    Abstract:

    Macrophages play important roles in host defense mechanisms. In the brain, besides microglial cells, meningeal macrophages are present. However, the pathobiological characteristics of meningeal macrophages in rats remain to be investigated. In normal Meninx, immunohistochemically, macrophages reacting to CD163 (macrophage scavenger receptor) and major histocompatibility complex (MHC) class II-expressing cells (involving activated macrophages or dendritic cells) were sporadically seen without age-dependent changes. Injection of lipoplysaccharide (LPS) (5 µg; Escherichia coli) into the cerebrum increased the number of anti-CD68-positive macrophages (with greater phagocytic activity) in the Meninx, with a peak at 12 h during observation period until 48 h; MHC class II-expressing cells showed a gradual increase in number from 3 h after injection; however, anti-CD163-positive macrophages did not show significant change. In in vitro studies, LPS (0, 0.02, 0.05, 0.5, 5, 50 and 100 µg/ml) was added to KMY-1 or KMY-2 cells, both of which had been established from a rat malignant meningioma. KMY-1 originally reacted to CD163, but LPS addition at 0.5 µg/ml and greater concentrations decreased the anti-CD163-positive cell number and instead increased the anti-CD68-positive cell number. LPS-treated KMY-2 increased the anti-CD163-positive cell number at 0.05 and 0.5 µg/ml. By RT-PCR methods, LPS (0, 0.5, 5, 50, and 100 µg/ml)-treated KMY-1 and KMY-2 showed an increase in mRNA of monocyte chemoattractant protein-1 (MCP-1, a chemokine), and LPS-treated KMY-2 increased mRNA of nerve growth factor (NGF, an immunological effecter). Collectively, under LPS treatment, macrophages with heterogeneous functions appear in rat Meninx; rat Meninx-forming cells may be involved in pathogenesis of meningeal inflammation by expressing different immunophenotypes and by producing regulatory proinflammatory factors such as MCP-1 and NGF.     Abstract:

    To study the clinicopathologic characteristics,immunohistochemical features,differential diagnosis,and prognosis of solitary fibrous tumours(SFT)/hemangiopericytomas(HPC)in the maters(Meninx). Methods A series of 7 cases previously diagnosed as SFT/HPC at the Department of Pathology,Peking Union Medical College Hospital,during the period from 2008 to 2018 were analyzed for clinical data,histopathology,and immunohistochemical findings.The patients were followed up and the relevant literatures were reviewed. Results These seven patients included two males and 5 females aged 22 to 77 years(mean,49 years).Headache was the most common symptom.The magnetic resonance imaging of SFT/HPC showed irregularly contoured masses and dural tail sign was observed at the periphery of the lesion in 4 cases.The major axis of the tumor ranged from 1.8 cm to 10 cm(mean,4 cm).The tumors were located in the mater in 6 cases and in the spinal Meninx in 1 case.The tumors were surgically removed in all cases.Under light microscope,the tumors were formed by long round,oval or spindle cells,with rich branching vascular pattern and varying quantity of collagenous fibers bands in both sparse areas and dense areas.According the WHO classification,2 cases were in WHO grade Ⅰ,2 cases in WHO grade Ⅱ,and 3 cases in WHO grade Ⅲ.Immunohistochemistry of the paraffin-embedded tissues in all cases showed positive immunoreativity for CD34 and vimentin in all seven cases,along with positive signal transducer and activator of transcription 6 in 4 cases,negative epithelial membrane antigen and S-100 in 7 cases,and negative progestational hormone and somatostatin receptor 2 in 6 cases.The Ki-67 index ranged from 1% to 15%.Five patients with follow-up data(including 1 current case)were alive,while 2 patients were lost to follow-up. Conclusions The SFT/HPC are rare in the maters(Meninx)and is clinically difficult to be differentiated from other meningioma.The combination of CD34 and signal transducer and activator of transcription 6 helps to diagnose this disease.

