Journal of Xianning University, 2010Co-Authors: S I HuoxiAbstract:Objective To detect the expression of Melanoma Antigen-1 gene(MAGE-1),Melanoma Antigen-3 gene(MAGE-3) in rectal carcinoma with hepatic metastasis and explore its clinical significance.Methods The expression of MAGE-1 and MAGE-3 in central tumor tissue of rectal carcinoma with or without hepatic metastasis were detected by RT-PCR.Results Expression of MAGE-1 was positive in 14 out of 30(46.67%) cases and expression of MAGE-3 was positive in 17 out of 30(56.67%) cases in rectal carcinoma with hepatic metastasis respectively;11 out of 30 cases(36.67%) expressed MAGE-1 and MAGE-3 simutaniously;20 out of 30 cases(66.67%) expressed at least one of MAGE-1 and MAGE-3;Expression of MAGE-1 was positive in 8 out of 30(26.67%) cases and expression of MAGE-3 was positive in 12 out of 30(40.00%) cases in rectal carcinoma without hepatic metastasis respectively;7 out of 30 cases(23.33%) expressed MAGE-1 and MAGE-3 simutaniously;13 out of 30 cases(43.33%) expressed at least one of MAGE-1 and MAGE-3 in rectal carcinoma without hepatic metastasis;the positive rates of MAGE-1 or MAGE-3 of rectal carcinoma with hepatic metastasis were significant higher than those of rectal carcinom without hepatic metastasis(P0.05).Conclusion MAGE-1 and MAGE-3 protein may be used as a target molecule of immunotherapy for rectal cancer and indexes for follow-up study of rectal carcinoma as well.

The expression of MAGE-1 or MAGE-3 was not related to age, sex, histological grades, metastasis to lymph nodes, duke stages (P>0.05). Conclusion As their significance, MAGE-1, MAGE-3 protein may be used as a target molecule of immunotherapy for rectal cancer and indexes for follow-up study of rectal cancer as well. Real-time rapid reverse transcriptase—polymerase chain reaction for intraoperative diagnosis of lymph node micrometastasis: Clinical application for cervical lymph node dissection in esophageal cancers

The aim of this study was to establish a sensitive and rapid genetic assay to detect lymph node micrometastasis and to assess its usefulness clinically for cervical lymphadenectomy in esophageal cancer. We have recently shown that metastasis in the lymph node chain along the recurrent laryngeal nerves (rec LNs) is a predictor of cervical node metastasis in esophageal cancer. In our retrospective study, the positive rate of cervical lymph node metastasis with rec LNs metastasis was 51.6%, and the rate without rec LNs metastasis was 11.6%. There was a significant difference in both positive rates (P =.0002). METHODS: Rec LNs obtained from 50 patients with esophageal cancer were assessed prospectively by intraoperative histopathologic examination (HE) and genetic analysis. The latter involved a real-time quantitative reverse transcriptase-polymerase chain reaction (RT-PCR) system with multiple markers, carcinoembryonic Antigen, squamous cell carcinoma, and Melanoma Antigen-3, whose messenger RNAs are highly and frequently expressed in esophageal cancers. Cervical lymphadenectomy was subsequently performed in a subset of these patients. RESULTS: Ten of 50 patients (20%) were scored as node positive by HE, and 24 patients (48%) were scored positive by genetic diagnosis, including 9 HE-positive cases. Genetic diagnosis of rec LNs accurately predicted all 9 cases with cervical lymph node metastasis and 2 cases with cervical lymph node recurrence, whereas HE missed 2 cases with cervical lymph node metastasis and 2 cases with cervical lymph node recurrence. CONCLUSIONS: Our real-time rapid RT-PCR assay can improve the sensitivity of HE for detection of lymph node metastasis and might be potentially useful for intraoperative genetic diagnosis for subsequent cervical lymphadenectomy in esophageal cancer surgery.

expression of Melanoma Antigen gene 3 mage 3 in gastric neoplasm and its clinical significance
Chinese Journal of Current Advances in General Surgery, 2007Co-Authors: Zhang YanyunAbstract:Objectives:To detect the expression of Melanoma Antigen-3(MAGE-3) in human stomach neoplasms and explore the possibility of applying the Antigen as target for immunotherapy of stomach neoplasms.Methods:Expression of MAGE-3 gene in 40 stomach neoplasms,para-tumorous mucosa,and 20 samples of chronic gastritis were examined by reverse transcription-polymerase chain reaction(RT-PCR).The samples selected randomly from each gene with positive PCR were confirmed by DNA sequence analysis.Results:In 40 stomach neoplasms,MAGE-3 gene was expressed in 27 tumor tissues(67.5%).The positive rates of MAGE-3 gene expression in neoplasm tissues were significant higher than those of para-tumorous mucosa(15%) and 20 samples of chronic gastritis(10%).The DNA sequence confirmed that the RT-PCR products was truly target cDNA.The expression of MAGE-3 gene in stomach neoplasm tissues was relevant to the intruded depth of neoplasm cells(P0.05),but not relevant to pathological staging,the differentiatation degree of neoplasm cells and lymph node metastasis(P0.05).Conclusion:MAGE-3 gene is expressed in stomach neoplasm at a high frequency,which suggests the possibility of using them as targets for Antigen-specific immunotherapy.
