme histochemical and immunohistochemical techniques. All toluidine blue-positive mast cells also reacted with FceRI and c-kit antibodies, almost all stained for tryptase, many for chymase and the myeloid workshop mast cell antibodies, few for FceRII and none for the proliferation marker Ki-67. Urticaria pigmentosa lesions contained fewer epidermal Langerhans cells and a lower percentage of avidin-positive mast cells than Mastocytomas and normal skin. Mastocytomas exhibited generally weaker staining for mast cell markers and mostly lacked FceRI-bound IgE on mast cells and Langerhans cells, although the receptor was able to bind IgE in tissue sections. Most of the mast cell antibodies also reacted with other cell types. Only toluidine blue, avidin, tryptase and chymase stains were mast cell specific. Mast cells in mastocytosis thus differed only to a minor degree from normal mast cells, although distinct pathomechanisms may play a role in urticaria pigmentosa and mastocytosis.