  • Wool sheep and purple snails – Long‐term continuity of animal exploitation in ancient Meninx (Jerba/Tunisia)

    Abstract:

    Archaeological research at the ancient city of Meninx in Jerba, Tunisia, carried out by the Institut National du Patrimoine Tunisie and Ludwig Maximilian University of Munich (LMU) produced more than 10,000 faunal specimens and shed light on subsistence activities spanning from the fourth century BCE until the seventh century CE. Despite its highly diverse fauna totalling at least 69 species of mammals, birds, reptiles, fish and molluscs, domestic livestock formed the mainstay of the economy at Meninx. Throughout site occupation and compared with contemporaneous sites in coastal Tunisia and Libya, sheep were of prime importance at Meninx. Diachronic demographic profiling illustrates an emphasis on the production of wool for making textiles. Together with the ubiquitous presence of crushed banded dye‐murex (Hexaplex trunculus) shells implying exploitation of purple dyes, we assume that both activities were integrated into a single chaine operatoire for making purple‐dyed fabrics that were traded across the Mediterranean from Punic until Late Roman times. Zooarchaeological findings also suggest that during the Byzantine Period, this major economic activity came to a standstill, with people returning to more self‐sufficient subsistence strategies. An intersite comparison furthermore revealed that high proportions of ovicaprines are a typical feature of Punic–Roman sites in Jerba. But even at the height of Roman power in the region, autochthonous husbandry traditions continued to exist on the island, as illustrated by the fauna from Henchir Bourgou.

  • Expression of menin gene mRNA in pituitary tumours

    Abstract:

    Objective: Multiple endocrine neoplasia type 1 (MEN 1) is an autosomal dominant inherited disorder characterised by the combined occurrence of parathyroid, endocrine pancreas and anterior pituitary tumours. The gene responsible for MEN 1, the menin gene, a putative tumour-suppressor gene located on human chromosome 11q13, has been cloned. To investigate the role of the menin gene in sporadic anterior pituitary tumorigenesis, its mRNA was assessed in a group of pituitary tumours. Methods: Menin gene expression, along with glyceraldehyde phosphate dehydrogenase (GAPDH) gene expression, has been studied in a group of normal pituitaries and in 23 pituitary tumours not associated with the MEN 1 syndrome. The pituitary tumours included 4 prolactinomas, 11 growthhormone-secreting tumours and 8 non-functional tumours. Total RNA was extracted from the normal pituitaries and tumours, and cDNA was synthesised with standard reverse transcriptase methods. Duplex polymerase chain reaction (PCR) was standardised in order to quantify the expression of the menin gene using intron-spanning primers across exons 9 and 10 in relation to the ‘house-keeping’ gene GAPDH. The PCR products were separated on agarose gel and densitometric analysis of the bands allowed semi-quantification. Results: There was no evidence for a change in menin gene expression in any of the pituitary tumours when compared with normal pituitaries. Conclusions: These studies complement previous work on mutational analysis, and do not suggest a major role for the menin suppressor gene in sporadic pituitary tumorigenesis.

The work also provided new insights into all periods of the city’s history, from the 4th c. B.C. to the late 7th c. A.D. It also answered the question of where the harbor facilities of Meninx were situated.

    Abstract:

    This paper presents the results of the geophysical prospection conducted at the site of Meninx (Jerba) in 2015. This was the first step in a Tunisian-German project (a cooperation between the Institut National du Patrimoine, Tunis, and the Institut für Klassische Archäologie der Ludwig-Maximilans-Universität München), the aim of which is to shed light on the urban history of the most important city on the island of Jerba in antiquity.Meninx, situated on the SE shore of the island (fig. 1), was the largest city on Jerba during the Roman Empire and eponymous for the island's name in antiquity. The outstanding importance of this seaport derived from the fact that it was one of the main production centers of purple dye in the Mediterranean. With the earliest secure evidence dating to at least the Hellenistic period, Meninx saw a magnificent expansion in the 2nd and 3rd c. A.D. It was inhabited until the 7th c. when the city was finally abandoned.
