Meclofenoxate hydrochlor and hyperbaric oxygen hbo in the treatment of hypoxic ischemic encephalopathy of newborn
Hainan Medical Journal, 2010Co-Authors: Chen HuijieAbstract:Objective To assess the efficacy of hyperbaric oxygen (HBO) combined with Meclofenoxate hydrochlor in the tratment of infants with hypoxic-ischemic encephalopathy (HIE).Methods 120 full-term infants with hypoxic-ischemic encephalopathy were randomly divided into two groups:the hyperbaric oxygen group(60 cases) was conducted with routine therapy + HBO (one daily),the study group(60 cases) was treated with routine therapy + hyperbaric oxygen (HBO) + Meclofenoxate hydrochlor.Two group had been treated for ten days.Assessment of effects was performed using NABA criteria before and after treatment.Results The effective rate in the study group was significantly higher than that in the control group (P0.01).Conclusion HBO combined with Meclofenoxate hydrochlor is very effective in the treatmetn of infants with hypoxic-ischemic encephalopathy,with less side-effects.

bioequivalence study of Meclofenoxate hydrochloride capsule in healthy volunteers
China Pharmacy, 2011Co-Authors: Luo Xia, Cao JunhuaAbstract:OBJECTIVE:To evaluate the bioequivalence of two kinds of Meclofenoxate hydrochloride capsule in Chinese healthy volunteers.METHODS:18 healthy male volunteers were randomly divided into 2 groups and given single oral dose of Meclofenoxate test preparation or reference preparation 200 mg.The blood samples were collected at 0 h,and 0.33,0.67,1,1.5,2,2.5,3,4,6,8,10,14,24 h after adiministrotion.The concentration of chlorophenoxyacetic acid in plasma was determined by HPLC.The pharmacokinetic parameters were calculated and the relative bioequivalence was evaluated by DAS Ver 2.0 software.RESULTS:The main pharmacokinetic parameters of trial capsules vs.reference capsule of Meclofenoxate hydrochloride were as follows:tmax(2.11±0.37)h vs.(2.06±0.34)h;cmax(12.56±2.85)μg·mL-1 vs.(12.78±2.84)μg·mL-1,t1/2(5.81±2.68)h vs.(6.00±2.14) h;AUC0~24(34.11±7.89)μg·h·mL-1 vs.(34.12±7.99)μg·h·mL-1;AUC0~∞(34.83±7.68)μg·h·mL-1 vs.(34.79±7.97)μg·h·mL-1,respectively。The relative bioavailability of test capsule was(100.7±11.7)%.CONCLUSION:Two kinds of Meclofenoxate hydrochloride capsules are bioequivalent in Chinese healthy volunteers.

Chinese Journal of New Drugs and Clinical Remedies, 2010Co-Authors: Luo XiaAbstract:AIM To evaluate the bioequivalence of Meclofenoxate dispersible tablets and capsules in Chinese healthy volunteers.METHODS The study was performed in 18 healthy male volunteers according to a randomized 2-way crossover design.The blood samples were collected at 0 h and 0.33,0.67,1,1.5,2,2.5,3,4,6,8,10,14,24 h after taking 200 mg of Meclofenoxate dispersible tablets(test drug)and Meclofenoxate capsules(reference drug),and with repeated sampling of the crossover drug administration 7 d after clearance.The concentration of chlorophenoxyacetic acid in plasma was determined by HPLC.The pharmacokinetic parameters were calculated and compared statistically to evaluate the relative bioequivalence between the two preparations of Meclofenoxate by DAS Ver2.0 software.RESULTS The major pharmacokinetic parameters of Meclofenoxate dispersible tablets and Meclofenoxate capsules were as follows:tmax were(1.8 ± s 0.3)and(2.1 ± 0.3)h,ρmax were(13.9 ± 2.6)and(12.8 ± 2.8)mg·L-1,t1/2 were(6.1 ± 2.0)and(6.0 ± 2.1)h,AUC0-t were(36 ± 9)and(34 ± 8)mg·h·L-1,AUC0-∞ were(36 ± 9)and(35 ± 8)mg·h·L-1,respectively.The relative bioavailability of test drug was(105 ± 10)%.CONCLUSION Meclofenoxate dispersible tablets and Meclofenoxate capsules are bioequivalent in Chinese healthy volunteers.

