Otten, Derek O'haganAbstract:The inclusion of a potent TLR4 immune potentiator to a recombinant antigen Vaccine formulation enhances both the magnitude and the breadth of the engendered immune response. One such immune potentiator (TLR4 agonist E6020) was evaluated with recombinant Men B antigens delivered in MF59 sub-micron adjuvant emulsion. The ability of this formulation to enhance serum antibody and bactercidal titers was investigated. The co-delivery of E6020 within MF59 enhanced both the serum and bactericidal titers for Men B antigens and for Men B antigens combined with Men ACWY-CRM conjugate Vaccine. The delivery of TLR4 agonist within MF59 emulsion oil droplets leads to a more potent response in comparison to the TLR4 when admixed with MF59 emulsion.

Serological Analysis of Immunogenic Properties of Recombinant Meningococcus IgA1 Protease-Based Proteins

MelikhovaAbstract:Using the genome sequence of IgA1 protease of N. meningitidis of serogroup B, four recombinant proteins of different structure and molecular weight were constructed. These proteins were equal in inducing the formation of specific antibodies to IgA1 protease and had protective properties against meningococci. In the sera of immunized mice, anti-IgA1 protease antibodies were detected by whole-cell ELISA, which indicated the presence of IgA1 protease on the surface of these bacteria. We hypothesized that the protective properties of IgA1 protease-based antigens and IgA1 protease analogs could be realized not only via impairment of bacterium adhesion to the mucosa, but also via suppression of this pathogen in the organism. The presented findings seem promising for using these proteins as the basis for anti-Meningococcus Vaccine.

Structural basis for cooperativity of human monoclonal antibodies to meningococcal factor H-binding protein.

Cooperativity has been deeply investigated among mAbs elicited by factor H-binding protein (fHbp), a Neisseria meningitidis surface-exposed lipoprotein and one of the key antigens included in both serogroup B Meningococcus Vaccine Bexsero and Trumenba. Here we report the structural and functional characterization of two cooperative mAbs pairs isolated from Bexsero Vaccines. The 3D electron microscopy structures of the human mAb-fHbp-mAb cooperative complexes indicate that the angle formed between the antigen binding fragments (fAbs) assume regular angle and that fHbp is able to bind simultaneously and stably the cooperative mAbs pairs and human factor H (fH) in vitro. These findings shed light on molecular basis of the antibody-based mechanism of protection driven by simultaneous recognition of the different epitopes of the fHbp and underline that cooperativity is crucial in Vaccine efficacy.

Peschiera et al demonstrate the capability of four monoclonal antibodies from Bexsero Vaccine to simultaneously bind the human factor H protein in vitro. an epidemiological analysis on epidemic cerebrospinal meningitis of dongguan city in 2005
Journal of Tropical Medicine, 2007Co-Authors: Zhang ZewuAbstract:Objective The epidemic situation and surveillance data of epidemic cerebrospinal meningitis of Dongguan in 2005 were analyzed in order to provide basis on making measures of prevention and control of the disease. Methods The descriptive and analytical epidemiological methods were used to analyze the epidemic situation of epidemic cerebrospinal meningitis, and the antibody level of IgG and the carrier rate of Meningococcus of epidemic cerebrospinal meningitis in healthy population. Results Migrant young workers are high-risk population of the disease in Dongguan city. The carrier rate of Meningococcus of healthy people was 0.16%. The antibody level of IgG of type A and type C were 2.72 μg/ml and 3.35 μg/ml, respectively. The protection rate of type A was 56.22%. The antibody levels of local population were higher than those of migrant people. The superior epidemic strains of Meningococcus and their drug sensitivity have changed a lot. Conclusion More attention should be made to the migrant population in the prevention and control of epidemic cerebrospinal meningitis. General immunization by group A and C Meningococcus Vaccine in migrant young workers is suggested. Some drugs, such as Penicillin, Ciprofloxacin and Rifampin, to which Meningococcus are sensitive, are recommended instead of traditional sulfonamides in treating patients and selecting preventive drugs in controlling the spread of epidemic cerebrospinal meningitis.