China Modern Medicine, 2009Co-Authors: Luo XiaAbstract:Objective:To establish a high performance liquid chromatograph (HPLC) method for the determination of Meclofenoxate hydrochloride dispersible tablets and its related substances. Methods:The chromatographic separation was performed on a Diamonsil C18 (250 mm×4.6 mm, 5 μm)column with mobile phases of acetonitrile-0.02 mol/ml potassium dihydrogen phosphate solution (33:67)( pH adjusted to 4.0 with phosphoric acid). The detection wavelength was 225 nm. The flow rate was 1.0 ml/min.The column temperature was 25℃ and the sample size was 20 μl. Results: Meclofenoxate hydrochloride and its related substances were well separated by this method,and the resolution was accord with the demands. The detection limit of Meclofenoxate hydrochloride was 2.0 ng.The average content of the related substances in three batches of samples was 1.56%.The linear range of Meclofenoxate hydrochloride was 5.2-20.8 μg/ml(r=1.000)and the average recovery was 99.26%(RSD=1.12%). Conclusion: The method is sensitive, accurate, rapid and can be used to the quality control of Meclofenoxate hydrochloride dispersible tablets.

Journal of Pharmacy and Pharmacology, 2011Co-Authors: Sumie Yoshioka, Yukio Aso, Mitsuru UchiyamaAbstract:The kinetics of hydrolysis of Meclofenoxate hydrochloride in human plasma have been compared with those of clofibrate. The hydrolysis rate in fractionated plasma was determined in the presence and absence of a plasma esterase inhibitor, tetraethyl pyrophosphate. The kinetic data indicated that clofibrate decomposed only by esterase-induced hydrolysis, which was inhibited by binding of clofibrate to plasma proteins. In contrast to clofibrate, Meclofenoxate decomposed rapidly in human plasma via spontaneous hydrolysis as well as esterase-induced hydrolysis. The spontaneous hydrolysis appeared to be inhibited by some components present in the esterase fraction isolated from plasma, while no significant inhibition of the hydrolysis by protein binding was observed.

Analytica Chimica Acta, 2012Co-Authors: Rasha M Elnashar, Nour Abdel T Ghani, Sherif M HassanAbstract:This work offers construction and comparative evaluation the performance characteristics of conventional polymer (I), carbon paste (II) and carbon nanotubes chemically modified carbon paste ion selective electrodes (III) for Meclofenoxate hydrochloride are described. These electrodes depend mainly on the incorporation of the ion pair of Meclofenoxate hydrochloride with phosphomolybdic acid (PMA) or phosphotungestic acid (PTA). They showed near Nernestian responses over usable concentration range 1.0 × 10(-5) to 1.0 × 10(-2)M with slopes in the range 55.15-59.74 mV(concentrationdecade)(-1). These developed electrodes were fully characterized in terms of their composition, response time, working concentration range, life span, usable pH and temperature range. The electrodes showed a very good selectivity for Meclo with respect to a large number of inorganic cations, sugars and in the presence of the degradation product of the drug (p-chloro phenoxy acetic acid). The standard additions method was applied to the determination of MecloCl in pure solution, pharmaceutical preparations and biological samples. Dissolution testing was also applied using the proposed sensors.

These changes could be due to modification in membrane phospholipid composition of their blood vessels. Lipid peroxide in vivo has been identified as a basic deteriorative reaction in cellular mechanisms of aging in human. The effect of a nootropic drug, Meclofenoxate (MF) or its combination with extract of ginkgo biloba (EGb-761) or zinc (Zn) on malondialdehyde (MDA) product as an index of endogenous lipid peroxidation; phospholipid; glutathione (GSH) and protein thiols (PrSHs) contents as well as superoxide dismutase (SOD) activity in blood, brain, heart and liver of 24-month-old male rats was investigated. Aged rats were treated with MF once daily at oral doses of 100 mg kg-1 body wt. alone or with either EGb at a dose of 150 mg kg-1 body wt. or Zn at 10.5 mg kg-1 body wt. for 4 weeks. This study showed that aging caused a higher increment in MDA level of brain and heart than liver and plasma accompanied with reduction in brain and heart phospholipid contents as well as alteration of the antioxidant systems as compared to 4-month-old rats. Treatment of aged rats with MF alone or combined with either EGb or Zn caused improvement in the measured free radical scavengers especially in brain and heart tissues. Our results also showed that both EGb and Zn induced a significant potential effect of MF action on blood pressure and heart rate. The results were explained in the light of the antioxidant properties of EGb and Zn. Thus it is concluded that EGb and Zn have a beneficial role with MF in diminishing cumulative oxidative changes in aging.
